Trans-Pacific transport of enhanced ozone plumes has been mainly attributed to fossil fuel combustion in Asia in spring, but less attention has been paid to vegetation fires in Asia. Here we show that the El Niño-Southern Oscillation (ENSO)-modulated fires in Southeast Asia, rather than Asian fossil fuel plumes, dominate the interannual variability of springtime trans-Pacific transport of ozone across the entire North Pacific Ocean. During El Niño springs, the intensified fires from both the Indochinese Peninsula and Indonesia, together with large-scale circulation anomalies, result in enhanced ozone plumes that stretch over 15 000 km in both the lower-middle and upper troposphere. This enhancement is also observed in the in situ measurements of ozone concentration, with an almost 10% increase at Mauna Loa Observatory, Hawaii, a unique site to monitor the long-distance transport over the North Pacific. This study reports an unexpectedly strong influence of vegetation fires, linked with climate variability, on global tropospheric chemistry and proves once more how complex the interactions in the climate system are.  相似文献

To sequentially handle fluids is of great significance in quantitative biology, analytical chemistry, and bioassays. However, the technological options are limited when building such microfluidic sequential processing systems, and one of the encountered challenges is the need for reliable, efficient, and mass-production available microfluidic pumping methods. Herein, we present a bubble-free and pumping-control unified liquid handling method that is compatible with large-scale manufacture, termed multilayer microfluidic sample isolated pumping (mμSIP). The core part of the mμSIP is the selective permeable membrane that isolates the fluidic layer from the pneumatic layer. The air diffusion from the fluidic channel network into the degassing pneumatic channel network leads to fluidic channel pressure variation, which further results in consistent bubble-free liquid pumping into the channels and the dead-end chambers. We characterize the mμSIP by comparing the fluidic actuation processes with different parameters and a flow rate range of 0.013 μl/s to 0.097 μl/s is observed in the experiments. As the proof of concept, we demonstrate an automatic sequential fluid handling system aiming at digital assays and immunoassays, which further proves the unified pumping-control and suggests that the mμSIP is suitable for functional microfluidic assays with minimal operations. We believe that the mμSIP technology and demonstrated automatic sequential fluid handling system would enrich the microfluidic toolbox and benefit further inventions.  相似文献

The effect of acute hypoxic hypoxia on the profile of plasma amino acids in rats was studied and compared to that resulting from acute liver injury induced by giving carbon tetrachloride. In hypoxic rats exposed to 45% air in N2 for 5 h, the concentrations of branched chain amino acids, including valine, leucine and isoleucine, and aromatic amino acids such as phenylalanine and tyrosine were significantly increased as compared to those in normoxic rats. The ratio of branched-chain to aromatic amino acids (Fischer’s ratio) was significantly decreased. The levels of arginine and citrulline, which are related to the urea cycle, were also depressed. Furthermore, plasma proline level was reduced in hypoxic rats. The activities of plasma marker enzymes for tissue damage remained unchanged during hypoxia, indicating that tissue injury was not induced by exposure to hypoxic conditions. We suggest that the characteristic profile of plasma amino acids and the Fischer ratio are valuable tools for understanding the pathology of acute hypoxia in the absence of systemic tissue damage.  相似文献

The shakeout period after the emergence of a dominant design is very important to technology followers and niche market producers, who need adjust to significant changes in the competitive landscape. We posit that such innovating firms use strategic signals that address technical-, market-, and standards-uncertainty associated with their innovations to communicate with market participants to reduce participants' perceived uncertainties associated with the innovative products. Studying technology followers and niche market producers in the personal computer industry during its 1982–1989 shakeout period, we found that technology followers and niche market producers differ in their use of technical-, market-, and standards-related signals. Additionally, using event study methodology, we find that technology followers' strategic signals have significant positive impacts on both their own and other followers' stock prices. Meanwhile, niche market producers' strategic signals have significant positive impacts on their own stock prices, but not significant impacts on other niche market producers and technology followers' stock prices.  相似文献

Although much attention has been paid to investigating and controlling air pollution in China, the trends of air-pollutant concentrations on a national scale have remained unclear. Here, we quantitatively investigated the variation of air pollutants in China using long-term comprehensive data sets from 2013 to 2017, during which Chinese government made major efforts to reduce anthropogenic emission in polluted regions. Our results show a significant decreasing trend in the PM2.5 concentration in heavily polluted regions of eastern China, with an annual decrease of ∼7% compared with measurements in 2013. The measured decreased concentrations of SO2, NO2 and CO (a proxy for anthropogenic volatile organic compounds) could explain a large fraction of the decreased PM2.5 concentrations in different regions. As a consequence, the heavily polluted days decreased significantly in corresponding regions. Concentrations of organic aerosol, nitrate, sulfate, ammonium and chloride measured in urban Beijing revealed a remarkable reduction from 2013 to 2017, connecting the decreases in aerosol precursors with corresponding chemical components closely. However, surface-ozone concentrations showed increasing trends in most urban stations from 2013 to 2017, which indicates stronger photochemical pollution. The boundary-layer height in capital cities of eastern China showed no significant trends over the Beijing–Tianjin–Hebei, Yangtze River Delta and Pearl River Delta regions from 2013 to 2017, which confirmed the reduction in anthropogenic emissions. Our results demonstrated that the Chinese government was successful in the reduction of particulate matter in urban areas from 2013 to 2017, although the ozone concentration has increased significantly, suggesting a more complex mechanism of improving Chinese air quality in the future.  相似文献

Droplet-based microfluidic technologies are powerful tools for applications requiring high-throughput, for example, in biochemistry or material sciences. Several systems have been proposed for the high-throughput production of monodisperse emulsions by parallelizing multiple droplet makers. However, these systems have two main limitations: (1) they allow the use of only a single disperse phase; (2) they are based on multiple layer microfabrication techniques. We present here a pipette-and-play solution offering the possibility of manipulating simultaneously 10 different disperse phases on a single layer device. This system allows high-throughput emulsion production using aqueous flow rates of up to 26 ml/h (>110 000 drops/s) leading to emulsions with user-defined complex chemical composition. We demonstrate the multiplex capabilities of our system by measuring the kinetics of β-galactosidase in droplets using nine different concentrations of a fluorogenic substrate.  相似文献

In order to quantize Dirac's classical point electron1 we supplement Einstein's classical equation (E/c)2 ? p2 = b2 with a reciprocal classical equation (CΔt)2 ? (Δr)2 = a2 where b = mc and a is Dirac's signal radius. Δt is the time saved by a light signal in various states of motion of the electron, and a/c is the rest time saved. Our former efforts2 of obtaining an integral equation for the probability amplitude have been rectified by Born.3 There is no solution of the integral equation, however, unless advanced and retarded phases are introduced simultaneously, along with Dirac's advanced and retarded potentials. We have obtained a transcendental equation for the eigen-value μ = αγ where α is the Sommerfeld fine-structure constant, and γ is the numerical factor in Dirac's signal radius a = γe2/mc2. We investigated the searching behaviors of twenty-four children in grades 6, 7, and 8 (ages 11–13) in finding information on three types of search tasks in Google. Children conducted 72 search sessions and issued 150 queries. Children's phrase- and question-like queries combined were much more prevalent than keyword queries (70% vs. 30%, respectively). Fifty two percent of the queries were reformulations (33 sessions). We classified children's query reformulation types into five classes based on the taxonomy by Liu et al. (2010). We found that most query reformulations were by Substitution and Specialization, and that children hardly repeated queries. We categorized children's queries by task facets and examined the way they expressed these facets in their query formulations and reformulations. Oldest children tended to target the general topic of search tasks in their queries most frequently, whereas younger children expressed one of the two facets more often. We assessed children's achieved task outcomes using the search task outcomes measure we developed. Children were mostly more successful on the fact-finding and fully self-generated task and partially successful on the research-oriented task. Query type, reformulation type, achieved task outcomes, and expressing task facets varied by task type and grade level. There was no significant effect of query length in words or of the number of queries issued on search task outcomes. The study findings have implications for human intervention, digital literacy, search task literacy, as well as for system intervention to support children's query formulation and reformulation during interaction with Google.  相似文献

Halogen atoms affect the budget of ozone and the fate of pollutants such as hydrocarbons and mercury. Yet their sources and significances in polluted continental regions are poorly understood. Here we report the observation of unprecedented levels (averaging at 60 parts per trillion) of bromine chloride (BrCl) at a mid-latitude site in North China during winter. Widespread coal burning in rural households and a photo-assisted process were the primary source of BrCl and other bromine gases. BrCl contributed about 55% of both bromine and chlorine atoms. The halogen atoms increased the abundance of ‘conventional’ tropospheric oxidants (OH, HO2 and RO2) by 26%–73%, and enhanced oxidation of hydrocarbon by nearly a factor of two and the net ozone production by 55%. Our study reveals the significant role of reactive halogen in winter atmospheric chemistry and the deterioration of air quality in continental regions where uncontrolled coal combustion is prevalent.  相似文献

We developed a new method for releasing viable cells from affinity-based microfluidic devices. The lumen of a microchannel with a U-shape and user-designed microstructures was coated with supported lipid bilayers functionalized by epithelial cell adhesion molecule antibodies to capture circulating epithelial cells of influx solution. After the capturing process, air foam was introduced into channels for releasing target cells and then carrying them to a small area of membrane. The results show that when the air foam is driven at linear velocity of 4.2 mm/s for more than 20 min or at linear velocity of 8.4 mm/s for more than 10 min, the cell releasing efficiency approaches 100%. This flow-induced shear stress is much less than the physiological level (15 dyn/cm2), which is necessary to maintain the intactness of released cells. Combining the design of microstructures of the microfluidic system, the cell recovery on the membrane exceeds 90%. Importantly, we demonstrate that the cells released by air foam are viable and could be cultured in vitro. This novel method for releasing cells could power the microfluidic platform for isolating and identifying circulating tumor cells.  相似文献

The exponent and the constant must be estimated from the given set of author productivity data. A step-by-step outline is presented for testing the applicability of Lotka's law. While advances in genomics have enabled sensitive and highly parallel detection of nucleic acid targets, the isolation and extraction of the nucleic acids remain a critical bottleneck in the workflow. We present here a simple 3D printed microfluidic chip that allows for the vortex and centrifugation free extraction of nucleic acids. This novel microfluidic chip utilizes the presence of a water and oil interface to filter out the lysate contaminants. The pure nucleic acids, while bound on cellulose particles, are magnetically moved across the oil layer. We demonstrated efficient and rapid extraction of spiked Human Papillomavirus (HPV) 18 plasmids in specimen transport medium, in under 15 min. An overall extraction efficiency of 61% is observed across a range of HPV plasmid concentrations (5 × 101 to 5 × 106 copies/100 μl). The magnetic, interfacial, and viscous drag forces inside the microgeometries of the chip are modeled. We have also developed a kinetics model for the adsorption of nucleic acids on cellulose functionalized superparamagnetic beads. We also clarify here the role of carrier nucleic acids in the adsorption and isolation of nucleic acids. Based on the various mechanistic insights detailed here, customized microfluidic devices can be designed to meet the range of current and emerging point of care diagnostics needs.  相似文献

We present a novel 3D hybrid assembly of a polymer microfluidic chip with polycarbonate track-etched membrane (PCTEM) enabling membrane-supported cell culture. Two chip designs have been developed to establish either diffusive or convective reagent delivery using the integrated PCTEM. While it is well suited to a range of cell-based assays, we specifically employ this platform for the screening of a common antitumor chemotoxic agent (mitomycin C – MMC) on the HL60 myeloid leukemia cell line. The toxic activity of MMC is based on the generation of severe DNA damage in the cells. Using either mode of operation, the HL60 cells were cultured on-chip before, during, and after exposure to MMC at concentrations ranging from 0 to 50 μM. Cell viability was analysed off-chip by the trypan blue dye exclusion assay. The results of the on-chip viability assay were found to be consistent with those obtained off-chip and indicated ca. 40% cell survival at MMC concentration of 50 μM. The catalogue of capabilities of the here described cell assay platform comprises of (i) the culturing of cells either under shear-free conditions or under induced through-membrane flows, (ii) the tight time control of the reagent exposure, (iii) the straightforward assembly of devices, (iv) the flexibility on the choice of the membrane, and, prospectively, (v) the amenability for large-scale parallelization.  相似文献

This paper argues that the United Kingdom Ministry of Defence's (MoD) sponsorship of industrial semiconductor research and development (R&D) has, by its very nature, influenced the type of R&D performed, and the directions it has taken, to the extent of limiting its potential for subsequent commercial utilization. While much of the fault appears to lie with the semiconductor industry's response to MoD funding of its R&D, especially its over-reliance on such external support, the paper also suggests that the structure, modus operandi, and funding mechanisms of the MoD department concerned have significantly affected the industry's R&D activities. With these criticisms in mind, while at the same time noting the advantageous features of this department's activities, such as its relative autonomy, brief suggestions for ensuring more coherent and continuous state support for semiconductor R&D, including, for example, the use of innovation-oriented procurement policies, are offered.  相似文献

BackgroundAlthough bioactive metabolites capable of causing oxidative photo-necrosis in plant tissues have been identified in fungi, little is known about this type of mechanism in bacteria. These metabolites act as photosensitizers that generate reactive oxygen species (ROS) capable of causing damage to cells. In addition, these metabolites can pass into an energetically excited state when they receive some luminous stimulus, a condition in which they interact with other molecules present in the environment, such as molecular oxygen (O2), also known as triplet oxygen (3O2), generating ROS.ResultsThe suspension of the bacterial culture of Pseudomonas cedrina was shown to produce foliar necrosis in papaya leaves (Carica papaya L.) only in the presence of sunlight, which is evidence of photosensitizing mechanisms that generate singlet oxygen (1O2). From the chemical study of extracts obtained from this bacteria, 3-(4-(2-carboxipropyl) phenyl) but-2-enoic acid (1) was isolated. This compound, in the presence of light and triplet oxygen (3O2), was able to oxidize ergosterol to its peroxide, since it acted as a photosensitizer producing 1O2, with which it was corroborated that a photosensitization reaction occurs, mechanism by which this bacterium could prove to cause oxidative foliar photo-necrosis.ConclusionsP. cedrina was able to induce oxidative foliar photo-necrosis because of its potential ability to produce photosensitizing metabolites that generate singlet oxygen in the plants it colonizes. Organization of airway epithelium determines ciliary beat direction and coordination for proper mucociliary clearance. Fluidic shear stresses have the potential to influence ciliary organization. Here, an in vitro fluidic flow system was developed for inducing long-term airflow shear stresses on airway epithelium with a view to influencing epithelial organization. Our system consists of a fluidic device for cell culture, integrated into a humidified airflow circuit. The fluidic device has a modular design and is made from a combination of polystyrene and adhesive components incorporated into a 6-well filter membrane insert. We demonstrate the system operates within physiologically relevant shear and pressure ranges and estimate the shear stress exerted on the epithelial cell layer as a result of air flow using a computational model. For both the bronchial epithelial cell line BEAS2B and primary human tracheal airway epithelial cells, we demonstrate that cells remain viable within the device when exposed to airflow for 24 h and that normal differentiation and cilia formation occurs. Furthermore, we demonstrate the utility of our device for exploring the impact of exposing cells to airflow: our tool enables quantification of cytoskeletal organization, and is compatible with in situ bead assays to assess the orientation of cilia beating.  相似文献

The diffusion of innovations is identified as an important aspect of technological and social change. Innovations diffuse through segmented networks of knowledge that limit the flow of knowledge from any one technological domain to any other. Despite this segmentation, some organizations are capable of developing pieces of knowledge that overcome these limitations. Within this context, we develop four hypotheses regarding specific R&D strategies that affect a firm’s ability to develop inventions that diffuse beyond the firm’s technological boundaries. Specifically, we examine how a firm's scientific intensity, technological collaborations, technological diversity, and internal focus impact breadth of innovation diffusion. We use two of the main determinants of innovation diffusion, namely, the relative advantage and the observability, as theoretical mechanisms to build our arguments. We empirically test our hypotheses on longitudinal data from the industries of pharmaceuticals, biotechnology, and chemicals. Our findings show that the extent to which the knowledge embedded in a firm’s inventions diffuses in distant technological areas is positively related to the firm’s scientific intensity and to its extent of collaboration, but it is negatively related to its technological diversity.  相似文献
