The effect of total dissolved gas (TDG) supersaturation on fish living downstream of dams is one of the main ecological risks of high dam construction. A strategy for mitigating the negative effects is needed urgently since many high dams are under construction in the upper reaches of the Yangtze River in China. Experiments on the hatching process of David’s schizothoracin were carried out and the results show that the hatching rate decreased with increasing TDG levels, and that most eggs hatched within a very short time in the higher TDG saturation groups. By using a stereomicroscope, damages to the head, yolk sac, body, anus, etc. were found in larvae which hatched in TDG supersaturated water. Results show that the lesion rate increased with increasing TDG levels. Furthermore, 7-d-old David’s schizothoracin were exposed to TDG supersaturated water levels of 100%, 105%, 110%, 115%, 120%, 125%, 130%, 135%, and 140% for testing their tolerance to TDG supersaturation. We found that the median lethal concentrations (LC50 ) for 13, 14, 20, 35, 52, 73, and 96 h exposure were 138%, 138%, 134%, 130%, 129%, 128%, and 126%, respectively. The median lethal times (LT50 ) were 7.49, 11.04, 19.25, and 35.38 h for exposure to water with TDG levels of 145%, 140%, 135%, and 130%, respectively.  相似文献

In this research, the changing rules of billet forged Ti-22 Al-24 Nb alloy with various solution time were studied, and the mechanical properties of different microstructure were tested at the room temperature. The results indicate that the(α2+O+B2) three-phase microstructure with equiaxed grains was acquired by Ti-22 Al-24 Nb alloy billet forging, and the average size of the grains was about 300 μm. With the increase of solution time, the primary lath-shaped O phase began to dissolve, and then the equiaxial α2 phase started to dissolve, but the undissolved α2/O phase began to be equiaxial and grow. The grains of original B2 phase recrystallized and grew. After the 2 h solution treatment, the recrystallization of grains was completed basically and the average size of the grains was about 100 μm. After the 2.5 h solution treatment, the strength of the alloy decreased, and the plasticity increased. However, when the solution treatment increased to 3 h, the plasticity decreased but the strength increased. The optimal solution parameters of Ti-22 Al-24 Nb alloy were the holding time of 2 h, the solution temperature of 1 000 ?C, and water cooling. Excellent comprehensive mechanical properties can be accquired under these parameters. The tensile strength, the yield strength, the elasticity modulus, the elongation, and the section shrinkage were 950 MPa, 915 MPa, 90 GPa, 15.69% and 42.28%, respectively.  相似文献

Insecticidal effects of different doses of the dust and methanol extracts of Garcinia kolae on Collosobruchus maculatus and Sitophilus zeamais were tested. The dust had no significant effect on the two insects; none of them died even at 3 d after treatment. The methanol extracts, however, had rapid lethal effects on both C. maculatus and S. zeamais. The mortality of C. maculatus by the lowest concentration of methanol extracts ranged from 95%∼100% whereas in S. zeamais, the mortality ranged from 87.5%∼100% and 70%∼100% in concentrations of 1 g extract+3 ml methanol and 1 g extract+5 ml methanol, respectively, from 24 to 48 h. The least concentration of 1 g extract+15 ml methanol had no significant lethal effect on Sitophilus zeamais.   相似文献

The objective of this study is to propose a more accurate and faster MTT 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide colorimetric assay (MCA) for quantitative measurement of polypeptide bacteriocins in solutions with nisin as an example. After an initial incubation of nisin and indicator bacterium Micrococcus luteus NCIB 8166 in tubes, MTT was added for another incubation period. After that, nisin was quantified by estimating the number of viable bacteria based on measuring the amount of purple formazan produced by cleavage of yellow tetrazolium salt MTT. Then MCA was compared to a standard agar diffusion assay (ADA). The results suggested a high correlation coefficient (r2=0.975±0.004) between optical density (OD) and the inhibitory effect of nisin on a bacterial strain Micrococcus luteus NCIB 8166 at a range of 0.125~32 IU/ml. The MCA described in this study was very quick. Quantification of nisin took only 7~8 h and the detection limit was at the level of 0.125 IU/ml when compared to 12 IU/ml and 24~28 h for ADA. The MCA provides an accurate and rapid method for quantifi-cation of nisin in solutions and is expected to be used for quantification of other antimicrobial substances.  相似文献

Pseudomonas aeruginosa strain (AS 1.50) and Bacillus subtilis strain (AS 1.439) from Ming lake were decomposed by photocatalytic nanostructure N-TiO2 thin films in a photo-reactor under UV irradiation. The different thickness nanostructure N-TiO2 thin films coated on mesh grid were prepared by sol-gel method and immobilized at 500 ℃ (films A) or 350 ℃ (films B) for 1 h in a muffle furnace. The results showed that N-TiO2 thin film B (8.18 nm thickness, 2.760 nm height and 25.15 nm diameter) has more uniform granular nanostructure and thinner flat texture than N-TiO2 thin film A (12.17 nm thickness, 3.578 nm height and 27.50 nm diameter). The bactericidal action of N-TiO2 thin film A and film B for Pseudomonas aeruginosa strain (AS 1.50) and Bacillus subtilis varniger strain (AS1.439) were investigated in this work. More than 95% of photocatalytic bactericidal efficiency for Pseudomonas aeruginosa strain (AS 1.50) and 75% for Bacillus subtilis strain (AS 1.439) were achieved by using N-TiO2 thin films-B for 70-80 rain of irradiation during the photo-bactericidal experimental process. The results indicated that the photo-induced bactericidal efficiency of N-TiO2 thin films probably depended on the characteristics of the films.  相似文献

通过对杂交鳜胚胎发育的连续观察,研究了其胚胎发育的时序和特点.结果显示,杂交鳜的受精卵呈淡黄色圆球形,卵径为1.52±0.16mm,油球多个且大小不一.杂交鳜胚胎发育的主要特征与已报道的鳜鱼胚胎发育相同,可分为胚盘形成阶段、卵裂阶段、囊胚阶段、原肠胚阶段、神经胚阶段、器官形成阶段和出膜阶段7个阶段及27个发育时期.在水温19.5±1.2℃条件下,34h50min开始形成器官,受精约98h后仔鱼孵出,孵出的仔鱼全长为3.55±0.10mm.胚胎发育总积温为1 982.38℃·h.  相似文献

Simple procedures for extraction and chromatographic determination ofdimethachlon residues in fresh tobacco leaves and cut-tobacco are described. The determination was carried out by capillary gas chromatography (GC) with electron capture detection (ECD) and confirmed by GC-MS. The mean recoveries and relative standard deviation (RSD) were 93.2%-112.9% and 3.5%-6.7%, respectively at levels ranging from 0.01 to 0.1 mg/kg. The limit of determination was 0.001 mg/kg. Tobacco samples in routine check were successfully analyzed using the proposed method.  相似文献

The effects of dietary supplementation with Clostridium butyricum on growth performance and humoral immune response in Miichthys miiuy were evaluated. One hundred and fifty Miichthys miiuy weighing approximately 200-260 g were divided into five groups and reared in 15 tanks with closed circuiting culture system. The animals were fed 5 diets: basal diet only (control) or supplemented of the basal diet with C. butyricum at doses of 10^3 (CB1), 10^5 (CB2), 10^7 (CB3) or 10^9 (CB4) CFU/g. Compared with the control, the serum phenoloxidase activity was significantly increased by the supplementation (P〈0.05), acid phosphatases activity was increased significantly (P〈0.05) at the doses of 10^9 CFU/g. Serum lysozyme activity peaked at dose of 10^7 CFU/g and in the skin mucus at dose of 10^9 CFU/g. Immunoglobulin M level in the serum and skin mucus was increased except at dose of 10^3 CFU/g (P〈0.05). The growth at the dose of 10^9 CFU/g was higher than that of the control (P〈0.05). It is concluded that supplementation of C. butyricum can mediate the humoral immune responses and improve the growth performance in Miichthys miiuy.  相似文献
