The hepatoprotective and antioxidant activities of the n-butanol extract of Rubus parvifolius L. (RPL), a widely used medicinal plant, were evaluated. Results demonstrated that RPL extract possessed pronounced hepatoprotective effects against carbon tetrachloride (CCl4)-induced hepatic injury in mice, which was at least partially attributed to its strong antioxidant capacity. Treatment with RPL extract markedly attenuated the increases in serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels caused by CCl4 intoxication. It also significantly prevented the decrease in superoxide dismutase (SOD) activity and the increase in malondialdehyde (MDA) content of liver tissue. Meanwhile, histopathological changes of hepatic damage were also remarkably ameliorated. Phytochemical analysis based on high-performance liquid chromatography/tandem mass spectrometry (HPLC-MS/MS) revealed the presence of various phenolic compounds, including caffeic acid conjugates, ellagic acid glycosides, and flavonol glycosides, which might be responsible for the hepatoprotective and antioxidant activities of RPL.  相似文献

Objective: The prevalence of non-alcoholic fatty liver disease (NAFLD) has markedly increased. Insulin resistance has been implicated in the pathogenesis of NAFLD. This study was aimed at observing the relationship between insulin resistance and NAFLD, and evaluating the role of pioglitazone (PGZ) acting as insulin-sensitizing agents in the prevention and treatment of rat fatty liver induced by high fat feeding. Methods: The rats were separated randomly into 6 groups: model group Ⅰ were fed high fat diet for 8 weeks, PGZ prevention group were given PGZ 4 mg/(kg.d) simultaneously, while control group Ⅰ were fed normal food for 8 weeks; model group Ⅱ were fed high fat diet for 16 weeks, PGZ treatment group were given PGZ 4 mg/(kg.d) orally simultaneous with high fat diet for 8 weeks after high fat feeding for 8 weeks, control group Ⅱ were fed normal food for 16 weeks. The rats were sacrificed after 8 weeks and 16 weeks respectively. Liver weight, body weight, serum activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), tumor necrosis factor alpha (TNF-α), fasting blood glucose (FBG), fasting plasma insulin (FINS), HOMA (homeostasis model assessment) insulin resistance index (HOMA-IR), and the liver histology of rats of all groups were assayed. Results: After 8 weeks, the liver in model group Ⅰ showed typical steatosis, accompanied with mild to moderate lobular inflammatory cell infiltration, liver indexes and serum levels of ALT, AST, ALP, TNF-α were significantly increased (P〈0.05) compared with control group Ⅰ. Whereas, the degree of hepatic injury was attenuated in PGZ prevention group, liver indexes and serum levels of ALT, ALP were significantly decreased (P〈0.05) compared with model group Ⅰ. After 16 weeks, notable steatosis, and lobular inflammation were observed in model group Ⅱ rat liver, while the degree of hepatic injury was attenuated in the PGZ treatment group. Liver index, serum levels ofALT, AST, ALP, FINS and HOMA-IR were significantly increased (P〈0.05) in model group Ⅱ compared with control group Ⅱ. Whereas, in PGZ treatment group, serum levels of AST and FINS showed decreasing tendency, liver indexes, serum levels of ALT, ALP, TNF-α and HOMA-IR were significantly decreased compared with model group Ⅱ. Conclusion: Insulin resistance plays a role in the pathogenesis of NAFLD in rats. Pioglitazone can attenuate insulin resistance and biochemical and histological injury in high fat-induced fatty liver in rats.  相似文献

目的 研究氟对人胎肝细胞脂质过氧化的影响。方法 体外培养的人胎肝细胞接触不同浓度的氟化钠24小时后,检测人胎肝细胞脂质过氧化物(LPO)水平、还原型谷胱甘肽(GSH)含量以及培养液中LPO、乳酸脱氢酶(LDH)、丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)的活性。结果 各氟染毒组培养液中LDH和AST活性均显著高于对照组(P<0.05);细胞内和培养液中LPO水平升高(P<0.01),细胞内GSH含量则明显下降(P<0.01)。结论 氟可使人胎肝细胞脂质过氧化水平升高和CSH含量下降,并且二者均呈现明显的剂量-效应关系。  相似文献

目的:探讨乙型肝炎患者机体抗氧化能力的变化及其与病情的关系.方法: 门诊乙型肝炎患者共124例和健康者30例,均经放射免疫方法检测后加以确诊.乙型肝炎患者分为三组:乙型肝炎病毒复制组、乙型肝炎病毒恢复组、乙型肝炎合并肝癌组.患者组和对照组均经7150型全自动生化分析仪测定血清ALT、AST含量,应用改良TBA微量法测定血清丙二醛(MDA),以黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)、Cu/Zn-SOD及Mn-SOD水平的变化.结果: 乙型肝炎病毒复制组、乙型肝炎病毒恢复组、乙型肝炎合并肝癌组三组患者血清MDA水平均显著高于对照组(P<0.01),而血清SOD、Cu/Zn-SOD及Mn-SOD水平均显著低于对照组(P<0.01);乙型肝炎病毒复制组、乙型肝炎合并肝癌组患者血清MDA水平均显著高于乙型肝炎病毒恢复组(P<0.01),而血清SOD、Cu/Zn-SOD及Mn-SOD水平均显著低于乙型肝炎病毒恢复组(P<0.01);乙型肝炎合并肝癌组患者血清MDA水平均显著高于乙型肝炎病毒复制组(P<0.01),而血清SOD、Cu/Zn-SOD及Mn-SOD水平均显著低于乙型肝炎病毒复制组(P<0.01).但血清MDA水平与ALT、AST水平(r=0.151),血清SOD、Cu/Zn-SOD及Mn-SOD水平与ALT、AST水平(r=-0.218)均无相关性(P>0.05).结论: 乙型肝炎患者出现明显抗氧化能力的异常.通过血清MDA、SOD、Cu/Zn-SOD及Mn-SOD水平的检测,可为乙型肝炎的临床防治及疗效观察提供一定的依据.  相似文献

144只一日龄的AA肉仔鸡随机分为4组(n=36),预饲一周后分别在试验组基础日粮中添加50mg/kg、100mg/kg、150mg/kg的半胱胺,分别于2、4、6周龄采颈静脉血,分离血清,测定四组血清钙、磷和葡萄糖含量及ALT、AST、ALP活性,研究半胱胺对肉鸡部分血清化学参数及酶活性的影响.结果表明,饲料中添加适量的Cs能升高AA肉鸡的血钙水平,而对鸡血清磷、葡萄糖含量没有明显影响;添加50~100mg/kg的Cs对ALT、AST活性没有明显的影响;而饲料中添加Cs对ALP活性有一定影响.  相似文献

The attenuating effect of daidzein (DAD on oxidative toxicity induced by Aroclor 1254 (A 1254) was investigated in mouse testicular cells. Cells were exposed to A1254 alone or with DAI. The oxidative damage was estimated by measuring malondialdehyde (MDA) formation, superoxide dismutase (SOD) activity and glutathione (GSH) content. Results show that A 1254 induced a decrease of germ cell number, an elevation in thiobarbituric acid reactive substances (TBARS) but a decrease in SOD activity and GSH content. However, simultaneous supplementation with DAI decreased TBARS level and increased SOD activity and GSH content. Consequently, dietary DAI may restore the intracellular antioxidant system to attenuate the oxidative toxicity of A1254 in testicular cells.  相似文献

Astaxanthin (AST), a carotenoid molecule extensively found in marine organisms and increasingly used as a dietary supplement, has been reported to have beneficial effects against oxidative stress. In the current paper, the effects of AST on viability of prostate cells were investigated by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay; cell apoptosis and intracellular reactive oxygen species (ROS) levels were determined by flow cytometry; the mitochondrial membrane potential (MMP) was measured by fluorospectrophotometer; and activities of malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) were evaluated by a detection kit. The results show that copper ion (Cu2+) induced apoptosis, along with the accumulation of intracellular ROS and MDA, in both prostate cell lines (RWPE-1 and PC-3). AST treatments could decrease the MDA levels, increase MMP, and keep ROS stable in RWPE-1 cell line. An addition of AST decreased the SOD, GSH-Px, and CAT activities in PC-3 cell line treated with Cu2+, but had a contrary reaction in RWPE-1 cell lines. In conclusion, AST could contribute to protecting RWPE-1 cells against Cu2+-induced injuries but could cause damage to the antioxidant enzyme system in PC-3 cells.  相似文献

目的:观察脂肪肝大鼠在生化、病理、脂肪代谢、细胞因子、胰岛素及瘦素各方面指标的变化.方法:雄性Wistar大鼠22只,按体重层次随机分为2组,对照组(A)11只,脂肪肝组(B)11只.对照组饲以普通鼠饲料,脂肪肝组饲以高脂饲料.8周后,全部处死,取血及肝组织,测定血中AIT、AST、TG、CHO、INS、FBS、FFA、Ieptin、II-6、TNF.肝组织匀浆测定肝组织TC,TG,SOD、MDA,观察肝脏组织学变化.结果:脂肪肝大鼠存在明显脂肪代谢紊乱,肝功能异常,胰岛素抵抗及瘦素表达增高;细胞因子升高.结论:实验性高脂饮食脂肪肝大鼠存在瘦素抵抗及胰岛素抵抗.  相似文献

Chlorogenic acid (CGA), a polyphenolic compound, is abundant in fruits, dietary vegetables, and some medicinal herbs. This study investigated the prevention of CGA against acetaminophen (AP)-induced hepatotoxicity and its engaged mechanisms. CGA reversed the decreased cell viability induced by AP in L-02 cells in vitro. In addition, CGA reduced the AP-induced increased serum levels of alanine/aspartate aminotransferase (ALT/AST) in vivo. The effect of CGA on cytochrome P450 (CYP) enzymatic (CYP2E1, CYP1A2, and CYP3A4) activities showed that CGA caused very little inhibition on CYP2E1 and CYP1A2 enzymatic activities, but not CYP3A4. The measurement of liver malondialdehyde (MDA), reactive oxygen species (ROS), and glutathione (GSH) levels showed that CGA prevented AP-induced liver oxidative stress injury. Further, CGA increased the AP-induced decreased mRNA expression of peroxiredoxin (Prx) 1, 2, 3, 5, 6, epoxide hydrolase (Ephx) 2, and polymerase (RNA) II (DNA directed) polypeptide K (Polr2k), and nuclear factor erythroid-2-related factor 2 (Nrf2). In summary, CGA ameliorates the AP-induced liver injury probably by slightly inhibiting CYP2E1 and CYP1A2 enzymatic properties. In addition, cellular important antioxidant signals such as Prx1, 2, 3, 5, 6, Ephx2, Polr2k, and Nrf2 also contributed to the protection of CGA against AP-induced oxidative stress injury.  相似文献

The aim of this study was to evaluate the possible therapeutic or protective effects of Helichrysum plicatum DC. subsp. plicatum ethanol extract (HPE) against gentamicin-induced nephrotoxicity. Thirty-six Sprague Dawley male rats weighing between 200 and 250 g were used as live material. They were formed into six groups containing 6 rats each and were allowed to adapt to laboratory conditions for 7 d. Group I: control, 5% DMSO intraperitoneal (i.p.); Group II: HPE 100 mg/(kg·d) i.p.; Group III: HPE 200 mg/(kg·d) i.p.; Group IV: gentamicin as 80 mg/(kg·d) i.p.; Group V: gentamicin as 80 mg/(kg·d) i.p.+HPE 100 mg/(kg·d) i.p.; and Group VI: gentamicin as 80 mg/(kg·d) i.p.+HPE 200 mg/(kg·d) i.p. for 8 d. Following treatment, serum, liver, and kidney tissues were used to assess blood urea nitrogen (BUN), creatinine, enzymatic and non-enzymatic antioxidants, and lipid peroxidation. Gentamicin significantly increased serum BUN, creatinin, and liver and kidney levels of malondialdehyde (MDA). It also decreased the activity of catalase (CAT), glutathione peroxidase (GPx), and superoxide dismutase (SOD). Treatment with the HPE 100 mg/kg reversed gentamicin-induced alterations as evidenced by decreased serum BUN and creatinin, liver and kidney oxidant marker, and tubular necrosis as well as by an increase in antioxidant enzymes. It was found that HPE 200 mg/kg significantly increased liver and kidney tissue MDA levels in nephrotoxicity in rats. As a result, these findings support the proposition that HPE in 100 mg/kg dose demonstrates in the kidney and liver as free radicals and scavenger to prevent the toxic effects of gentamicin in both the biochemical and histopathology parameters.  相似文献

文章研究了不同剂量甲醛等挥发性物质对发育期大鼠体重、血液常规指标、肝肾功能的影响。研究结果表明:甲醛等挥发性物质的混合物能显著降低大鼠体重,而对肝、肾重量无明显影响;可明显提高大鼠血液中血小板(PLT)和嗜酸性粒细胞(EOS)的含量,但对血液中其它常规指标无明显影响;能明显提高大鼠血清中的谷丙转氨酶(ALT)和天冬氨酸氨基转移酶(AST)的活性.对血清中碱性磷酸酶(ALP)、γ-谷氨酰转肽酶(GGT)、血尿素氮(BUN)和肌酐(Cr)无明显影响。  相似文献

The present study is designed to investigate the protection by ferulic acid against the hepatotoxicity induced by diosbulbin B and its possible mechanism, and further observe whether ferulic acid augments diosbulbin B-induced anti-tumor activity. The results show that ferulic acid decreases diosbulbin B-increased serum alanine transaminase/aspartate transaminase (ALT/AST) levels. Ferulic acid also decreases lipid peroxide (LPO) levels which are elevated in diosbulbin B-treated mice. Histological evaluation of the liver demonstrates hydropic degeneration in diosbulbin B-treated mice, while ferulic acid reverses this injury. Moreover, the activities of copper- and zinc-containing superoxide dismutase (CuZn-SOD) and catalase (CAT) are decreased in the livers of diosbulbin B-treated mice, while ferulic acid reverses these decreases. Further results demonstrate that the mRNA expressions of CuZn-SOD and CAT in diosbulbin B-treated mouse liver are significantly decreased, while ferulic acid prevents this decrease. In addition, ferulic acid also augments diosbulbin B-induced tumor growth inhibition compared with diosbulbin B alone. Taken together, the present study shows that ferulic acid prevents diosbulbin B-induced liver injury via ameliorating diosbulbin B-induced liver oxidative stress injury and augments diosbulbin B-induced anti-tumor activity.  相似文献

目的:观察黄芩甙注射液对四氯化碳(CCl4)致急性肝损伤小鼠肝抗氧化功能的影响。方法:50只小鼠随机分为5组,每组10只,分别为对照组、模型组、黄芩甙注射液低、中、高(0.25 g/kg、0.50 g/kg、1.00g/kg)剂量组。用CCl4制备小鼠急性肝损伤模型,测定小鼠体重变化,肝指数,肝超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)活性,谷胱甘肽(GSH)及丙二醛(MDA)含量。结果:与对照组及模型组比较,黄芩甙各剂量组小鼠试验前后的体重及肝指数差异不显著(P>0.05)。与模型组比较,低剂量组小鼠肝SOD、GSH-Px活性及MDA含量差异不显著(P>0.05),而GSH含量显著升高(P<0.05);中、高剂量组SOD、GSH-Px活性极显著提高(P<0.01),GSH含量极显著升高(P<0.01),MDA含量极显著降低(P<0.01)。结论:黄芩甙能显著增强急性肝损伤小鼠肝SOD、GSH-Px活性,提高GSH含量,降低MDA含量,并表现正向的量效关系。  相似文献

研究目的:本研究旨在观察槲皮素对四氯化碳(CCl4)诱导的肝损伤的治疗解毒作用及其机理。创新要点:首次发现槲皮素对CCl4诱导的肝损伤有治疗作用,并且首次发现Prx和Trx家族参与其中。研究方法:检测小鼠血清转氨酶含量,并检测肝组织中丙二醛(MDA)、谷胱甘肽(GSH)和4.羟基壬烯醛(4-HNE)含量,并用实时聚合酶链式反应(PCR)检测肝组织中Prx1-6、TrxR1/2、Trx1/2、Nrf2和HO.1的mRNA表达情况。重要结论:CCl4造模成功后口服槲皮素对其造成的急性肝损伤有治疗作用,给药组小鼠血清中的转氨酶与模型组相比均有显著下降,通过MDA和免疫组化分析其机理可能和保护氧化应激损伤有关,通过实时PCR分析发现CCl4抑制了抗氧化酶Prx家族、TrxRd1、TrxRd2、Trx1、Trx2和Nrf2及其下游HO-1的基因表达,而槲皮素可以逆转CCl4降低的这些基因的表达。  相似文献

Salvianolic acid B(Sal B) is an active component of traditional Chinese medicine Salvia miltiorrhiza and is used to treat vascular diseases. To better understand its mechanism, the antioxidant capacities of Sal B was evaluated with human endothelial cells under oxidative stress. Human endothelial cells were pretreated with Sal B for 12 h followed by hydrogen peroxide for another 12 h. Production of reactive oxygen species (ROS), activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX), and concentration of glu-tathione were measured: Protective effect of Sal B on the endothelial cells from hydrogen peroxide-induced damage ' was observed, and ROS production in the cells was found significantly inhibited. Sal B remarkably enhanced the activities of antioxidant enzymes SOD, CAT and GPX. Furthermore, Sal B up-regulated the intracellular glutathione concentration. The results indicate that Sal B protected endothelial cells from oxidative stress by improving the redox status of the cells through enhancing the antioxidant enzyme activities and increasing the reductive glutathione concentration after the oxidative challenge.  相似文献

目的:探讨葡萄籽原花青素对大鼠肝缺血再灌注损伤的抗氧化作用、凋亡相关蛋白Bcl-2和Bax表达的影响。方法:SD雄性大鼠30只,复制肝缺血再灌注损伤模型。随机分为:假手术组;HIRI模型组;葡萄籽原花青素预处理(GSPE)组(160 mg/kg),每组10只。于再灌注后3 h检测肝组织谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量及血清谷丙转氨酶(ALT)、谷草转氨酶(AST)的含量;光镜下比较各组肝组织形态学的变化;免疫组化法检测GSPE对凋亡相关蛋白Bcl-2,Bax表达的影响。结果:与HIRI模型组比较,GSPE预处理组血清ALT、AST水平明显降低(P〈0.05);肝组织GSH-Px、SOD的活性明显升高(P〈0.05),而MDA含量下降(P〈0.05)。光镜下GSPE预处理组肝细胞损伤程度较模型组明显减轻;免疫组化结果显示,GSPE预处理组肝组织Bcl-2蛋白的表达量较模型组明显增多,而Bax蛋白的表达量则较模型组明显减少。结论:GSPE对缺血再灌注肝脏具有一定的抗氧化损伤和抗凋亡的作用。  相似文献

探索了将较为成熟的科研成果"文蛤酶解产物防醉解酒"的科研实验设计转化成药理学综合教学实验。以工艺成熟的文蛤酶解产物为评价对象,采用白酒灌胃的方式建立小鼠中剂量饮酒模型,疲劳转棒仪评价文蛤酶解产物对饮酒造成的小鼠平衡失调的缓减作用;建立高剂量饮酒模型,以翻正实验观察文蛤酶解产物的防醉解酒效果,同时测定血液及肝组织相关生化指标,光学显微镜下观察小鼠肝脏组织病理学变化。该实验可帮助学生巩固实验动物的捉、拿、固定、灌胃等基本操作,掌握醉酒模型评价实验的一般思路与方法,促进学生掌握生化分析、病理学切片等综合实验技能,提高分析数据及团队协作能力,符合国家对创新性人才的培养需求。  相似文献

本文对50例次经导管肝动脉化疗栓塞(TAE)治疗肝癌的并发症及其防治进行了分析。全部病例在TAE术后均有消化道症状,术后血清胆红质、尿素氮、丙氨酸氨基转移酶(ALT)和天门冬酸氨基转移酶(AST)均较术前有不同程度增高,重复化疗栓塞者易出现急性糜烂性胃炎,少见并发症有胆囊梗死、胸腔积液等。笔者还对部分并发症的防治进行了探讨。  相似文献

为探讨金樱子多糖对免疫抑制小鼠肝功能及血脂代谢的调节作用,取50只昆明小鼠随机平均分为正常对照组、模型组、RLP低剂量组、RLP中剂量组、RLP高剂量组.低、中、高剂量组分别按100ms/kg·d、200mg/kg·d、400mg/kg·d连续灌胃30d;正常对照组和模型组灌胃生理盐水.于试验前一周在继续灌胃的基础上,除正常对照组外,分剐于第l、3、5天给小鼠按25mg/kg体重腹腔注射地塞米松(Dex)建立免疫抑制动物模型.摘小鼠眼球取血,用试剂盒测定小鼠血清的总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL—C)、低密度脂蛋白胆固醇(LDL—C)、谷草转氨酶(AST)、谷丙转氨酶(ALT).结果表明,金樱子多糖能显著降低免疫抑制小鼠血清中TC、LDL—C、AST、Au含量,对HDL—C和TG有一定的调节作用.因此,金樱子多糖对免疫抑制小鼠肝功能及血脂代谢具有一定的调节作用.  相似文献

目的:通过实验观察甲醛对运动大鼠心肌组织抗氧化系统的影响及服用刺五加后的作用,探讨甲醛对运动大鼠的心肌组织毒性及刺五加的保护作用。方法:采用人为模拟不同程度甲醛染毒环境进行动式染毒,甲醛剂量分别为0.8 mg/m3,2.4 mg/m3。每天染毒30 min,每周6次,连续4周,刺五加的灌胃剂量为104.17 mg/kg,测定运动大鼠心肌组织的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)的活力和丙二醛(MDA)的含量。结果:染毒组与对照组相比,2.4 mg/m3剂量染毒运动组大鼠心肌组织 CAT、GSH-PX、SOD活性明显低于对照组(P〈0.01),MDA含量明显高于对照组(P〈0.01)。服药染毒运动组与染毒运动组相比,0.8mg/m3剂量染毒服药组大鼠心肌组织CAT、GSH-PX、SOD活性明显高于染毒运动组(P〈0.01),MDA含量明显低于染毒运动组(P〈0.01)。结论:甲醛可以降低染毒大鼠心肌组织抗氧化酶的活性,造成心肌组织氧化性损伤,而灌服中药刺五加可以有效缓解其毒性。  相似文献
