BackgroundLactic acid bacteria are able to reduce the immunoreactivity of proteins of cereal grains during wheat dough fermentation or may be a source of proteolytic preparations added during bread making. The key enzyme in prolamin degradation is prolyl endopeptidase. This study was aimed at optimizing the composition of a culture medium and culture conditions that would enhance the synthesis of intracellular prolyl endopeptidase (PEP) by Lactobacillus acidophilus 5e2.ResultsThe application of Plackett–Burman screening plans enabled demonstrating that the concentration of a nitrogen source in the culture and the initial pH value of the culture medium were significant for PEP synthesis. Further optimization conducted with the method of central composite designs (CCD) confirmed both the linear and square impact of nitrogen concentration and initial pH value of the culture medium on PEP production. In turn, the response surface method (RSM) allowed determining the optimal nitrogen concentration and pH value at 26.88 g/l and pH 4.85, respectively.ConclusionsValidation of the resultant model enabled over 3-fold increase in the quantity of the synthesized enzyme.  相似文献

For some years now, the history of modern mathematics and the history of modern science have developed independently. A step toward a reunification that would benefit both disciplines could come about through a revived appreciation of mathematical practice. Detailed studies of what mathematicians actually do, whether local or broadly based, have often led in recent work to examinations of the social, cultural, and national contexts, and more can be done. Another recent approach toward a historical understanding of the abstractness of modern mathematics has been to see it as a species of modernism, and this thesis will be tested by the raft of works on the history of modern applied mathematics currently under way.  相似文献

This essay examines different approaches to writing the history of science in light of the increased importance of microhistorical studies in the past two decades. It specifically examines the role of microhistory within the history of science and the importance of Thomas Kuhn's concept of the "normal exception" in early methodological statements about the function of microhistory. It also considers the possibilities for writing archivally based history of science for a general readership as a means of bridging the divide between specialized research within the subfields of the history of science and more general accounts of the nature and growth of science.  相似文献

We argue that some algorithms are value-laden, and that two or more persons who accept different value-judgments may have a rational reason to design such algorithms differently. We exemplify our claim by discussing a set of algorithms used in medical image analysis: In these algorithms it is often necessary to set certain thresholds for whether e.g. a cell should count as diseased or not, and the chosen threshold will partly depend on the software designer’s preference between avoiding false positives and false negatives. This preference ultimately depends on a number of value-judgments. In the last section of the paper we discuss some general principles for dealing with ethical issues in algorithm-design.  相似文献

This paper is a review of the changes brought about in the magnetic properties of “iron” during the period 1870 to 1928 and shows the absurdity of using “iron” as a standard for comparison. The latest (1928) value for the initial permeability (μ0) of “iron” is given as 1150, its maximum permeability (μmax) as 61,000, and its hysteresis loss (Wh) as 300 ergs per cubic centimeter per cycle for B = 10,000 gausses. Lazear (2005) suggests that entrepreneurs should be generalists, while those who work for others should be specialists. Many prospective entrepreneurs will develop varied skills by engaging in a variety of employment activities prior to becoming an entrepreneur, and incomes are higher for those that do so. An alternative view predicts that those with greater taste for variety are more likely to become entrepreneurs. Varied employment prior to becoming an entrepreneur is simply an expression of this taste, and is associated with lower earnings. Data from a survey of 830 independent inventors and 300 individuals from the general population are used to discriminate between these two theories. The results show that inventor-entrepreneurs typically have a more varied labor market experience, and that varied work experience is associated with lower household income.  相似文献

History of science has developed into a methodologically diverse discipline, adding greatly to our understanding of the interplay between science, society, and culture. Along the way, one original impetus for the then newly emerging discipline--what George Sarton called the perspective "from the point of view of the scientist"--dropped out of fashion. This essay shows, by means of several examples, that reclaiming this interaction between science and history of science yields interesting perspectives and new insights for both science and history of science. The authors consequently suggest that historians of science also adopt this perspective as part of their methodological repertoire.  相似文献

This essay argues that science education can gain from close engagement with the history of science both in the training of prospective vocational scientists and in educating the broader public about the nature of science. First it shows how historicizing science in the classroom can improve the pedagogical experience of science students and might even help them turn into more effective professional practitioners of science. Then it examines how historians of science can support the scientific education of the general public at a time when debates over "intelligent design" are raising major questions over the kind of science that ought to be available to children in their school curricula. It concludes by considering further work that might be undertaken to show how history of science could be of more general educational interest and utility, well beyond the closed academic domains in which historians of science typically operate.  相似文献

Performance incentives have become increasingly important in academia. Evidence suggest that they increase not only researchers’ effort but also their cheating. We develop a principal–agent framework in which a research institution (principal) aims to reward scientific productivity considering the social benefit of research and the remuneration costs. Researchers (agents) take into account what others are doing. As a consequence, the introduction of performance incentives not only leads to fraudulent behavior in the short run, but may also deteriorate the scientific norm in the long run. In the face of these dynamics, the principal pursues two goals: to increase productivity (effort incentive policy) and to deal with fraud (anti-fraud policy). Several factors determine the optimal mix of policies: economic factors (researchers’ cost of providing high or low effort, the societal value of scientific knowledge and the societal costs of scientific misbehavior) and psychological factors (the evolution of norms over time). An important insight of our theory is that when research institutions seek to encourage the production of high quality research, they should also raise the salary for normal research to avoid misconduct.  相似文献

To what extent should humans transfer, or abdicate, “responsibility” to computers? In this paper, I distinguish six different senses of ‘responsible’ and then consider in which of these senses computers can, and in which they cannot, be said to be “responsible” for “deciding” various outcomes. I sort out and explore two different kinds of complaint against putting computers in greater “control” of our lives: (i) as finite and fallible human beings, there is a limit to how far we can acheive increased reliability through complex devices of our own design; (ii) even when computers are more reliable than humans, certain tasks (e.g., selecting an appropriate gift for a friend, solving the daily crossword puzzle) are inappropriately performed by anyone (or anything) other than oneself. In critically evaluating these claims, I arrive at three main conclusions: (1) While we ought to correct for many of our shortcomings by availing ourselves of the computer's larger memory, faster processing speed and greater stamina, we are limited by our own finiteness and fallibility (rather than by whatever limitations may be inherent in silicon and metal) in the ability to transcend our own unreliability. Moreover, if we rely on programmed computers to such an extent that we lose touch with the human experience and insight that formed the basis for their programming design, our fallibility is magnified rather than mitigated. (2) Autonomous moral agents can reasonably defer to greater expertise, whether human or cybernetic. But they cannot reasonably relinquish “background-oversight” responsibility. They must be prepared, at least periodically, to review whether the “expertise” to which they defer is indeed functioning as he/she/it was authorized to do, and to take steps to revoke that authority, if necessary. (3) Though outcomes matter, it can also matter how they are brought about, and by whom. Thus, reflecting on how much of our lives should be directed and implemented by computer may be another way of testing any thoroughly end-state or consequentialist conception of the good and decent life. To live with meaning and purpose, we need to actively engage our own faculties and empathetically connect up with, and resonate to, others. Thus there is some limit to how much of life can be appropriately lived by anyone (or anything) other than ourselves.  相似文献

Few studies have attempted to link e-mail use to power and politics. The purpose of this article is to offer some initial evidence and analysis of e-mail's role in organizational power and politics. To frame the discussion, the article reviews the relevant writings on power and politics. The review is concluded with a theoretical critique that suggests that contextual and, particularly, temporal aspects should be incorporated into research on power and politics in organizations. To demonstrate how an analysis based on temporal aspects can be undertaken, a case study that outlines the use of e-mail for power and politics in a university is presented. The discussion and conclusions sections synthesize the analysis by demonstrating that what was interpreted by many organization members at the early stages of the conflict as a ''battle of the sexes'' can be seen (and, indeed, was interpreted by many organization members at the later stages of the conflict) as an elaborate exercise in misinformation created by top management to further its political objectives. The article concludes with a discussion of the implications from this case to e-mail research and practice.  相似文献

With labour market problems pressing national governments to support private innovation, the market mechanism is reinforced as it sets the rate and direction of innovation: (1) in the mix of improvement innovations in established industries, labour-saving technologies are outnumbering the product innovations that offer additional employment and investment opportunities, and (2) basic innovations are rare (stale-mate in technology), but when they occur they tend to occur in ‘clusters’. The competitive nature of allocative decisions on corporate R&D and innovative investment is complemented by the competitive R&D policy of most European countries. As those decisions concern large-scale, highcomplexity technologies, the perspective that the market mechanism tends to produce them in ‘clusters’ poses serious problems of timing, spacing, and — of course — of value judgement about alternative technological developments in the European context. These strategic problems of innovation policy have risen to such a level of potential conflict that they need harmonization.  相似文献
