采用豌豆根尖微核技术研究了氯化钠对豌豆根尖细胞的遗传损伤,测定了不同浓度氯化钠(0.05,0.15,0.25mol/L)诱导下的豌豆根尖细胞微核率、有丝分裂指数和染色体畸变率.结果表明,氯化钠能诱发较高的微核率,降低根尖细胞有丝分裂指数;并能诱导豌豆根尖细胞产生较多类型的染色体畸变.氯化钠对豌豆根尖细胞有一定的遗传毒性.  相似文献

用二甲戊灵作为诱变剂诱导大蒜根尖染色体加倍,并与秋水仙素诱导法、低温诱导法对比。结果显示,二甲戊灵处理的大蒜根尖可观察到的中期分裂相多倍体细胞数目与秋水仙素处理效果相近,提示二甲戊灵适合作为多倍体诱导实验的诱变剂。  相似文献

以染色体较大、容易观察的大蒜为材料,将不同体积分数(0.1%、0.3%、0.5%)的甲醛水溶液作为诱变剂,分别处理大蒜根尖24 h、48 h、72 h和96 h.通过常规染色体压片技术,研究不同体积分数的甲醛溶液对大蒜根尖细胞有丝分裂行为和微核产生的影响.研究发现,大蒜根尖细胞对甲醛的毒性反应比较敏感,不同体积分数的甲醛水溶液均能使大蒜根尖细胞有丝分裂指数明显下降,且不同体积分数的甲醛处理均可诱发细胞微核的产生.结果表明:甲醛对大蒜根尖细胞有明显的细胞毒性和遗传毒性,甲醛浓度与根尖细胞有丝分裂指数呈负相关.  相似文献

以染色体较大、容易观察的大蒜为材料,研究不同体积分数的甲醛水溶液对大蒜根尖细胞有丝分裂行为和微核产生的影响.结果表明,大蒜对甲醛的毒性反应比较敏感,用0.01%~0.04%的甲醛水溶液处理4 h~12 h,细胞分裂指数较对照平均下降69.13%;处理24 h,细胞分裂完全停止,分裂指数为0;不同体积分数的甲醛处理均可诱发细胞微核的产生,频率在0.162%~0.235%;细胞分裂后期个别染色体表现落后,并随之解体、凝聚,是导致微核产生的一个重要成因.  相似文献

以蚕豆根尖为材料,研究洗手液对蚕豆根尖细胞的致畸效应,采用蚕豆根尖细胞的微核试验方法和染色体畸变试验方法,以不同浓度的洗手液为诱变剂,测定蚕豆根尖细胞的微核率、有丝分裂指数和染色体畸变率.结果表明:不同浓度的洗手液能诱发较高频率的微核,还能诱导染色体产生多种类型的染色体畸变.结论是洗手液具有一定的遗传毒性.  相似文献

为研究硒镉联合作用对番茄根尖细胞遗传学效应,用不同硒浓度和100μmol/L镉联合处理番茄根尖24 h、48 h、72 h、96 h,统计有丝分裂指数、染色体畸变率和微核率.结果表明:单一镉处理时番茄根尖细胞有丝分裂指数下降,染色体畸变率和微核率增加,且随时间的延长而毒害加深.硒镉处理时,硒浓度低于1.0μmol/L时番茄根尖细胞有丝分裂指数增加,染色体畸变率和微核率下降,高于1.0μmol/L时番茄根尖细胞有丝分裂下降,染色体畸变率和微核率升高,但浓度过高时,微核率随着镉处理时间的延长反而有所降低,表现出低浓度硒对镉的毒害有缓解作用,高浓度硒与镉协同毒害作用.  相似文献

以大蒜为材料,用不同质量浓度的秋水仙素(0,0.1,0.5,1.0,1.5,2.0g/L)和8-羟基喹啉(0,0.002,0.004,0.006,0.008,0.010mol/L)作为预处理剂,设计个处理时间(0.5,1.5,3.0,4.5,6.0h),分析不同预处理剂对大蒜根尖细胞有丝分裂的影响。结果表明,各预处理剂在不同浓度和处理时间情况下对大蒜根尖细胞中期分裂指数的影响是不相同的。(1)1.0g/L的秋水仙素处理3.0h,细胞的中期分裂指数最高,为6.11;(2)0.004mol/L的8-羟基喹啉处理3.0h,细胞的中期分裂指数也最高,为3.24。两种预处理剂互作处理时,细胞分裂中期的染色体分散性最好。  相似文献

本文通过对有丝分裂实验材料的改进,将人教版教材中洋葱或大蒜的根尖改为葱或大蒜的茎尖进行实验。实验发现,葱的茎尖及大蒜鳞芽形成期前的茎尖均可作为实验材料且分裂相明显。结果表明,葱或大蒜的茎尖比根尖易获取,且制成的装片可观察到更多分裂相。  相似文献

以不同浓度的垃圾渗出液为诱变剂,测定蚕豆根尖细胞的有丝分裂指数、微核率和染色体畸变率。结果表明:垃圾渗出液能够降低蚕豆根尖细胞有丝分裂指数,能诱发较高频率的微核和染色体畸变,产生染色体断片、滞后染色体和染色体桥。随着垃圾渗出液处理浓度的增加,有丝分裂指数降低,而微核率和染色体畸变率均升高;随着垃圾渗出液浓度的进一步增加,染色体畸变率却下降,但仍明显高于对照组。  相似文献

In this study on the mutagenic effects of different concentrations of chromium trioxide (CrO3) on Vicia faba root tip, micronucleus assay and chromosome aberration assay were used to determine the mitotic indexes, micronucleus rate and chromosome aberration rate of Viciafaba root tip cells. The results showed that the effects of CrO3 concentration on the mitotic indexes were complicated. CrO3 increases the micronucleus rate of Vicia faba root tip cells. It was found that within certain range of CrO3 concentration the micronucleus rate increased systematically with increased concentration of CrO3, but that the micronucleus rate decreased at higher level of CrO3 and that CrO3 also caused various types of chromosome aberration at a rate which increased systematically with increased concentration of CrO3. We concluded that CrO3 has significant mutagenic effect on Viciafaba root tip cells.  相似文献

采用不同浓度的氯化镉溶液处理龙葵根尖,通过常规染色体压片技术,观察其根尖分生组织细胞有丝分裂的情况及染色体的变化.结果表明,随氯化镉溶液浓度升高和处理时间的延长,其根尖生长速度和有丝分裂指数降低,并能引起染色体畸变和微核现象.说明氯化镉对龙葵根尖细胞有丝分裂有一定的抑制作用,具有一定的遗传毒性效应.  相似文献

The mutagemc effects of microwave and chromium trioxide (CrO3) on Vicia faba root tip were studied. Micronucleus assay and chromosomal aberration assay were used to determine the mitotic index, the micronucleus frequency and chromosomal aberration frequency of Vicia faba root tip cells induced by microwave and CrO3. The results showed that the micronucleus frequency decreased, and that the mitotic index and chromosomal aberration frequency showed linear dose responses to CrO3, in treatment of microwave for 5 s. In microwave of 25 s, the mitotic index decreased, the micronucleus frequency and chromosomal aberration frequency increased with increase of CrO3 concentration. We concluded that microwave and CrO3 had antagonistic effect on the mitotic index of Vicia faba root tip cells, but had synergetic effect on micronucleus frequency and chromosomal aberration frequency of Viciafaba root tip cells.  相似文献

Turmeric has long been used as a spice and food colouring agent in Asia. In the present investigation, the antimutagenic potential of curcumin was evaluated in Allium cepa root meristem cells. So far there is no report on the biological properties of curcumin in plant test systems. The root tip cells were treated with sodium azide at 200 and 300 μg/ml for 3 h and curcumin was given at 5, 10 and 20 μg/ml for 16 h, prior to sodium azide treatment. The tips were squashed after colchicine treatment and the cells were analyzed for chromosome aberration and mitotic index. Curcumin induces chromosomal aberration in Allium cepa root tip cells in an insignificant manner, when compared with untreated control. Sodium azide alone induces chromosomal aberrations significantly with increasing concentrations. The total number of aberrations was significantly reduced in root tip cells pretreated with curcumin. The study reveals that curcumin has antimutagenic potential against sodium azide induced chromosomal aberrations in Allium cepa root meristem cells. In addition, it showed mild cytotoxicity by reducing the percentage of mitotic index in all curcumin treated groups, but the mechanism of action remains unknown. The antimutagenic potential of curcumin is effective at 5 μg/ml in Allium cepa root meristem cells.  相似文献

以豌豆根尖细胞为材料,用5种品牌的废旧干电池的混合液配制成体积比为5%、10%、15%、20%、25%、30%(V废旧电池液/V蒸馏水)和原液为诱变剂染色体压片技术,测定豌豆根尖细胞的微核率,染色体畸变率和有丝分裂指数等指标.结果表明:不同体积比的废旧电池液均能诱发豌豆根尖细胞较高的微核和染色体畸变,在5%-25%体积比内,微核率随电池液体积比的增加而升高,当体积比大于25%后,微核率逐渐下降;不同体积比的电池液均能降低根尖细胞有丝分裂指数.废旧电池液对豌豆根尖细胞有较强的遗传毒性.  相似文献
