Human thioredoxin reductase (TrxR) system is associated with cancer cell growth and anti-apoptosis process. Effects of 1,2-[bis(1,2-benzisoselenazolone-3(2H)-ketone)]ethane (BBSKE), a novel TrxR inhibitor, were investigated on human leukemia cell lines HL-60 and K562. BBSKE treatment induced cell growth inhibition and apoptosis in both cell lines. Apoptosis induced by BBSKE is through Bcl-2/Bax and caspase-3 pathways. Ehrlich's ascites carcinoma-bearing mice were used to investigate the anti-tumor effect of BBSKE in vivo. Tumor-bearing mice treated with BBSKE showed an increase of life span with a comparable effect to cyclophosphamide (CTX). These results suggest a potential usage of BBSKE as a therapeutic agent against non-solid tumors.  相似文献

The treatment of relapsing primary nephrotic syndrome in children   总被引:2,自引:0,他引:2  
Objective: To explore better therapy and reduce the rate of re-relapse of primary nephritic syndrome in children who had been treated with corticosteroids but relapsed. Methods: Eighty relapsers were enrolled from Jan. 1994 to Apr. 2000, who were randomly divided into two groups. The treatment group (n=39) had been treated with tripterysium glucosides for three months, with the control group (n=41) members were treated with cyclophosphmide (CTX) by intermission intravenous pulse, with total dose of CTX not being more than 150 mg/kg. Prednisone, meanwhile, was given to both groups. The total treatment period of prednisone was prolonged by 12-18 months. Results: After following up for 3-7 years, the re-relapse rates of both groups were observed. The re-relapse rate of the treatment group was 28.2% to 29.3% in the CTX-controlled group. The re-relapse rates between two groups were almost similar, and with no observed significant difference (P>0.05). The side effect of tripterysium glucosides was less than that of CTX. Conclusion: For the treatment of relapsing nephritic syndrome in children, the combination of tripterysium glucosides and prolonged corticosteroid therapy is as effective as the regimen of CTX plus prolonged use of prednisone.  相似文献

The effects of select drugs, dosages, and modes of administration upon learned taste aversions were compared among groups of wild-caught male and female Philippine rice rats (R. r. mindanensis). Experiment 1 compared two-choice saccharin aversions for 28 days among groups intubated with copper sulfate, cyclophosphamide, lithium chloride, red squill, sodium chloride (control), or deionized water (control). Main results were that 375 mg/kg lithium chloride produced the greatest sustained aversions, whereas 198 mg/kg cyclophosphamide and 210 mg/kg red squill produced moderate aversions, with males showing more resistance to extinction than females. Experiment 2 compared saccharin aversion among matched groups of male and female rats that received low (36 mg/kg), moderate (105 mg/kg), or high (368 mg/kg) dosages of lithium by gavage, ip injection, or ingestion. Sex differences in rates of extinction were found for the ingestion and injection-dosed rats, but no sex difference was again found for rats dosed by gavage. A significant mode × day interaction indicated that extinction progressed more rapidly for rats dosed by gavage. For all modes of administration, high dosages yielded intense 28-day aversion, moderate dosages produced intermediate 3–5 day aversion, and low dosages caused no aversion.  相似文献

研究目的:在小鼠模型中利用吲哚美辛阻断COX-1/2通路,探讨非甾体类抗炎药对肾缺血再灌注损伤的保护作用。创新要点:非甾体类抗炎药被认为具有肾毒性,本研究首次在小鼠模型中探讨非甾体类抗炎药对肾缺血再灌注损伤的保护作用。研究方法:小鼠左侧肾蒂夹闭后,通过腹腔注射不同剂量的吲哚美辛,在肾缺血再灌注损伤24小时后,获取血液和肾脏标本。利用酶联免疫(ELISA)试剂盒测定血清肌酐和细胞因子浓度来评估肾功能,肾组织样本进行苏木精-伊红染色和免疫组化分析。重要结论:腹腔注射吲哚美辛5 mg/kg组的小鼠血清肌酐值与对照组相比显著降低,肾小管损伤也显著减轻(见图1和2);腹腔注射5和7 mg/kg吲哚美辛组的小鼠血清肾肿瘤坏死因子-α和白介素-6的浓度显著降低(见图3a和3b);腹腔注射3和5 mg/kg吲哚美辛组的小鼠血清血栓素B2与6-酮前列腺素F1α的比值明显降低(见图3c);腹腔注射5 mg/kg吲哚美辛组小鼠肾组织COX-1和COX-2染色较弱(见图4)。因此,吲哚美辛对小鼠肾缺血再灌注损伤的作用与其剂量相关,在某个特定的剂量范围内具有肾保护作用。吲哚美辛对小鼠肾缺血再灌注损伤的保护作用与阻断COX-1/2有关。  相似文献

Bulblet development is a problem in global lily bulb production and carbohydrate metabolism is a crucial factor. Micropropagation acts as an efficient substitute for faster propagation and can provide a controllable condition to explore bulb growth. The present study was conducted to investigate the effects of humic acid (HA) on bulblet swelling and the carbohydrate metabolic pathway in Lilium Oriental Hybrids ‘Sorbonne’ under in vitro conditions. HA greatly promoted bulblet growth at 0.2, 2.0, and 20.0 mg/L, and pronounced increases in bulblet sucrose, total soluble sugar, and starch content were observed for higher HA concentrations (≥2.0 mg/L) within 45 d after transplanting (DAT). The activities of three major starch synthetic enzymes (including adenosine 5′-diphosphate glucose pyrophosphorylase, granule-bound starch synthase, and soluble starch synthase) were enhanced dramatically after HA application especially low concentration HA (LHA), indicating a quick response of starch metabolism. However, higher doses of HA also caused excessive aboveground biomass accumulation and inhibited root growth. Accordingly, an earlier carbon starvation emerged by observing evident starch degradation. Relative bulblet weight gradually decreased with increased HA doses and thereby broke the balance between the source and sink. A low HA concentration at 0.2 mg/L performed best in both root and bulblet growth. The number of roots and root length peaked at 14.5 and 5.75 cm, respectively. The fresh bulblet weight and diameter reached 468 mg (2.9 times that under the control treatment) and 11.68 mm, respectively. Further, sucrose/starch utilization and conversion were accelerated and carbon famine was delayed as a result with an average relative bulblet weight of 80.09%. To our knowledge, this is the first HA application and mechanism research into starch metabolism in both in vitro and in vivo condition in bulbous crops.  相似文献

本实验对松寿丹(SSD)对实验性小鼠心脑缺氧的保护作用及与血脂的关系进行了研究。实验结果证明:①SSD对NaNO_2中毒所致急性血液性缺氧有保护作用,1.6g/kgP<0.05;②能显著降低高脂饲养动物血清胆固醇含量,0.8g/kg和0.4g/kg两组均P<0.01;③对高脂饲养动物耐NaNO_2缺氧有保护作用,0.8g/kg组P<0.05。提示松寿丹不仅对单纯血液性缺氧有保护作用,而且还通过其显著的降血脂作用改善循环性缺氧,提高机体耐缺氧能力。  相似文献

探讨氟苯尼考的小鼠免疫器官和脾淋巴细胞毒性及其对小鼠肌肉理化特性的影响,本研究采用高、中、低剂量(40 mg/kg、20 mg/kg、10 mg/kg)对小鼠连续7d腹腔注射给药,每日一次;通过计算小鼠胸腺和脾脏器指数、观察脾淋巴细胞形态、测量肌肉蛋白含量和pH值等做为实验内容.结果显示:与对照组比较,胸腺和脾脏器指数除高剂量组数值明显减小外(P<0.05),中、低剂量组对应值降低都不明显(P>0.05);肌肉蛋白的含量和pH值下降不明显(P>0.05);脾淋巴细胞形态除高剂量组形态异常外,其余各组变化不大.说明氟苯尼考对小鼠有一定的免疫毒性,对肌肉蛋白质的含量、肌肉pH影响较小.  相似文献

为探讨金樱子多糖对免疫抑制小鼠肝功能及血脂代谢的调节作用,取50只昆明小鼠随机平均分为正常对照组、模型组、RLP低剂量组、RLP中剂量组、RLP高剂量组.低、中、高剂量组分别按100ms/kg·d、200mg/kg·d、400mg/kg·d连续灌胃30d;正常对照组和模型组灌胃生理盐水.于试验前一周在继续灌胃的基础上,除正常对照组外,分剐于第l、3、5天给小鼠按25mg/kg体重腹腔注射地塞米松(Dex)建立免疫抑制动物模型.摘小鼠眼球取血,用试剂盒测定小鼠血清的总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL—C)、低密度脂蛋白胆固醇(LDL—C)、谷草转氨酶(AST)、谷丙转氨酶(ALT).结果表明,金樱子多糖能显著降低免疫抑制小鼠血清中TC、LDL—C、AST、Au含量,对HDL—C和TG有一定的调节作用.因此,金樱子多糖对免疫抑制小鼠肝功能及血脂代谢具有一定的调节作用.  相似文献
