养殖斑节对虾红体病病原及组织病理研究   总被引:3,自引:0,他引:3  
养殖班节对虾红体病病原经组织病理、电镜观察和白斑杆状病毒PCR检测,初步证实是一种枝原体.枝原体高度多态性,主要以球状体和分枝丝状体形式存在.球状体直径60~108um,分枝丝状体膨大部直径达150um,丝状部为25um.枝原体主要寄生在斑节对虾肝胰腺上皮细胞、造血组织细胞、鳃上皮细胞、甲壳表皮细胞等,致肝胰腺萎缩糜烂.受感染细胞细胞质空泡变性,细胞核固缩解体,内质网、高尔基体和溶酶体消失.对红体带白斑斑节对虾、白斑(无红体特征)斑节对虾和红体(无白斑特征)斑节对虾组织病理、电镜和白斑杆状病毒PCR检测,证实斑节对虾红体病不是由白斑杆状病毒引起,红体斑节对虾具有白斑杆状病毒是病毒继发感染所致.  相似文献

班节对虾白斑综合症杆状病毒(Whitespotsyndromebaculoviruses,WSSBV)感染胃上皮细胞,胃环肌和纵肌细胞,海绵结缔组织细胞;鳃下皮细胞;触角腺分泌型和非分泌型上皮细胞,触角腺体胶囊上皮细胞;造血组织细胞;心脏心肌细胞,随体细胞,结统组织细胞;神经系统胶质细胞,神经膜细胞,神经细胞和球形细胞;甲壳表皮细胞、肌细胞.WSSBV对中肠和中肠育囊上皮细胞,肝胰腺上皮细胞,雄性生殖系统精原细胞,输精管上皮细胞等不敏感.人工肌注感染,WSBV有3d内可致斑节对虾90%,甚至100%死亡.感染斑节对虾鳃经电镜观察发现了3种杆状病毒,2种为无包涵体细胞核杆状病毒,其中一种大小为(119~136)um×(289~306)um,核农壳(98~105)nm×(265~272)um,另一种大小为(101~112)um×(391~420)um,核衣壳(78~85)nm×(276~298)um;1种为细胞质杆状病毒,大小为(102~112)nm×(326~340)um,核衣壳(71~78)nm×(306~308)um.  相似文献

从感染斑节对虾杆状病毒(MBV)的斑节对虾仔虾肝胰腺中纯化MBV,经抽提后以之为模板进行聚合酶链反应(PCR),引物根据苜蓿丫纹夜蛾核型多角体病毒(AcNPV)多角体蛋白基因序列的高度保守区设计并合成.PCR扩增得到650b单一核苷酸片段.PCR产物部分序列分析表明此PCR产物与AcNPV多角体蛋白基因序列有约60%的同源性.以此650pDNA片段为探针作打点杂交,探针能与受MBV感染的斑节对虾肝胰腺和消化道中抽提的总DNA杂交,不能与其它组织中抽提的总DNA杂交.应用PCR方法检测MBV的组织特异性,得出与打点杂交相同的结果;检测斑节对虾各生长阶段及卤虫感染MBV情况,认为MBV可能为水平传播;PCR检测方法与组织学方法比较,认为前者更加敏感.  相似文献

对虾白斑综合症病毒(WSSV)编码与核酸合成代谢相关的核糖核苷酸还原酶(Ribonucleotide Reductases,RR),与病毒DNA的复制有关.利用克隆技术将RR基因克隆到L4440载体,构建体内合成dsRNA的大肠杆菌HTn5工程菌.诱导该工程菌合成了RR基因的特异双链RNA(RR—dsRNA)和非特异双链RNA(gfP—dsRNA),分别与WSSV混合共注射凡纳滨对虾,在感染72h后用病毒检测试剂盒提取DNA模板用于荧光定量PCR分析,结果显示RR—dsRNA能有效抑制WSSV病毒粒子的增值.  相似文献

红底板中华鳖和白底板中华鳖中均发现细菌和病毒.经细菌培养证实红底板和白底板鳖肝、脾、肾、肺、心和鳃样组织中均有细菌,电镜观察也证实白底板鳖肝和肺中有细菌,红底板鳖肺和鳃样器官中有细菌.细菌短杯状,大小(0.5~0.6)μm×(0.8~1.0)μm.在白底板鳖脾、肾和胃中和红底板小场中分别检测到直径为70~75um和85~90um的病毒粒子.红底板病鳖和白底板病鳖均表现为广泛地系统性的组织坏死,白底板病鳖较红底板病鳖病变严重.  相似文献

Epstein-Barr virus(EBV),a human gammaherpesvirus carried by more than 90% of the world’s population,is associated with malignant tumors such as Burkitt’s lymphoma(BL),Hodgkin lymphoma,post-transplant lymphoma,extra-nodal natural killer/T cell lymphoma,and nasopharyngeal and gastric carcinomas in immune-compromised patients.In the process of infection,EBV faces challenges:the host cell environment is harsh,and the survival and apoptosis of host cells are precisely regulated.Only when host cells receive sufficient survival signals may they immortalize.To establish efficiently a lytic or long-term latent infection,EBV must escape the host cell immunologic mechanism and resist host cell apoptosis by interfering with multiple signaling pathways.This review details the apoptotic pathway disrupted by EBV in EBV-infected cells and describes the interactions of EBV gene products with host cellular factors as well as the function of these factors,which decide the fate of the host cell.The relationships between other EBV-encoded genes and proteins of the B-cell leukemia/lymphoma(Bcl) family are unknown.Still,EBV seems to contribute to establishing its own latency and the formation of tumors by modifying events that impact cell survival and proliferation as well as the immune response of the infected host.We discuss potential therapeutic drugs to provide a foundation for further studies of tumor pathogenesis aimed at exploiting novel therapeutic strategies for EBV-associated diseases.  相似文献

植物在其与痛原物相互关系长期进化过程中,通过各种代谢途径在体内可以形成各种抵抗病原物有关的物质。这包括植物凝集素、富含羟脯氨酸的糖蛋白、胼胝质、类黄酮和萜类植保素、过氧化物酶、几丁质酶(chitinease)和1,3-β-葡聚糖酶、病程相关蛋白和水杨酸等等。寄主植物细胞壁在感染病原菌后的木质化作用-木质素含量的增加是寄主植物抗性反应的一种特性,为阻止病原菌的进一步侵染提供保护圈。苯丙氨酸解氨酶(PAL),肉桂酸-4-羟化酶(CA4H)和4-香豆酸-CoA联结酶(4CL)是苯丙烷代谢途径的关键酶。这三个酶的活性在植物体内的变化与植物的抗病性存在一定的联系。水杨酸的Mr小,适宜于植物韧皮部中的长距离传输,被认为是植物诱导抗性的信号分子。  相似文献

Herpes simplex virus type 1 (HSV-1), a neurotropic member of the alphaherpes virus family, is among the most prevalent and successful human pathogens. HSV-1 can cause serious diseases at every stage of life including fatal disseminated disease in newborns, cold sores, eye disease, and fatal encephalitis in adults. HSV-1 infection can trigger rapid immune responses, and efficient inhibition and clearance of HSV-1 infection rely on both the innate and adaptive immune responses of the host. Multiple strategies have been used to restrict host innate immune responses by HSV-1 to facilitate its infection in host cells. The adaptive immunity of the host plays an important role in inhibiting HSV-1 infections. The activation and regulation of T cells are the important aspects of the adaptive immunity. They play a crucial role in host-mediated immunity and are important for clearing HSV-1. In this review, we examine the findings on T cell immune responses during HSV-1 infection, which hold promise in the design of new vaccine candidates for HSV-1.  相似文献

Silver carp, Hypopthalmichthys molitrix is one of the most economically valuable fish species in Bangladesh. However, its production is often hindered by parasite-induced mortality. The present study reports the intensity of parasitic infestation in 216 specimens of H. molitrix collected from different fish markets in Rajshahi City, Bangladesh. Nine different parasite species (Trichodina pediculatus, Dactylogyrus vastator, Ichthyophthirius multifilis, Gyrodactylus elegans, Lernaea sp., Apiosoma sp., Myxobolus rohitae, Camallanus ophiocephali, and Pallisentis ophiocephali) were recovered from the gill, skin, stomach, and intestine of host fish. The highest level of infection was observed for host skin, while lower levels were observed for host gill, stomach, and intestine. The results also revealed that the intensity of parasite infection in different organs of H. molitrix varied with the season. In particular, the highest levels of infection were recorded during the winter period (November-February), when fish are most susceptible to parasites. The findings of the study will help in the management and conservation of H. molitrix.  相似文献

为获得宿主菌 ,研究了十二烷基硫酸钠 (SDS)对枯草芽孢杆菌中质粒的消除 .将过夜培养的枯草芽孢杆菌2 4/pMX45接种于含SDS( 0— 0 .0 0 8% )的LB培养基中 ,当SDS浓度 (w /v)大于或等于 0 .0 0 6 %时 ,菌体不能生长 .SDS的亚致死浓度为 0 .0 0 5 % ,其致死率达 99% .菌液稀释后涂LB平板 ,再随机挑选单菌落至抗性平板 ,检测由质粒编码的红霉素抗性是否丢失 .氯化铯 溴化乙锭梯度离心及质粒DNA的电泳图像证实了 2 4/pMX45的衍生菌株A7中的质粒已完全被消除 .A7延迟期较短 ,并且细胞浓度高于 2 4/pMX45 .用SDS处理 8h后 ,2 4/pMX45的遗传标记开始丢失 ,消除率持续增高至 2 2h ,随之消除质粒的菌体量保持恒定 .在未经SDS处理的对照实验中 ,相同条件下培养 2 4h及 48h后 ,没有发现质粒自然丢失的现象 ,因此SDS能消除枯草芽孢杆菌中的质粒  相似文献

目的:探讨HBsAg、HBeAg阳性孕妇外周血单个核细胞(PBMC)内乙型肝炎病毒(HBV)DNA感染状况及其在宫内母婴垂直传播中的作用.方法:对HBsAg/HBeAg双阳性共67对孕妇及其新生儿静脉血分离和提纯PBMC后,经抽提、纯化后的DNA进入PCR扩增反应,引物为HBV C区基因序列.结果:67名HBsAg及HBeAg双阳性的孕妇中有35例(52.2%)PBMC中HBV DNA阳性,25例孕妇在血清及PBMC中均发现HBV DNA.67名新生儿有22例感染HBV DNA,感染率32.8%,其中血清HBV DNA阳性者10例,PBMC HBVDNA阳性者19例,二者均阳性者7例.结论:母亲PBMC内HBV DNA阳性可能导致新生儿PBMC中HBV DNA阳性,PBMC内的HBV DNA可能是HBV母婴垂直传播的一条重要途径,同时,HBsAg及HBeAg阳性母亲若血清HBV DNA为阳性就极大增加了其新生儿感染HBV的危险性.  相似文献

Appressorium formation is an important event in establishing a successful interaction between the rice blast fungus, Magnaporthe oryzae, and its host plant, rice. An understanding of molecular events occurring in appressorium differentiation will give new strategies to control rice blast. A quick and reliable method to extract total RNA from appressorium is essential for studying gene expression during appressorium formation and its mechanism. We found that duplicate film is an efficient substratum for appressorium formation, even when inoculated with high density conidia. When inoculated with conidia at 1×106ml-1, the percentages of conidium germination and appressorium formation were (97.98±0.67)% and (97.88±0.45)%, respectively. We applied Trizol before appressorium collection for total RNA isolation, and as much as 113.6 pg total RNA was isolated from the mature appressoria at 24 h after inoculation. Functional analysis of two genes, MNH6 and MgATG1, isolated from the cDNA subtractive library, revealed that the quantity of RNA was good enough to construct a cDNA (complementary DNA) library or a cDNA subtractive library. This method may be also applicable for the appressorium RNA isolation of other pathogenic fungi in which conidia differentiate into appressoria in the early stages of host infection.  相似文献
