Determination of blood glucose levels is very important to know the physiological condition of the human beings as the hormonal
imbalance may cause abnormalities in glucose metabolism. The traditional methods of glucose estimation by colorimetric and
titrimetric methods were involved with huge expenditure and time. The modified colorimetric microwell reader method proposed
in the present study was performed with small quantities of sample and reagents with the same linearity that was observed
in the normal colorimetric analysis. The modified method not only reduces the cost of the test to almost one third of the
normal colorimetric method but also provide an opportunity to screen the large number of samples in a short duration of time. 相似文献

The present study reveals the effect of Musa paradisiaca stem juice on blood glucose level (BGL) of normal & diabetic rats.
The dose of 500 mg/kg bodyweight produces a significant rise of 28.3% in blood glucose level after 6h of oral administration
in normal rats. Whereas, in sub diabetic rats the same dose produces a rise of 16.4% in blood glucose levels within 1h during
glucose tolerance test (GTT) and a rise of 16% after 4 h in fasting blood glucose levels of severe diabetic cases. These results
were unexpected and important to report as other species of Musa like Musa sapientum has been reported for its hypoglycemic

This study was undertaken to evaluate the effects of n-3 fatty acid supplementation on blood glucose, lipid profile and cytokines
in humans. Twenty adult healthy subjects were supplemented with 1g/day fish oil concentrate capsules for 2 weeks. Fasting
blood samples were taken at baseline and again after 2 week intervention. Fish oil supplementation significantly lowered fasting
serum concentrations of total cholesterol, triacylglycerol, very low density lipoprotein and low density lipoprotein (P <
0.01). Also high density lipoprotein increased significantly compared with baseline value (P < 0.01). Fish oil did not significantly
altere the fasting blood glucose concentration but significantly reduced the serum cytokines (P < 0.001). This study demonstrated
that eicosapentanoic acid (EPA) and docosahexaenoic acid (DHA) supplementation altered serum lipids, lipoprotein and cytokine
concentrations in normal human subjects even at the small dose of 660 mg of EPA plus DHA per day. It appears that EPA plus
DHA can be a safe and perhaps beneficial supplement to inflammatory diseases. 相似文献

The present study investigated the effect of an antianxiety drug, buspirone on blood glucose and plasma insulin level concerning
the role of 5-HT1A receptors in blood glucose regulation in healthy humans. Twelve healthy male volunteers were administered single oral doses
of buspirone (10 mg) or placebo, in a randomized, crossover way, followed by oral glucose load (75 gm in 200 ml) at reported
Tmax i.e. the time of peak plasma concentration of the respective administered drug. The blood samples were collected as predose,
postdose and post oral glucose load at 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 hr to investigate the effect of buspirone or placebo
at basal blood glucose and plasma insulin level and after oral glucose load induced (postprandial) blood glucose and plasma
insulin level. Blood glucose and plasma insulin concentrations were estimated by glucose hexokinase method and enzyme linked
immunosorbent assay (ELISA) method respectively. The concentration of blood glucose was significantly (p<0.05) decreased after
oral glucose load following administration of buspirone in comparison with placebo however no significant change was observed
in the fasting blood glucose and plasma insulin (fasting and oral glucose load induced) level. In conclusions, the present
study findings show that buspirone produced a significant alteration in blood glucose level in healthy humans. In addition,
study results also indicate that the involvement of serotonergic (5-HT, receptors) mechanism of blood glucose regulation in

Introduction
Extremely high glucose concentrations have been shown to interfere with creatinine assays especially with Jaffe method in peritoneal dialysate. Because diabetes is the fastest growing chronic disease in the world, laboratories study with varying glucose concentrations. We investigated whether different levels of glucose spiked in serum interfere with 21 routine chemistry and thyroid assays at glucose concentrations between 17-51 mmol/L.Materials and methods
Baseline (group I) serum pool with glucose concentration of 5.55 (5.44-5.61) mmol/L was prepared from patient sera. Spiking with 20% dextrose solution, sample groups were obtained with glucose concentrations: 17.09, 34.52, and 50.95 mmol/L (group II, III, IV, respectively). Total of 21 biochemistry analytes and thyroid tests were studied on Abbott c8000 and i2000sr with commercial reagents. Bias from baseline value was checked statistically and clinically.Results
Creatinine increased significantly by 8.74%, 31.66%, 55.31% at groups II, III, IV, respectively with P values of < 0.001. At the median glucose concentration of 50.95 mmol/L, calcium, albumin, chloride and FT4 biased significantly clinically (-0.85%, 1.63%, 0.65%, 7.4% with P values 0.138, 0.214, 0.004, < 0.001, respectively). Remaining assays were free of interference.Conclusion
Among the numerous biochemical parameters studied, only a few parameters are affected by dramatically increased glucose concentration. The creatinine measurements obtained in human sera with the Jaffe alkaline method at high glucose concentrations should be interpreted with caution. Other tests that were affected with extremely high glucose concentrations were calcium, albumin, chloride and FT4, hence results should be taken into consideration in patients with poor diabetic control.Key words: assay interference, glucose interference, preanalytical phase, creatinine, Jaffe kinetic assay, thyroid function tests 相似文献7.

The estimation of electrolytes like sodium (Na+), potassium (K+) and chloride (Cl−) using direct and indirect ion-selective electrodes (ISE) is a routine laboratory practice. Interferents like proteins, triglycerides, drugs etc. are known to affect the results. The present study was designed to look into the effect of increasing glucose concentrations on estimation of Na+, K+ and Cl− by direct and indirect ISE. Pooled sera was mixed with glucose stock solution (20 g/dL) prepared in normal saline to obtain glucose concentrations ranging from ~100 to ~5000 mg/dL. Na+, K+ and Cl− levels were estimated by direct and indirect ISE analyzers and results were statistically analysed using ANOVA and Pearson’s correlation. Similar experiment was also performed in 24 h urine sample from healthy subjects. Significant difference was observed between Na+ and Cl− measurements by direct and indirect ISE, with indirect ISE values being consistently higher than direct ISE. Besides this, significant difference was observed amongst Na+ and Cl− values from baseline values obtained by indirect ISE at glucose concentrations ≥2486 mg/dL. However, no such difference was observed with direct ISE. Na+ and Cl− estimation by indirect ISE showed significant negative correlation with glucose concentration, more so, above ~2000 mg/dL. K+, however, showed no significant difference with varying glucose. Similar results were observed in 24 h urine samples with a significant difference observed amongst Na+ and Cl− values at ≥2104 mg/dL glucose. Thus we conclude that high glucose concentrations interfere significantly in estimation of Na+ and Cl− by indirect ISE in serum as well as urine.

An electrokinetic driven microfluidic lab-on-a-chip was developed for glucose quantification using double-enzyme assay. The enzymatic glucose assay involves the two-step oxidation of glucose, which was catalyzed by hexokinase and glucose-6-phosphate dehydrogenase, with the concomitant reduction of NADP+ to NADPH. A fluorescence microscopy setup was used to monitor the different processes (fluid flow and enzymatic reaction) in the microfluidic chip. A two-dimensional finite element model was applied to understand the different aspects of design and to improve the performance of the device without extensive prototyping. To our knowledge this is the first work to exploit numerical simulation for understanding a multisubstrate double-enzyme on-chip assay. The assay is very complex to implement in electrokinetically driven continuous system due to the involvement of many species, which has different transport velocity. With the help of numerical simulation, the design parameters, flow rate, enzyme concentration, and reactor length, were optimized. The results from the simulation were in close agreement with the experimental results. A linear relation exists for glucose concentrations from 0.01 to 0.10 g l−1. The reaction time and the amount of enzymes required were drastically reduced compared to off-chip microplate analysis. 相似文献

Reference intervals for pubertal characteristics are influenced by genetic, geographic, dietary and socioeconomic factors. Therefore, the aim of this study was to establish age-specific reference intervals of glucose and lipid levels among local school children. This was cross-sectional study, conducted among Saudi school children. Fasting blood samples were collected from 2149 children, 1138 (53%) boys and 1011 (47%) girls, aged 6 to 18 years old. Samples were analyzed on the Architect c8000 Chemistry System (Abbott Diagnostics, USA) for glucose, cholesterol, triglycerides, HDL and LDL. Reference intervals were established by nonparametric methods between the 2.5th and 97.5th percentiles. Significant differences were observed between boys and girls for cholesterol and triglycerides levels in all age groups (P < 0.02). Only at age 6–7 years and at adolescents, HDL and LDL levels were found to be significant (P < 0.001). No significant differences were seen in glucose levels except at age 12 to 13 years. Saudi children have comparable serum cholesterol levels than their Western counterparts. This may reflect changing dietary habits and increasing affluence in Saudi Arabia. Increased lipid screening is anticipated, and these reference intervals will aid in the early assessment of cardiovascular and diabetes risk in Saudi pediatric populations. 相似文献

Water extract of dry fruits ofTerminalia chebula (Hindi-Harda, Telugu-Karakkaya) at a dose of 200 mg/kg body weight improved the glucose tolerance as indicated by 44% of
reduction in the peak blood glucose at 2nd hour in glucose tolerance test in diabetic (streptozotocin induced) rats. Treatment of diabetic rats with an initial fasting
blood glucose of 253±9.4 mg/dl daily once with the water extract (200 mg/kg) for two weeks brought down the fasting blood
glucose to 123±8.4 mg/dl which is only slightly above the normal value. These results indicate that water extract of Terminalia
chebula improves glucose tolerance and brings down fasting blood glucose in diabetic rats. 相似文献

This paper describes a simple method for fabricating a series of poly(ethylene glycol) diacrylate (PEG-DA) hydrogel microstructures inside microfluidic channels as probe for proteins and glucose. In order to demonstrate the feasibility of this newly developed system, bovine serum albumin (BSA) was chosen as a model protein. PEG microcolumns were used for the parallel detection of multiple components. Using tetrabromophenol blue (TBPB) and the horseradish peroxidase/glucose oxidase reaction system, bovine serum albumin (BSA) and glucose in human urine were detected by color changes. The color changes for BSA within a concentration range of 1-150 μM, and glucose within a range of 50 mM-2 M could be directly distinguished by eyes or precisely identified by optical microscope. To show the practicability of the gel particle array, protein and glucose concentrations of real human urine samples were determined, resulting in a good correlation with hospital analysis. Notably, only a 5 μL sample was needed for a parallel measurement of both analytes. Conveniently, no special readout equipment or power source was required during the diagnosis process, which is promising for an application in rapid point-of-care diagnosis. 相似文献

During routine screening, some otherwise healthy individuals who showed remarkably lower post prandial (at 2 hour) plasma
glucose without any symptom were subjected to extended glucose tolerance test and a few of them to extended post meal tolerance
test as well. It was observed that post prandial (at 2 hour) plasma glucose after glucose administration was significantly
lower than the fasting level (p<0.05−p<0.001). However, post prandial plasma glucose at 2 hour after their usual meal exhibited
a significantly higher level than the fasting and post glucose level (p<0.05−p<0.001). Glucose appears to be a stronger agent
than the more natural mixed meal in these individuals in causing post prandial lowering of plasma glucose. Hence, these individuals
are to be evaluated with their usual meals before considering further investigations. Like upper limit, there is the need

Diabetes has been classified as a disease of glucose overproduction by tissues, mainly liver and glucose underutilization
by insulin requiring tissues like liver, adipose and muscle due to lack of insulin. There is, however, glucose over utilization
in tissues not dependent on insulin for glucose transport like kidney, nerve and brain. There are serious complications due
to this excess glucose in these tissues and their reversal is important for a good metabolic control and normalisation of
other parameters. Insulin, trace metals and some plant extracts have been used to see the reversal effects of the complications
of diabetes in liver and kidney in experimental diabetes. Almost complete reversal of the metabolic changes has been achieved
in the activities of key enzymes of metabolic pathways in liver and kidney and an effective glucose control has been achieved

During a dual-center study on obese and normal weight children and adolescents, focusing on glucose metabolism, we observed a marked difference in glucose results (N = 16,840) between the two sites, Salzburg, Austria and Uppsala, Sweden (P < 0.001). After excluding differences in patient characteristics between the two populations as cause of this finding, we investigated other preanalytic influences. Finally, only the tubes used for blood collection at the two sites were left to evaluate. While the Vacuette FC-Mix tube (Greiner Bio-One, Kremsmünster, Austria) was used in Uppsala, in Salzburg blood collections were performed with a lithium heparin tube (LH-Monovette, Sarstedt, Germany). To prove our hypothesis, we collected two blood samples in either of these tubes from 51 children (Salzburg N = 27, Uppsala N = 24) and compared the measured glucose results. Indeed, we found the suspected bias and calculated a correction formula, which significantly diminished the differences of glucose results between the two sites (P = 0.023). Our finding is in line with those of other studies and although this issue should be widely known, we feel that it is widely neglected, especially when comparing glucose concentrations across Europe, using large databases without any information on preanalytic sample handling. 相似文献

In persons with impaired glucose tolerance, serum glycated proteins (9.44±1.46μg hexose/mg protein) increased 1.69-fold (P<0.001)
over control subjects compared to an increase of 1.95-fold in overt diabetics. However, correlations between serum glycated
protein values and 2 h blood sugar values during the tolerance test (r=0.39) and between the former and graph area (r=0.30)
were not strong. Glycated albumin (2.25 mol hexose/mol protein) and glycated haemoglobin (0.398±0.07 mol hexose/mol protein)
in the study group also showed 1.67 and 1.24-fold increase respectively compared to controls. Fairly good correlations between
glycated serum protein and glycated albumin (r=0.50) and glycated haemoglobin (r=0.45) were observed during this study. The
data suggest that measurement of glycated proteins will be a useful index to detect impaired glucose tolerance. 相似文献

Although HbA1C is widely accepted as a useful index of mean blood glucose in type 2 diabetic patients its usefulness as screening test for
diabetes has been controversial. The present study was undertaken to determine whether the level of HbA1C predicted diabetes in a prediabetic group of subjects. Plasma lipids, oral glucose tolerance, HbA1C was determined in 90 normal control subjects, 57 offspring of one type 2 diabetes mellitus parent and 11 diagnosed type 2
diabetes mellitus individuals. The mean age of participants was 44.5 yrs (not significantly different amongst the three groups)
and the mean body mass index was 26.8 (not significantly different amongst the three groups). Two hours after a 75 g glucose
challenge, the offspring had a significantly higher plasma glucose level (mean = 7.1 mmol/L, p value = 0.002) than the normals. Similarly the HbA1C values were higher in the offspring than in the normals (mean = 5.78%, p value = 0.016). Besides the significantly higher values for oral glucose tolerance test and HbA1C, the diabetics also were significantly higher for triglycerides (mean = 2.25mmol/L), total cholesterol (mean = 6.24mmol/L)
and systolic blood pressure (mean = 138.45mm Hg) than the offspring (P value = 0.031, 0.006, 0.010) and the normals (P value = 0.026, 0.018, 0.002) respectively. The mean values of diastolic blood pressure, LDL cholesterol and HDL cholesterol

This research presents a multiple enzyme-doped thread-based microfluidic system for blood urea nitrogen (BUN) and glucose detection in human whole blood. A novel enzyme-doped thread coated with a thin polyvinylchloride (PVC) membrane is produced for on-site electrochemical detection of urea and glucose in whole blood. Multiple enzymes can be directly applied to the thread without delicate pretreatment or a surface modification process prior to sealing the thread with PVC membrane. Results indicate that the developed device exhibits a good linear dynamic range for detecting urea and glucose in concentrations from 0.1 mM–10.0 mM (R2 = 0.9850) and 0.1 mM–13.0 mM (R2 = 0.9668), which is suitable for adoption in detecting the concentrations of blood urea nitrogen (BUN, 1.78–7.12 mM) and glucose (3.89–6.11 mM) in serum. The detection result also shows that the developed thread-based microfluidic system can successfully separate and detect the ions, BUN, and glucose in blood. The calculated concentrations of BUN and glucose ante cibum (glucose before meal) in the whole blood sample are 3.98 mM and 4.94 mM, respectively. The developed thread-based microfluidic system provides a simple yet high performance for clinical diagnostics. 相似文献

Non-insulin dependent diabetes mellitus (NIDDM) is the most rapidly growing chronic metabolic disorder in the world. With
advancement in the age and duration of diabetes there is a gradual tendency for the level of blood sugar to rise along with
a subsequent increase in the HbA1c as well as in the fasting insulin level. Whether this is an aging process or increased
frequency of diabetes is still controversial. The correlation between glucose and insulin sensitivity is consistent with the
idea that the degree of chronic hyperglycemia is a cause of excessive insulin resistance in type 2 diabetes, i.e. the insulin
resistance which characterizes type 2 diabetes but not nondiabetic subjects matched for age, gender, family history and duration
of diabetes. The study comprised a total of 76 subjects out of which 30 were normal, non-diabetic persons and the rest 46
were diabetics with different duration of time in years, after being diagnosed diabetic. Data was analyzed after dividing
the subjects into four groups—Group 1 comprised of one year old diabetics, Group 2 was made up of those, who had diabetes,
for the past 2–5 years, Group 3 included patients who were diabetic since more than 5 years and Group 4 included non-diabetics
as the normal control group. The results obtained indicated that the HbA1c levels showed a significant increase with the duration
of diabetes as well as the insulin level showed a significant correlation after adjustment for age, sex and duration of diabetes. 相似文献

Monitoring cellular bioenergetic pathways provides the basis for a detailed understanding of the physiological state of a cell culture. Therefore, it is widely used as a tool amongst others in the field of in vitro toxicology. The resulting metabolic information allows for performing in vitro toxicology assays for assessing drug-induced toxicity. In this study, we demonstrate the value of a microsystem for the fully automated detection of drug-induced changes in cellular viability by continuous monitoring of the metabolic activity over several days. To this end, glucose consumption and lactate secretion of a hepatic tumor cell line were continuously measured using microfluidically addressed electrochemical sensors. Adapting enzyme-based electrochemical flat-plate sensors, originally designed for human whole-blood samples, to their use with cell culture medium supersedes the common manual and laborious colorimetric assays and off-line operated external measurement systems. The cells were exposed to different concentrations of the mitochondrial inhibitor rotenone and the cellular response was analyzed by detecting changes in the rates of the glucose and lactate metabolism. Thus, the system provides real-time information on drug-induced liver injury in vitro. 相似文献
