Circadian rhythm of serum glucose, pyruvate, lactate and lactate dehydrogenase (LDH) was studied in normal healthy young volunteers
at the onset (I Group, 7 cases) and peak (II Group, 8 cases) of winters in India with highly varied temperature. There was
a significant difference, in the circadian rhythm of glucose, pyruvate and lactate dehydrogenase (LDH) of the two groups.
A clockwise shift was observed in acrophase except in serum lactate, which is related to the energy demand in association
with time qualified changes in diurnal activity of the individuals and change in photoperiod. 相似文献

Erythrocyte membrane protein glycosylation increase by 3.4 fold in diabetes. Insulin or sulfonylurea treatment did not reduce
the extent of glycosylation. The serum protein glycosylation was comparable in all the groups including control. Erythrocyte
membrane Na+,K+-ATPase activity decreased in the diabetics; only insulin treatment partly restored the activity. Erythrocyte membrane acetylcholinesterase
activity decreased only in the sulfonylurea treated group. Serum butyrylcholinesterase activity was relatively low in the
diabetic and insulin treated diabetic groups. The Km and Vmax of the two components of Na+,K+-ATPase from erythrocyte membranes were differently affected in the diabetic and the two treatment groups. The Vmax of acetylcholinesterase
decreased only in the sulfonylurea treated group. Diabetic states resulted in decreased Vmax of components I and II of serum
butyrylcholinesterase. In insulin-treated diabetics, component II was absent. Sulfonylurea group resembled diabetics.In vitro incubation with insulin differentially affected the Na+,K+-ATPase and serum butyrylcholinesterase activities. 相似文献

The activity of enzymes of glycolysis has been studied in erythrocytes from type-II diabetic patients in comparison with control. RBC lysate was the source of enzymes. In the diabetics the hexokinase (HK) activity increased 50 % while activities of phosphoglucoisomerase (PGI), phosphofructokinase (PFK) and aldolase (ALD) decreased by 37, 75 and 64 % respectively but were still several folds higher than that of HK. Hence, it is possible that in the diabetic erythrocytes the process of glycolysis could proceed in an unimpaired or in fact may be augmented due to increased levels of G6P. The lactate dehydrogenase (LDH) activity was comparatively high in both the groups; the diabetic group showed 85 % increase. In control group the HK, PFK and ALD activities showed strong positive correlation with blood sugar level while PGI activity did not show any correlation. In the diabetic group only PFK activity showed positive correlation. The LDH activity only in the control group showed positive correlation with marginal increase with increasing concentrations of glucose. 相似文献

Sepsis is associated with various metabolic derangements as a consequence of inflammatory response, ischemia and oxidative stress. Four parameters of relevance are procalcitonin (PCT), ischemia modified albumin (IMA) pH and lactate. The study was carried out to highlight the concomitant occurrence of sepsis, ischemia and lactic acidosis, all of which could have deleterious effects on organ function. 26 critically ill patients with a provisional diagnosis of sepsis were the test subjects. The control group had 25 apparently healthy volunteers. PCT, lactate and IMA were assayed. PCT was estimated on an automated analyser using electro-chemiluminescence. Lactate and pH were estimated on a blood gas analyzer. Serum IMA was estimated spectrophotometrically by Albumin Cobalt Binding Test. Statistical tools like students ‘t’ test and Venn diagram were employed to depict the outcome of the study. All critically ill patients had significantly higher IMA levels (0.96746 ± 0.73407) as compared to the control group (0.00728 ± 0.00895) with a p value of <0.0001. The Venn diagram was used to depict the finding that all 26 test subjects had elevated levels of IMA, of which PCT was elevated in 22 and lactate in 20. Both PCT and lactate were abnormal in 17 patients. The most significant observation was that all critically ill patients, irrespective of the presence of sepsis or lactic acidosis had elevated levels of IMA which is clearly indicative of the ubiquitous presence of oxidative stress. The Venn diagram is an elegant representation of the concurrent multiple pathophysiological processes which occur in critically ill patients. 相似文献

The present study was undertaken in 100 subjects, 30 diabetics without complication (group I), 40 diabetics with retinopathy
(group II) and 30 non diabetic as normal control group (group III). Blood sugar levels, magnesium, cholesterol and triglyceride
were analyzed from plasma and serum. The results were correlated with degree of diabetic control from the levels of glycosylated
hemoglobin. Serum magnesium levels in group II were found to be significantly lowered than in group I. There was also significant
difference in magnesium levels of group I and group III. We found a significant correlation between the glycosylated hemoglobin
and magnesium levels in our study.
The results also indicate that the patients with diabetic retinopathy showed significant rise in serum cholesterol and triglyceride.
Probably hypomagnesemia and increased serum cholesterol and triglyceride levels are responsible for microvascular changes
in diabetes leading to retinopathy. The purpose of this study was thus to gather information about the degree of control of

In diabetes, persistent hyperglycemia results in increased production of free radicals especially oxygen free radicals, which can cause cell destruction and tissue injury resulting in cell dysfunction. With the premise that oxidative stress is a major cause of diabetic complications, we conducted a controlled laboratory based investigation on level of lipid peroxide levels in the serum of Type 1 and Type 2 diabetic patients attending Muhimbili National Hospital. From our clinical data it was observed that majority of the patients had higher waist to hip ration and body mass index, which suggests that the patients were either overweight or obese. The enrolled diabetic patients had higher lipid peroxide levels than controls and also Type 2 patients had higher lipid peroxide levels than Type 1 patients. Moreover, patients with known complications had higher lipid peroxide levels than patients without complications. The lipid peroxide levels in the diabetic patients were significantly different from that of the control subjects enrolled in the study. A majority of the diabetic patients had a poorly controlled blood sugar. Our finding hints that despite the fact that diabetic patients in our clinic are on follow up, they are at a risk of developing coronary heart diseases, neuropathy and other secondary diabetic complications. 相似文献

The aim of the present study was to evaluate the antidiabetic and ameliorative potential of aqueous extract of Ficus bengalensis
bark in streptozotocin induced diabetic rats. The effect of oral administration of aqueous extract of F. bengalensis bark
on blood glucose, serum electrolytes, serum glycolytic enzymes, liver microsomal protein, hepatic cytochrome P-450 dependent
monooxygenase enzymes and lipid peroxidation in liver and kidney of streptozotocin -induced diabetic rats was studied. Oral
administration of Ficus bengalensis to fed, fasted and glucose loaded diabetic rats significantly [F > 0.05 (ANOVA) and P<
0.05 (DMRT)] decreased the blood glucose level at 5 hrs and restored the levels of serum electrolytes, glycolytic enzymes
and hepatic cytochrome P-450 dependent enzyme systems and decreased the formation of liver and kidney lipid peroxides at the
end of 12 weeks. Further, the aqueous extract of Ficus bengalensis at a dose of 500mg/kg/day exhibits significant antidiabetic
and ameliorative activity as evidenced by histological studies in normal and Ficus bengalensis treated streptozotocin induced
diabetic rats. On the basis of our findings, it could be used as an antidiabetic and ameliorative agent for better management

The rate of glucose consumption and pyruvate and lactate production in red blood cells of normal, sickle cell trait and SS disease subjects were measured. Glucose consumption was found to be increased significantly (p<0.05) in red cells of sickle cell patients than the sickle cell trait and normal persons. There was also an increased rate (p<0.05) of pyruvate formation and a significant (p<0.05) increase in lactate formation in sickle red cells. 相似文献

Usually CPK, CK-MB and lactate dehydrogenase (LDH) enzymes are measured in blood during the period of myocardial infarction.
The changes in LDH assay indicates duration of the infarction.
The level of lactate dehydrogenase is 500 times greater in tissue than those found in serum. A small mass of damaged tissue
causes leakage of enzyme and increases its level in serum.
Any cause of tissue break down/hemolysis which is sufficiently severe can produce LDH pattern similar to that in myocardial
infarction.
We report this case of small bowel volvulus showing significant increase in LDH.
It may be due to tissue necrosis which increases the level of LDH in serum. 相似文献

The supply of oxygen is limited in certain intra abdominal conditions due to direct vascular invasion or inflammatory process, resulting in high lactate levels. Aim of this study was to find the predictive value of lactate levels in the peritoneal fluid (PF) and blood of patients with acute abdomen. The study comprised of fifty patients with acute abdominal conditions, admitted in emergency ward of tertiary care hospital, thirty patients were with surgical abdomen (group I) and twenty patients with non surgical abdomen (group II). Lactate was estimated in PF and blood on Blood Gas Analyzer (NOVA, M-7). The mean lactate levels in PF were significantly higher in group I as compared to group II (14.65 ± 1.195 vs. 5.92 ± 0.97 mmol/L, p < 0.001). There was no significant difference in blood lactate levels in both the groups. When PF and blood lactate levels were compared within groups, we found that PF levels were significantly higher than blood in group I (14.65 ± 1.195 vs. 3.85 ± 0.54 mmol/L, p < 0.001) but not in group II (5.92 ± 0.97 vs. 4.36 ± 0.95 mmol/L). Diagnostic value was obtained using ROC curve. Cut off values obtained for PF lactate, difference and ratio of PF and blood lactate (≥6.4 mmol/L, ≥3.3 and ≥2.1 respectively) are at very high degree of sensitivity and specificity. So it can be useful marker of surgical emergency in patients with acute intra abdominal pathology, especially in clinically ill patients or in whom physical examination is not yielding because of neurologic disorders or unresponsiveness. 相似文献

The influence of Crataeva nurvala bark decoction was studied in calcium oxalate stone forming rats, in relation to oxalate
metabolism in liver. The activities of the major oxalate synthesizing enzymes in liver namely, glycollate oxidase (GAO) and
lactate dehydrogenase (LDH) were significantly increased in the calculogenic group. Bark decoction treatment lowered the liver
GAO activity considerably. Transport ATPases (Na+, K+ and Ca2+-ATPases) and alkaline phosphatase were enhanced in rats fed calculi producing diet, while the activities of acid phosphatase,
inorganic pyrophosphatase and aminotransferases were slightly reduced. Bark decoction administration produced a marginal decrease
in Na+, K+-ATPase and increase in aspartate aminotransferase activities, without significantly altering other enzyme activities. The
decrease in liver GAO activity seen during bark decoction treatment, with concomitant decrease in kidney oxalate level, may
prove beneficial as a prophylactic measure in preventing stone recurrence. 相似文献

Adenosine deaminase and protein tyrosine phosphatase activities in liver and peritoneal macrophages were estimated in control,
streptozotocin induced diabetes and insulin treated diabetic groups of Swiss albino mice. Both the enzyme levels were elevated
while glycogen content decreased in the liver of diabetic group of animals as compared to controls, while in insulin treated
group, they were comparable to control levels. In macrophages also, both adenosine deaminase which plays an important role

We have studied the expression of lactate dehydrogenase and its isoforms in gall bladder cancer, cholelithiasis and chronic
cholecystitis. Quantitative and qualitative assays of lactate dehydrogenase and its various isoforms were carried out in the
blood sera of patients and healthy controls along with parallel estimation of various liver function test enzymes. Statistical
analysis was done using the software Graph Pad Prism. Significantly high expression of lactate dehydrogenase along with alkaline
phosphatase and total bilirubin (P ≤ 0.05) was observed in all the three clinical conditions as compared to controls. LDH showed an increasing trend from stage
I to stage IV of GBC indicating a significant positive association with the disease progression. The levels of LDH 3 and 4
isoforms appeared significantly more elevated in GBC than cholelithiasis or chronic cholecystitis. We suggest that a careful
estimation of total LDH and its isoforms 3 and 4 alone or along with alkaline phosphatase and total bilirubin during different
clinical stages, like chronic cholecystitis, cholelithiasis and GBC, may prove to be a potentially useful biomarker in the
prognostic management of gall bladder diseases, specifically GBC. 相似文献

To study the protective role of Arogh on isoproterenol induced myocardial damage in rats. The effect of Arogh pretreatment
on isoproterenol induced myocardial damage was assessed by studying the levels of lipid peroxides and changes in the activity
of marker enzymes such as creatine kinase, lactate dehydrogenase and transaminases in serum and heart tissue. In isoproterenol
administered rats, a significant decrease was observed in the activity of marker enzymes in the heart with a corresponding
increase in their levels in serum. Lipid peroxide levels increased significantly in the serum and heart. In rats pretreated
with arogh, the level of lipid peroxides and the activity of marker enzymes were maintained at near normal values. Pretreatment
with Arogh offered a protective effect against isoproterenol induced myocardial damage in rats as evidenced by LDH isoenzyme
patterm and histopathological studies of heart tissue. 相似文献

Non enzymatic glycosylation of IgG and their urinary excretion in patients with diabetic nephropathy
Diabetic nephropathy is a major cause of end stage renal disease. Increased excretion of albumin has widely been recognized
as an early manifestation of diabetic nephropathy particularly in subjects with diabetes mellitus. However, certain other
proteins besides albumin may be excreted in high amount during early phase of diabetic nephropathy. The serum and urinary
IgG, Glycosylated hemoglobin, fructosamine and glycosylated IgG were evaluated in the present study. Thirty-two patients of
Type 2 Diabetes without any complications, thirty-one patients of Type 2 Diabetes with nephropathy, twenty-six patients of
non-diabetic nephropathy and forty normal healthy individuals were enrolled in this study. Subjects were grouped based on
their serum creatinine level. Serum IgG, glycosylation of IgG and urinary IgG excretion were increased significantly in diabetic
patients compared to healthy controls, which were further increased significantly in chronic renal failure patients with respect
to the clinical stage of nephropathy. A positive correlation was observed between glycosylation of IgG and IgG excretion (R2=0.5995, 0.7114 respectively) in diabetic patients without any complications and diabetic nephropathy patients only, suggesting
a significant role of IgG glycosylation in the vascular clearances of IgG during diabetic nephropathy. 相似文献

The objective of the study was to estimate the serum concentrations of oxidative stress markers-lipid hydroperoxide (MDA-Malondialdehyde),
lactate dehydrogenase (LDH), nitrite; and ferric reducing antioxidant power (FRAP) — a measure of total antioxidant capacity
in serum. Serum nitrite levels were high in fertile women than in infertile women (p<0.001), whereas MDA was high in infertile
women than in fertile group (p<0.018). On classifying the entire group of women with infertility based on the etiology, serum
MDA and LDH levels were higher in women with infertility due to unexplained factor than in women with other causes contributing
to infertility compared to controls (p<0.001, p<0.02). A positive correlation among serum prolactin and nitrite suggested
that hyperprolactinemia could contribute to infertility by inducing oxidative damage. Serum FRAP levels showed no significant
difference in the groups studied. Lipid peroxidation was high in women with unexplained infertility which may be due to hyperprolactinemia. 相似文献

The objective of the present study is to evaluate the beneficial effect of tomatoes, which are rich source of Lycopene, relatively
a new carotenoid known to play an important role in human health and disease. In this study lipid peroxidation rate was measured
by estimating Malondialdehyde and the levels of serum enzymes involved in antioxidant activities like Super Oxide Dismutase,
Glutathione Peroxidase, Glutathione Reductase, Reduced Glutathione, in type-II diabetic group (n=40) and age matched control
group (n=50), and observed significantly lower levels of antioxidant enzymes and very high lipid peroxidation rate in type-II
diabetes when compared to control group (p<0.001). Short term supplementation with tomatoes (cooked) to diabetic group for
a period of 30 days, showed a significant improvement in antioxidant enzyme levels (p<0.001) and decreased lipid peroxidation
rate (p<0.001) suggesting the supplementation with tomato lycopene may serve as the best method of preventing the oxidative

In this study, we compared the lipid profile and antioxidant enzymes of normal and diabetic subjects consuming two different
types of oil as cooking medium. 70 normal, healthy subjects were taken as controls and 70 subjects with Type 2 diabetes were
recruited in patient group. Each group was further subdivided into two subgroups of 35 subjects each, consuming coconut oil
and sunflower oil respectively as cooking medium. Samples of blood were collected and analyzed for serum total cholesterol,
triacylglycerols, and cholesterol in lipoprotein fractions. Total glutathione and glutathione peroxidase were measured in
erythrocytes and superoxide dismutase in serum. Triacylglycerols, LDL and VLDL cholesterol levels were high in the diabetic
subjects compared to the controls. Total glutathione and glutathione peroxidase values showed significant decrease in diabetic
subjects as compared to the controls, while superoxide dismutase values showed significant difference between coconut oil
consuming groups. Though lipid profile parameters and oxidative stress were high in Type 2 diabetic subjects compared to controls,
no pronounced changes for these parameters were observed between the subgroups (coconut oil vs. sunflower oil). 相似文献

Objective To investigate the protective effect of nicorandil and amlodipine in isoproterenol induced myocardial necrosis in rats.
Methods The effect of nicorandil and amlodipine on bio-chemical parameters during isoproterenol induced myocardial necrosis in rats
was examined by determining the activity of creatine phosphokinase (CK), lactate dehydrogenase (LDH), and transaminases (AST,
ALT) in the serum of the animals.
Results Isoproterenol (150mg/kg/day) administered rats showed statistically significant rise in activities of LDH(1.02±0.19 to 1.85±0.05),
CK(4.3±0.19 to 7.37±.27), AST(0.38±.03 to 0.78±.05) and ALT(0.19±.017 to 0.346±.027) in the serum. Pre-co-treatment of rats
with nicorandil and amlodipine significantly lowered the raised levels of these enzymes and thereby restoring the enzyme activity
to near normal as was clear from the chart i.e. LDH was 1.10±0.04, CK was 4.37±0.19, AST and ALT were 0.39±0.028 and 0.199±0.04
respectively.
Conclusion It is demonstrated that pre-co-treatment with nicorandil and amlodipine either alone or in combination help in providing protective
effect on isoproterenol induced myocardial necrosis. 相似文献

Diabetes Mellitus in obese and non-obese Indian individuals.AIMS: Effect of Obesity and insulin resistance on diabetic control.SETTINGS AND DESIGN: 50 each groups Diabetic individuals obese and non-obese.METHODS AND MATERIAL: On selected 50 each group diabetic patient and normal, following blood investigations has been performed—Plasma Glucose,
Glycohemoglobin and Serum Insulin.STATISTICAL ANALYSIS USED: Individuals patient’s results were analyzed and compared with the normal controls.RESULTS AND CONCLUSION: The changes in glycosylated haemoglobin are mainly proportional to the post lunch glucose level (r=0.773) (p<0.01) and not
correlated to circulating insulin or the body mass index. However the levels were higher in obese diabetes (Type I and II
both) than in non-obese. Mechanism of resistance in insulin receptor interactions due to obesity is well known. However, obesity
does not seem to affect directly glycosylated haemoglobin. Under such circumstances, the reduction of weight for a diabetic
person can improve sugar control by minimizing insulin resistance and thereby can improve glycosylated haemoglobin levels. 相似文献
