Trapping and preconcentrating particles and cells for enhanced detection and analysis are often essential in many chemical and biological applications. Existing methods for diamagnetic particle trapping require the placement of one or multiple pairs of magnets nearby the particle flowing channel. The strong attractive or repulsive force between the magnets makes it difficult to align and place them close enough to the channel, which not only complicates the device fabrication but also restricts the particle trapping performance. This work demonstrates for the first time the use of a single permanent magnet to simultaneously trap diamagnetic and magnetic particles in ferrofluid flows through a T-shaped microchannel. The two types of particles are preconcentrated to distinct locations of the T-junction due to the induced negative and positive magnetophoretic motions, respectively. Moreover, they can be sequentially released from their respective trapping spots by simply increasing the ferrofluid flow rate. In addition, a three-dimensional numerical model is developed, which predicts with a reasonable agreement the trajectories of diamagnetic and magnetic particles as well as the buildup of ferrofluid nanoparticles.  相似文献

A new microfluidic method that allows hydrodynamic focusing in a microchannel with two sheath flows is demonstrated. The microchannel network consists of a T-shaped main channel and two T-shaped branch channels. The flows of the sample stream and the sheath streams in the microchannel are generated by electroosmotic flow-induced pressure gradients. In comparison with other flow focusing methods, this novel method does not expose the sample to electrical field, and does not need any external pumps, tubing, and valves.  相似文献

The present study uses the dielectrophoresis (DEP) and electrothermal (ET) forces to develop on-chip micromixers and microconcentrators. A microchannel with rectangular array of microelectrodes, patterned either on its bottom surface only or on both the top and the bottom surfaces, is considered for the analysis. A mathematical model to compute electrical field, temperature field, the fluid velocity, and the concentration distributions is developed. Both analytical and numerical solutions of standing wave DEP (SWDEP), traveling wave DEP (TWDEP), standing wave ET (SWET), and traveling wave ET (TWET) forces along the length and the height of the channel are compared. The effects of electrode size and their placement in the microsystem on micromixing and microconcentrating performance are studied and compared to velocity and concentration profiles. SWDEP forces can be used to collect the particles at different locations in the microchannel. Under positive and negative DEP effect, the particles are collected at electrode edges and away from the electrodes, respectively, irrespective of the position, size, and number of electrodes. The location of the concentration region can be shifted by changing the electrode position. SWET and TWET forces are used for mixing and producing concentration regions by circulating the fluid at a given location. The effect of forces can be changed with the applied voltage. The TWDEP method is the better method for mixing along the length of the channels among the four options explored in the present work. If two layers of particle suspension are placed side by side in the channel, triangular electrode configuration can be used to mix the suspensions. Triangular and rectangular electrode configurations can efficiently mix two layers of particle suspension placed side-by-side and one-atop-the-other, respectively. Hence, SWDEP forces can be successfully used to create microconcentrators, whereas TWDEP, SWET, and TWET can be used to produce efficient micromixers in a microfluidic chip.  相似文献

Droplet-based microfluidics has gained extensive research interest as it overcomes several challenges confronted by conventional single-phase microfluidics. The mixing performance inside droplets/slugs is critical in many applications such as advanced material syntheses and in situ kinetic measurements. In order to understand the effects of operating conditions on the mixing performance inside liquid slugs generated by a microfluidic T-junction, we have adopted the volume of fluid method coupled with the species transport model to study and quantify the mixing efficiencies inside slugs. Our simulation results demonstrate that an efficient mixing process is achieved by the intimate collaboration of the twirling effect and the recirculating flow. Only if the reagents are distributed transversely by the twirling effect, the recirculating flow can bring in convection mechanism thus facilitating mixing. By comparing the mixing performance inside slugs at various operating conditions, we find that slug size plays the key role in influencing the mixing performance as it determines the amount of fluid to be distributed by the twirling effect. For the cases where short slugs are generated, the mixing process is governed by the fast convection mechanism because the twirling effect can distribute the fluid to the flow path of the recirculating flow effectively. For cases with long slugs, the mixing process is dominated by the slow diffusion mechanism since the twirling effect is insufficient to distribute the large amount of fluid. In addition, our results show that increasing the operating velocity has limited effects on improving the mixing performance. This study provides the insight of the mixing process and may benefit the design and operations of droplet-based microfluidics.  相似文献

For the diagnosis of biochemical reactions, the investigation of microflow behavior, and the confirmation of simulation results in microfluidics, experimentally quantitative measurements are indispensable. To characterize the mixing and reaction of fluids in microchannel devices, we propose a mixing quality index (Mqi) to quantify the cross-sectional patterns (also called mixing patterns) of fluids, captured with a confocal-fluorescence microscope (CFM). The operating parameters of the CFM for quantification were carefully tested. We analyzed mixing patterns, flow advection, and mass exchange of fluids in the devices with overlapping channels of two kinds. The mixing length of the two devices derived from the analysis of Mqi is demonstrated to be more precise than that estimated with a commonly applied method of blending dye liquors. By means of fluorescence resonance-energy transfer (FRET), we monitored the hybridization of two complementary oligonucleotides (a FRET pair) in the devices. The captured patterns reveal that hybridization is a progressive process along the downstream channel. The FRET reaction and the hybridization period were characterized through quantification of the reaction patterns. This analytical approach is a promising diagnostic tool that is applicable to the real-time analysis of biochemical and chemical reactions such as polymerase chain reaction (PCR), catalytic, or synthetic processes in microfluidic devices.  相似文献

Heat transfer in laminar tube flow with convective boundary conditions and axial heat conduction in the fluid is solved exactly. The effects of the external Biot numbers and the axial conduction in fluid on the temperature distribution and the local Nusselt number are determined for both the upstream and the downstream regions. In order to illustrate the effects of velocity profile, the results for the slug flow and the parabolic velocity profiles are plotted simultaneously. The heat transfer characteristics of the flow are found to be rather sensitive to the Péclét number, the external Biot numbers and the velocity profile in the thermal entrance region.  相似文献

Electric field-driven separation and purification techniques, such as isoelectric focusing (IEF) and isotachophoresis, generate heat in the system that can affect the performance of the separation process. In this study, a new mathematical model is presented for IEF that considers the temperature rise due to Joule heating. We used the model to study focusing phenomena and separation performance in a microchannel. A finite volume-based numerical technique is developed to study temperature-dependent IEF. Numerical simulation for narrow range IEF (6 < pH < 10) is performed in a straight microchannel for 100 ampholytes and two model proteins: staphylococcal nuclease and pancreatic ribonuclease. Separation results of the two proteins are obtained with and without considering the temperature rise due to Joule heating in the system for a nominal electric field of 100 V/cm. For the no Joule heating case, constant properties are used, while for the Joule heating case, temperature-dependent titration curves and thermo-physical properties are used. Our numerical results show that the temperature change due to Joule heating has a significant impact on the final focusing points of proteins, which can lower the separation performance considerably. In the absence of advection and any active cooling mechanism, the temperature increase is the highest at the mid-section of a microchannel. We also found that the maximum temperature in the system is a strong function of the ΔpK? value of the carrier ampholytes. Simulation results are also obtained for different values of applied electric fields in order to find the optimum working range considering the simulation time and buffer temperature. Moreover, the model is extended to study IEF in a straight microchip where pH is formed by supplying H+ and OH, and the thermal analysis shows that the heat generation is negligible in ion supplied IEF.  相似文献

We present a new approach to enhance mixing in T-type micromixers by introducing a constriction in the microchannel under periodic electro-osmotic flow. Two sinusoidal ac electric fields with 180° phase difference and similar dc bias are applied at the two inlets. The out of phase ac electric field induces oscillation of fluid interface at the junction of the two inlet channels and the constriction. Due to the constriction introduced at the junction, fluids from these two inlets form alternative plugs at the constricted channel. These plugs of fluids radiate downstream from the constriction into the large channel and form alternate thin crescent-shaped layers of fluids. These crescent-shaped layers of fluids increase tremendously the contact surface area between the two streams of fluid and thus enhance significantly the mixing efficiency. Experimental results and mixing mechanism analysis show that amplitude and frequency of the ac electric field and the length of the constriction govern the mixing efficiency.  相似文献

This study describes a novel microfluidic reactor capable of flow-through polymerase chain reactions (PCR). For one-heater PCR devices in previous studies, comprehensive simulations and experiments for the chip geometry and the heater arrangement were usually needed before the fabrication of the device. In order to improve the flexibility of the one-heater PCR device, two heat pipes with one fan are used to create the requisite temperature regions in our device. With the integration of one heater onto the chip, the high temperature required for the denaturation stage can be generated at the chip center. By arranging the heat pipes on the opposite sides of the chip, the low temperature needed for the annealing stage is easy to regulate. Numerical calculations and thermal measurements have shown that the temperature distribution in the five-temperature-region PCR chip would be suitable for DNA amplification. In order to ensure temperature uniformity at specific reaction regions, the Re of the sample flow is less than 1. When the microchannel width increases and then decreases gradually between the denaturation and annealing regions, the extension region located in the enlarged part of the channel can be observed numerically and experimentally. From the simulations, the residence time at the extension region with the enlarged channel is 4.25 times longer than that without an enlarged channel at a flow rate of 2 μl/min. The treated surfaces of the flow-through microchannel are characterized using the water contact angle, while the effects of the hydrophilicity of the treated polydimethylsiloxane (PDMS) microchannels on PCR efficiency are determined using gel electrophoresis. By increasing the hydrophilicity of the channel surface after immersing the PDMS substrates into Tween 20 (20%) or BSA (1 mg/ml) solutions, efficient amplifications of DNA segments were proved to occur in our chip device. To our knowledge, our group is the first to introduce heat pipes into the cooling module that has been designed for a PCR device. The unique architecture utilized in this flow-through PCR device is well applied to a low-cost PCR system.  相似文献

Hyperthermia can be used as an adjunctive method of chemotherapy, radiotherapy, and gene therapy to improve cancer treatment. In this study, we investigate the hyperthermic cell death of cervix cancer CaSki cells in a microchannel integrated with a directional heating scheme. Heat was applied from the inner end to the outer end of the channel and a temperature distribution from 60 °C to 30 °C was established. A three dimensional (3D) numerical model was conducted for the heat transfer simulation, based on which a simple fitting method was proposed to easily estimate the temperature distribution along the channel. Cell death along the channel was mapped 22 h after the heating treatment by dual fluorescent labeling and phase-contrast microscopy imaging. Upstream, where the temperature is higher than 42 °C, we observe necrotic death, late-stage and early stage apoptotic death in sequence along the channel. Downstream and in the middle of the channel, where the temperature is lower than 42 °C, significant cell detachment was noted. Vigorous detachment was observed even in the non-hyperthermic zone (temperature lower than 37 °C), which we believe is due to the direct effect of the hyperthermic zones (higher than 37 °C). The present work not only gives a vivid map of cell responses under a temperature gradient, but also reveals the potential interactions of the heated tumor cells and non-heated tumor cells, which are seldom investigated in conventional petri-dish experiments.  相似文献

Inertial microfluidics has brought enormous changes in the conventional cell/particle detection process and now become the main trend of sample pretreatment with outstanding throughput, low cost, and simple control method. However, inertial microfluidics in a straight microchannel is not enough to provide high efficiency and satisfying performance for cell/particle separation. A contraction–expansion microchannel is a widely used and multifunctional channel pattern involving inertial microfluidics, secondary flow, and the vortex in the chamber. The strengthened inertial microfluidics can help us to focus particles with a shorter channel length and less processing time. Both the vortex in the chamber and the secondary flow in the main channel can trap the target particles or separate particles based on their sizes more precisely. The contraction–expansion microchannels are also capable of combining with a curved, spiral, or serpentine channel to further improve the separation performance. Some recent studies have focused on the viscoelastic fluid that utilizes both elastic forces and inertial forces to separate different size particles precisely with a relatively low flow rate for the vulnerable cells. This article comprehensively reviews various contraction–expansion microchannels with Newtonian and viscoelastic fluids for particle focusing, separation, and microfluid mixing and provides particle manipulation performance data analysis for the contraction–expansion microchannel design.  相似文献

The present study concentrates to make a complete thermal analysis on a rectangular fin with its primary surface by taking into consideration of radiation heat exchange with the surrounding along with the convective mode of heat transfer. The one side of the primary surface is heated by a fluid with high temperature which may be required to dissipate heat quickly by a fin array system. To analyze an actual case study, the thermal conductivity of the fin material and convective heat transfer coefficient over the surface are treated as a variable and they are as a function of the local fin surface temperature. With the aforementioned condition, the energy equations for both the fin and primary surface become nonlinear. The decomposition method is suggested to solve these highly nonlinear equations to obtain a closed form temperature distribution. The result of temperature distribution determined by the present analysis is compared with that of the numerical values. With the adaptation of the simplified case, the present method is also compared with the exactly closed form results. From both of these comparisons, an exact matching of results is found. The fin performances, namely, fin efficiency, surface efficiency and augmentation factor are evaluated for a wide range of thermogeometric parameters.  相似文献

The T-shaped microchannel system is used to mix similar or different fluids, and the laminar flow nature makes the mixing at the entrance junction region a challenging task. Acoustic streaming is a steady vortical flow phenomenon that can be produced in the microchannel by oscillating acoustic transducer around the sharp edge tip structure. In this study, the acoustic streaming is produced using a triangular structure with tip angles of 22.62°, 33.4°, and 61.91°, which is placed at the entrance junction region and mixes the inlets flow from two directions. The acoustic streaming flow patterns were investigated using micro-particle image velocimetry (μPIV) in various tip edge angles, flow rate, oscillation frequency, and amplitude. The velocity and vorticity profiles show that a pair of counter-rotating streaming vortices were created around the sharp triangle structure and raised the Z vorticity up to 10 times more than the case without acoustic streaming. The mixing experiments were performed by using fluorescent green dye solution and de-ionized water and evaluated its performance with the degree of mixing (M) at different amplitudes, flow rates, frequencies, and tip edge angles using the grayscale value of pixel intensity. The degree of mixing characterized was found significantly improved to 0.769 with acoustic streaming from 0.4017 without acoustic streaming, in the case of 0.008 μl/min flow rate and 38 V oscillation amplitude at y = 2.15 mm. The results suggested that the creation of acoustic streaming around the entrance junction region promotes the mixing of two fluids inside the microchannel, which is restricted by the laminar flow conditions.  相似文献

A novel microfluidic device enabling selective generation of droplets and encapsulation of targets is presented. Unlike conventional methods, the presented mechanism generates droplets with unique selectivity by utilizing a K-junction design. The K-junction is a modified version of the classic T-junction with an added leg that serves as the exit channel for waste. The dispersed phase fluid enters from one diagonal of the K and exits the other diagonal while the continuous phase travels in the straight leg of the K. The intersection forms an interface that allows the dispersed phase to be controllably injected through actuation of an elastomer membrane located above the inlet channel near the interface. We have characterized two critical components in controlling the droplet size-membrane actuation pressure and timing as well as identified the region of fluid in which the droplet will be formed. This scheme will have applications in fluid sampling processes and selective encapsulation of materials. Selective encapsulation of a single cell from the dispersed phase fluid is demonstrated as an example of functionality of this design.  相似文献

A comprehensive study involving numerical analysis and experimental validation of temperature transients within a microchamber was performed for thermocycling operation in an integrated centrifugal microfluidic platform for polymerase chain reaction (PCR) amplification. Controlled heating and cooling of biological samples are essential processes in many sample preparation and detection steps for micro-total analysis systems. Specifically, the PCR process relies on highly controllable and uniform heating of nucleic acid samples for successful and efficient amplification. In these miniaturized systems, the heating process is often performed more rapidly, making the temperature control more difficult, and adding complexity to the integrated hardware system. To gain further insight into the complex temperature profiles within the PCR microchamber, numerical simulations using computational fluid dynamics and computational heat transfer were performed. The designed integrated centrifugal microfluidics platform utilizes thermoelectrics for ice-valving and thermocycling for PCR amplification. Embedded micro-thermocouples were used to record the static and dynamic thermal responses in the experiments. The data collected was subsequently used for computational validation of the numerical predictions for the system response during thermocycling, and these simulations were found to be in agreement with the experimental data to within ∼97%. When thermal contact resistance values were incorporated in the simulations, the numerical predictions were found to be in agreement with the experimental data to within ∼99.9%. This in-depth numerical modeling and experimental validation of a complex single-sided heating platform provide insights into hardware and system design for multi-layered polymer microfluidic systems. In addition, the biological capability along with the practical feasibility of the integrated system is demonstrated by successfully performing PCR amplification of a Group B Streptococcus gene.  相似文献

In the present article Parameterized Perturbation Method (PPM) is used to obtain the solutions of momentum and heat transfer equations of non-Newtonian fluid flow in an axisymmetric channel with porous wall for turbine cooling applications. The validity of the results of PPM solution were verified by comparison with numerical results obtained using a fourth order Runge–Kutta method. These comparisons reveal that Parameterized Perturbation Method is a powerful approach for solving this problem. The analytical investigation is carried out for different governing parameters namely, Reynolds number, injection Reynolds number, Prandtl number and power law index. The results show that skin friction coefficient increases with increase of Reynolds number, especially at high Reynolds numbers. Also it can be found that Nusselt number has direct relationship with Reynolds number, Prandtl number and power law index.  相似文献

Efficient mixing is difficult to achieve in miniaturized devices due to the nature of low Reynolds number flow. Mixing can be intentionally induced, however, if conducting or nonconducting obstacles are embedded within the microchannel. In the case of conducting obstacles, vortices can be generated in the vicinity of the obstacle due to induced charge electro-osmosis (ICEO) which enhances mixing of different streams: the obstacle shape affects the induced zeta potential on the conducting surface, which in turn influences the flow profile near the obstacle. This study deals with optimization of the geometric shape of a conducting obstacle for the purpose of micromixing. The obstacle boundary is parametrically represented by nonuniform rational B-spline curves. The optimal obstacle shape, which maximizes the mixing for given operating conditions, is found using genetic algorithms. Various case studies at different operating conditions demonstrated that the near right triangle shape provides optimal mixing in the ICEO flow dominant regime, whereas rectangular shape is the optimal shape in diffusion dominant regime. The tradeoff between mixing and transport is examined for symmetric and nonsymmetric obstacle shapes.  相似文献

Spherical and non-spherical wax microparticles are generated by employing a facile two-step droplet microfluidic process which consists of the formation of molten wax microdroplets in a flow-focusing microchannel and their subsequent off-chip crystallization and deformation via microdroplet impingement on an immiscible liquid interface. Key parameters on the formation of molten wax microdroplets in a microfluidic channel are the viscosity of the molten wax and the interfacial tension between the dispersed and continuous fluids. A cursory phase diagram of wax morphology transition is depicted depending on the Capillary number and the Stefan number during the impact process. A combination of numerical simulation and analytical modeling is carried out to understand the physics underlying the deformation and crystallization process of the molten wax. The deformation of wax microdroplets is dominated by the viscous and thermal effects rather than the gravitational and buoyancy effects. Non-isothermal crystallization kinetics of the wax illustrates the time dependent thermal effects on the droplet deformation and crystallization. The work presented here will benefit those interested in the design and production criteria of soft non-spherical particles (i.e., alginate gels, wax, and polymer particles) with the aid of time and temperature mediated solidification and off-chip crosslinking.  相似文献

A multidimensional, time-dependent model of the thermocline degradation in a packed bed thermal storage tank is presented. The formulation includes the effects of finite tank length, multidimensional thermal conduction, heat transfer between the fluid and solid portions of the bed, and heat losses across all tank surfaces. The technique used to solve the coupled pair of parabolic partial differential equations describing the degradation of the thermocline is based on a vectorized separation of variables approach. Assuming cylindrical geometry, the analysis leads to separate eigenvalue problems for the radial, angular, and axial spatial dependencies of the fluid and solid temperatures in the bed. The eigenvalues are readily calculated and the corresponding eigenfunctions are shown to form a complete set of spatial basis functions for the solution space. The method and results are illustrated by several examples.  相似文献
