植物组织培养是根据植物细胞具有全能性这一理论,在近几十年来发展起来的一项无性繁殖的新技术,是一项操作复杂而精细,操作难度较大的实践活动。其操作流程包括:配制母液、培养基配制与灭菌、外植体选择和消毒、接种、形成愈伤组织、诱导丛芽和生根、组培苗移栽等操作环节。由于操作持续时间长、环节多、要求高、难度大,目前基本停留在"讲实验"上。如何确保在有限的教学时间内顺利地开展实验教学呢?笔者以"菊花的组织培养"为例谈谈该课题教学与实验的可操作性。 相似文献

选取黄褐毛忍冬的茎尖、1年生的茎段作为外植体,分别对表面灭菌的方法、取材部位、初代培养、增殖培养、生根培养等进行了研究．结果显示,在黄褐毛忍冬外植体的表面灭菌过程中,氯化汞是较好的表面灭菌剂;在外植体的取材上,应以茎尖为主;在初代培养过程中,激素6-BA在0．5—1．0mg／L,NAA在0．1—0．2mg／L或IBA在0．1mg／L,茎尖萌发都比较好;在增殖培养过程中,添加0．5mg／L 6-BA＋0．1mg／LNAA和0．5mg／L 6-BA＋0．1mg／L IBA的培养基最有利于组培苗的增殖;在生根培养时,使用IBA为1．5mg／L时的培养基更有利于幼苗生根．筛选了一个较好的黄褐毛忍冬组培快繁种苗方案,为黄褐毛忍冬产业发展提供了一种快速、优质的种苗繁殖途径． 相似文献
