Objective: To conduct a randomized comparative trial of pharmacokinetics, efficacy and toxicity profile treatment with 1200 mg/m^2 gemcitabine using standard 30-min infusion or fixed dose rate (FDR) infusion [10 mg/(m^2-min)] on days 1 and 8 plus carboplatin AUC (area under curve) 5 on day 1 in Chinese non-small-cell cancer patients. Twelve patients were enrolled in this study. Methods: Plasma gemcitabine concentrations were measured by ion-pair reversed phase high performance liquid chromatography. Antitumoral activity and toxicity of gemcitabine was assessed according to World Health Organization criteria. Results: The obtained mean parameters, such as T1/2 (elimination half time), AUC, and CL (clearance), were consistent with those reported in literature. Qualified response rate in our study was 33.3% for standard arm and 50% for FDR ann. Additional 50% and 33.3% patients contracted stable disease (SD) in standard arm and FDR arm, respectively. The predominant toxicity was hematologic, and patients in the standard infusion ann experienced consistently more hematologic toxicity, Conclusion: Pharmacokinetic and clinical data in this trial support the continued evaluation of the FDR infusion strategy with gemcitabine.  相似文献

Angiogenesis is required for solid tumor growth and facilitates tumor progression and metastasis. The inhibition effects of O-(chloroacetyl-carbamoyl) fumagillol (TNP-470), an angiogenesis inhibitor, and gemcitabine, a chemotherapeutic agent, on expression of growth factors were investigated using human pulmonary adenocarcinoma cell line, A549. The A549 cells were divided into four groups: control group, 10^-6 mg/ml gemcitabine treated group, 10^-4 mg/ml TNP-470 treated group and gemcitabine+TNP-470 treated group. The mRNA and protein expression of vascular endothelial growth factor (VEGF) and its receptors, FMS-like tyrosine kinase-l (FLT-1) and kinase insert domain-containing receptor (KDR), in different groups were measured. The growth of A549 cell cultured with gemcitabine or TNP-470 was inhibited in an almost dose-dependent manner. Although gemcitabine (10^-6 mg/ml) alone and TNP-470 (10^-4 mg/ml) alone had no effect on the mRNA and protein expression of VEGF and its receptors (FLT-1, KDR) in A549 cells compared to the control (P〉0.05), 10^-6 mg/ml gemcitabine in combination with 10^-4 mg/ml TNP-470 had significant effect (P〈0.01). Moreover, combination of the two drugs significantly inhibited the mRNA expression of VEGF, FLT-1 and KDR compared to either drug alone (P〈0.05). This study suggests that combined treatment with TNP-470 plus gemcitabine may augment the antiangiogenic and antineoplastic effects in lung cancer cells in vitro.  相似文献

Decomposition of dimethyl sulfide in a wire-cylinder pulse corona reactor   总被引:1,自引:0,他引:1  
Decomposition of dimethyl sulfide (DMS) in air was investigated experimentally by using a wire-cylinder dielectric barrier discharge (DBD) reactor at room temperature and atmospheric pressure. A new type of high pulse voltage source with a thyratron switch and a Blumlein pulse-forming network (BPFN) was adopted in our experiments. The maximum power output of the pulse voltage source and the maximum peak voltage were 1 kW and 100 kV, respectively. The important parameters affecting odor decomposition, including peak voltage, pulse frequency, gas flow rate, initial concentration, and humidity, which influenced the removal efficiency, were investigated. The results showed that DMS could be treated effectively and almost a 100% removal efficiency was achieved at the conditions with an initial concentration of 832 mg/m3 and a gas flow rate of 1000 ml/min. Humidity boosts the removal efficiency and improves the energy yield (EY) greatly. The EY of 832 mg/m3 DMS was 2.87 mg/kJ when the relative humidity was above 30%. In the case of DMS removal, the ozone and nitrogen oxides were observed in the exhaust gas. The carbon and sulfur elements of DMS were mainly converted to carbon dioxide, carbon monoxide and sulfur dioxide. Moreover, sulfur was discovered in the reactor. According to the results, the optimization design for the reactor and the matching of high pulse voltage source can be reckoned.  相似文献

研究了以悬浮态TiO2为催化剂,在波长为365nm的125W紫外灯照射下加入H2O2对邻苯二甲酸乙酯(DEP)光催化降解,分别讨论了pH值、H2O2的加入量、邻苯二甲酸乙酯的初始浓度及光照时间对DEP的降解效果。结果表明:在pH=2、浓度为50mg/l的H2O2中对初始浓度为50mg/L的邻苯二甲酸乙酯溶液光照120分钟有较好的降解效果。  相似文献

Objective:To investigate the relationship between renal function and clinical outcomes among patients with acute ST-segment elevation myocardial infarction (ASTEMI), who were treated with emergency percutaneous coronary intervention (PCI). Methods: 420 patients hospitalized in Peking University First Hospital, diagnosed with ASTEMI treated with emergency (PCI) from January 2001 to June 2011 were enrolled in this study. Estimated glomerular filtration rate (eGFR) was used as a measure of renal function. We compared the clinical parameters and outcomes between ASTEMI patients combined renal insufficiency and the patients with normal renal function. Results:There was a significant increase in the concentrations of fibrinogen and D-Dimer (P<0.05) and a much higher morbidity of diabetes mellitus in the group of patients with chronic kidney disease (CKD; eGFR<60 ml/(min·1.73 m2)) (P<0.01). CKD (eGFR<60 ml/(min·1.73 m2)) was an independent predictor of in-hospital mortality for patients hospitalized with ASTEMI receiving PCI therapy rapidly (P=0.032, odds ratio (OR) 4.159, 95% confidence interval (CI) 1.127-15.346). Conclusions:Renal insufficiency is an independent predictor of in-hospital mortality for patients hospitalized with ASTEMI treated with primary PCI.  相似文献

In vitro cultures of loquat cultivar Mardan were established using shoot apices after treating with NaOCl (5%, 7%, 10%, 12%, 14% (v/v)) for 12 min and HgCl2 (0.01%, 0.05%, 0.10%, 0.20%, 0.25% (w/v)) for 2 min. A maximum survival rate of 70% was recorded after surface sterilization with 10% NaOCl. Caulogenic response was assessed on Murashige and Skoog (MS) medium fortified with assorted combinations of the cytokinins, benzylaminopurine (BAP), kinetin, and N6-(2-isopentyl)adenine (2iP). Treatment of BAP 1.5 mg/L combined with 2iP 9.0 mg/L and kinetin 1.5 mg/L was found to be optimum for shoot morphogenesis in terms of the number and subsequent growth of shoots, while the highest shoot length was yielded by the combination of BAP 0.5 mg/L, kinetin 0.5 mg/L, and 2iP 3 mg/L. Higher levels of cytokinins induced callogenesis, vitrification and stunted growth to some extent. For rhizogenesis, uniform sized micro-shoots were excised and transferred to half-strength MS medium containing auxins. The best rooting expression was observed with naphthaleneacetic acid (NAA) 1 mg/L combined with indole-3-butyric acid (IBA) 2 mg/L and paclobutrazol (PBZ) 1 mg/L.  相似文献

Objective: The aim of this paper is to compare the propofol concentration in plasma and cerebrospinal fluid (CSF) in patients scheduled for intracranial tumor removal and anaesthetized using propofol as part of a total intravenous anaesthesia technique. Methods: Twenty-seven patients (ASA I–II) scheduled for elective intracranial tumor removal were studied. Anesthesia was induced with 2 mg/kg propofol for 5 min and infused at 10 mg/(kg·h) for 5 min and then stopped. CSF and arterial blood were collected simultaneously before infusion of propofol and at different time points after infusion of propofol according to bispectral index (BIS) values. Concentrations of propofol in plasma and CSF were measured by HPLC with fluorescence detection. The correlation coefficient and regression equation between plasma and CSF concentration of propofol were worked out by linear simple regression. Results: The propofol CSF concentration that we measured was 1.46% of the plasma concentration. The coefficient of relation between plasma and CSF concentration was 76.7%. Conclusions: The propofol CSF concentration was positively correlated with and much lower than the plasma concentration. Discrepancies may result from high plasma protein binding of propofol, intracranial pathology and sampling volume. 探讨甲醛染毒对运动大鼠血红蛋白(HB)含量的影响及刺五加的保护作用.采用40只2月龄雄性Wistar大鼠,体重180±10g,随机分为对照组、模型组、刺五加组进行动式染毒,甲醛染毒剂量分别为0.8mg/m3、2.4mg/m3.每天染毒30min,每周6次,连续4周,刺五加的灌胃剂量为104.17mg/kg,测定运动大鼠HB含量.实验结果表明,模型组与对照组相比,模型组大鼠血HB含量均明显低于对照组(P0.05);刺五加组与模型组相比,刺五加组大鼠HB含量均明显高于模型组(P0.05).由实验得:甲醛可以造成运动大鼠HB含量降低,而灌服刺五加可以有效缓解其毒性.  相似文献

Scratch test and friction test were performed to evaluate the internal and external interface behaviors of TiAlSiN coating, respectively. The critical compressive and shearing stress of coating failure during scratch test were calculated and the values are 30.84 MPa and 4.98 MPa respectively. The average friction coefficients of TiAlSiN coating against 2Cr12Ni4Mo3VNbN steel are 0.70 (sliding speed 50 m/min), 0.63 (sliding speed 100 m/min), and 0.81 (sliding speed 150 m/min). The elements diffusion was analyzed by EDS. Al and Si element of coating material diffuse to the steel disc, except Ti element. The oxidation decreases with the increase of sliding speed, but the adhesion increases with the increase of sliding speed. More Al element diffuses to the steel disc at the high sliding speed, but the diffusion of Si element keeps almost constant at different sliding speeds.  相似文献

In this work, the orthogonal cutting experiments on Ti-6Al-4V alloy were conducted at different cutting speeds(10—160 m/min)and feed rates(20—160 μm/rev). The tool-chip contact length was measured by the track of tool rake face; meanwhile, the chip morphology caused by the localized and overall chip deformation was characterized by the degree of segmentation and the chip compression ratio, respectively. These parameters were analyzed and calculated according to the segmented chip morphology. In addition, three modified models considering the overall chip deformation and the localized deformation of adiabatic shear band were proposed, and the constants of the models were calculated by the genetic algorithm optimization. Considering the overall and localized chip deformation, the value and variation trend of the tool-contact length predicted by these three models agreed well with the experimental results.  相似文献

Objective: to evaluate cilazapril in vasovagal syncope treatment. Method: eighty-six cases of VVS patients found positive in TTT tilt were medicated with 2.5 mg cilazapril daily for three months and followed up by TTT. Results: seven cases quit due to cough or unexplained reason; 79 VVS patients had no more fainting spells; 75.95% of TTT results of patients changed to negative after 3 months therapy. The before and after cilazapril treatment average blood pressures (taken in lying position) were 121/73 mm Hg (1 mm Hg=0.133kPa) and 120/76 mm Hg respectively (P>0.05); and mean heart rates were 68.63±12.37/min and 70.13±13.15/min respectively with no significant changes (P>0.05). Conclusion: Cilazapril was effective in treatment of VVS; did not affect normal blood pressure and heart rate; was safe; and had little side effect.  相似文献

Removal of chromium (VI) dissolved in water by intermittent foam separation was implemented with cetyl trimethy-ammonium bromide as surfactant. The influence of various factors on removal efficiency was systematically studied. The removal efficiency has a maximum value near pH 4.0; thus, most experiments were carried out at pH 4.0. The orthogonal experiment was conducted to confirm the optimal operating parameters. The orthogonal experimental results show that when the liquid feed concentration is 10 mg/L, the pH value of feed solution is 4.00, air flow rates 0.9 L/min, surfactant dosage is 300 mg/L, the maximum removal efficiency of chromium (VI) reaches 97.80%, and condense multiple reaches 1 711. The kinetic test indicates that the foam separation of chromium is a first-order process. The equivalent rate constant calculated from the slope is 0.406 4, and the equivalent rate equation is obtained.  相似文献

为建立超声波提取-高效液相色谱法测定滁菊中木犀草素的方法。采用purospher star RP-18(250 mm×4.6 mm,5μm)色谱柱;以0.1%磷酸溶液(pH=2.6)-甲醇(40:60)为流动相;流速为1 mL/min;检测波长为327 nm。结果表明,木犀草素在10 min内实现快速基线分离,且在0.002~0.05 mg/mL范围内线性关系良好,r=0.9999,平均回收率为99.22%,RSD为1.7%(n=3)。该方法简便快速,可作为滁菊原药材的质量标准。  相似文献

选取王不留行饱满种子培养无菌苗,用无菌苗的茎和叶作为外植体,分别接种MS基本培养基外加激素NAA和6-BA诱导愈伤组织和不定芽,使用1/2MS培养基,附加IBA或NAA进行不定芽的生根实验,最后炼苗和移栽.实验结果表明王不留行的最优组织培养方案:种子用0.1%升汞消毒8m in,无菌湿脱脂棉催芽,茎和叶作为外植体,使用MS 0.1mg/L NAA 0.1mg/L 6-BA诱导愈伤组织和不定芽,1/2MS 0.15mg/L NAA诱导植株生根.  相似文献

利用酸催化的快速溶胶-凝胶法(sol-gel)制备了一系列不同La3+掺杂量(x=0.01%~3%)的TiO2复合光催化剂;在太阳光条件下,以亚甲基蓝溶液的光催化降解为模型反应,研究了染料的光催化降解动力学行为,考察了催化剂投加量和La3+掺杂量对复合光催化剂活性的影响.结果表明,亚甲基蓝的光催化降解反应遵循Langmuir-Hinshelwood动力学模型,表观反应速率常数随着反应体系中催化剂用量和La3+掺杂量的不同而不同,均存在一个最佳值.在本实验条件下,当催化剂投加量为1.5g/L,La3+掺杂量为0.5%时,测得表观反应速率常数为2.1.×10-2mg(L·min)-1,反应120min后亚甲基蓝的降解率可达91.55%.  相似文献

为拓宽茶产业剩余物的开发应用途径,以氯化锌为活化剂制备了废弃茶梗基活性炭,考察了各吸附条件对其吸附孔雀石绿效果的影响,并分析了其吸附动力学行为和等温吸附方程。结果表明:溶液pH等各因素对吸附效果都有一定影响;在染料初始浓度为100mg/L、吸附pH=7、吸附温度25℃、吸附剂投量为2g/L,吸附时间为150min的条件下,活性炭对孔雀石绿吸附量为57.8mg/g,对染料的去除率达到97.5%。活性炭对孔雀石绿的吸附过程是一个物理吸附控制的准二级动力学过程,等温吸附规律更符合Langmuir等温式。  相似文献
