The partition behaviors of β-1,3-1,4-glucanase, α-amylase and neutral proteases from clarified and whole fermentation broths of Bacillus subtilis ZJF-1A5 were investigated. An aqueous two-phase system (polyethylene glycol (PEG)/MgSO4)was examined with regard to the effects of PEG molecular weight (MW) and concentration, MgSO4 concentration, pH and NaCl concentration on enzyme partition and extraction. The MW and concentration of PEG were found to have significant effects on enzyme partition and extraction with low MW PEG showing the greatest benefit in the partition and extraction of β-glucanase with the PEG/MgSO4 system. MgSO4 concentration influenced the partition and extraction of β-glucanase significantly. pH had little effect on β-glucanase or proteases partition but affected α-amylase partition when pH was over 7.0. The addition of NaCl had little effect on the partition behavior of β-glucanase but had very significant effects on the partitioning of α-amylase and on the neutral proteases. The partition behaviors of β-glucanase, α-amylase and proteases in whole broth were also investigated and results were similar to those obtained with clarified fermentation broth. A two-step process for purifying β-glucanase was developed, which achieved β-glucanase recovery of 65.3% and specific activity of 14027 U/mg, 6.6 times improvement over the whole broth.  相似文献

The optimization of cultural conditions for β-glucanase production byBacillus subtilis ZJF-1A5 was investigated in flask trials. Temperature had great effect on β-glucanase production which maximized at optimal temperature of 37°C and decreased significantly when temperature was over 37°C. Charge quantity affected β-glucanase production significantly. Adding oxygen vector N-dodecane or acetic ether benefited β-glucanase production, but it depended on the concentration and charge quantity. The results of fractional factorial design showed that age and size of inoculum and shaking speed were the key factors affecting β-glucanase production and the cultivation time span to reach the highest β-glucanase activity. The optimal cultural conditions for β-glucanase production obtained with CCD were as follows: inoculum age and size (16 h, 3.82% (v/v)), shaking speed 210 r/min, charge quantity of 30 mL in 250 mL flask and initial pH 7.0, cultured at 37°C for 50 h. Repeated experimental results accorded with those predicted by a second-order polynomial model. The amount of β-glucanase, α-amylase and neutral protease produced byB subtilis ZJF-1A5 was associated partially with cell growth. Those three enzymes' activities increased following the cell growth and increased significantly when cells entered the stationary phase. β-glucanase was purified from a solid-state culture ofTrichoderma reesei on wheat bran in three steps which comprised ammonium sulfate precipitation, Sephadex G-100 chromatography, and DEAE-Sephadex A-50 chromatography. The molecular mass was determined to be 35.21 kilodaltons by sodium dodecyl sulfate-12.5% polyacrylamide gel electrophoresis. The β-glucanase at low pHs was more stable than that at high pHs, and optimum pH was 5.0. The optimum temperature was 60°C, and β-glucanase was relatively stable at below 40°C for 60 min. TheK m of the enzyme on β-glucan was 10.86 mg/ml, and theV max on β-glucan was 14286 μmol of glucose equivalents per mg of the pure enzyme per min. The β-glucanase activity was significantly inhibited by Fe3+ ions, and was reduced in the presence of Cu2+ ions, Mn2+ ions and Mg2+ ions at 5 mmol/L and 10 mmol/L, respectively. The β-glucanase activity was stimulated by Co2+ ions, Ca2+ ions, Zn2+ ions, and Fe2+ ions at 1 mmol/L and 5 mmol/L, respectively. In this paper, we consider some polynomial (α,β)-metrics, and discuss the sufficient and necessary conditions for a Finsler metric in the form F=α a1β a2β2/α a4β4/α3 to be projectively flat, where ai (i=1,2,4) are constants with a1≠0, α is a Riemannian metric and β is a 1-form. By analyzing the geodesic coefficients and the divisibility of certain polynomials, we obtain that there are only five projectively flat cases for metrics of this type. The effects of different timing of N fertilizer application at the same rate on grain β-amylase activity, protein concentration, weight and malt quality of barley were studied. Grain β-amylase activity and protein concentration were significantly higher in treatments where all top-dressed N fertilizer was applied at booting stage only or equally applied at two-leaf stage and booting stage than in the treatment where all top-dressed N fertilizer was applied at two-leaf age stage only. On the other hand, grain weight and malt extract decreased with increased N application at booting stage. There were obvious differences between barley varieties and experimental years in the grain and malt quality response to the timing of N fertilizer application. It was found that grain protein concentration was significantly and positively correlated with β-amylase activity, but significantly and negatively correlated with malt extract and Kolbach index. The effect of grain protein concentration on malt quality was predominant over the effect of grain β-amylase activity.  相似文献

The aim of this work was to construct a novel food-grade industrial arming yeast displaying β-1,3-1,4-glucanase and to evaluate the thermal stability of the glucanase for practical application. For this purpose, a bi-directional vector containing galactokinase (GAL1) and phosphoglycerate kinase 1 (PGK1) promoters in different orientations was constructed. The β-1,3-1,4-glucanase gene from Bacillus subtilis was fused to α-agglutinin and expressed under the control of the GAL1 promoter. α-galactosidase induced by the constitutive PGK1 promoter was used as a food-grade selection marker. The feasibility of the α-galactosidase marker was confirmed by the growth of transformants harboring the constructed vector on a medium containing melibiose as a sole carbon source, and by the clear halo around the transformants in Congo-red plates owing to the expression of β-1,3-1,4-glucanase. The analysis of β-1,3-1,4-glucanase activity in cell pellets and in the supernatant of the recombinant yeast strain revealed that β-1,3-1,4-glucanase was successfully displayed on the cell surface of the yeast. The displayed β-1,3-1,4-glucanase activity in the recombinant yeast cells increased immediately after the addition of galactose and reached 45.1 U/ml after 32-h induction. The thermal stability of β-1,3-1,4-glucanase displayed in the recombinant yeast cells was enhanced compared with the free enzyme. These results suggest that the constructed food-grade yeast has the potential to improve the brewing properties of beer.  相似文献

In this work, we study the Asanov Finsler metric F=α(β2/α2 gβ/α 1)1/2exp{(G/2)arctan[β/(hα) G/2]}, where α=(αijyiyj)1/2 is a Riemannian metric and β=biyj is a 1-form, g∈(-2,2), h=(1-g2/4)1/2, G=g/h. We give the necessary and sufficient condition for Asanov metric to be locally projectively flat, i.e., α is projectively flat and β is parallel with respect to α. Zhao (2003a) first established a congruence for any odd prime p>3, S(1,1,1;p)≡-2Bp-3 (mod p), which holds when p=3 evidently. In this paper, we consider finite triple harmonic sum S(α,β,γ;p) (mod p) is considered for all positive integers α,β,γ. We refer to w=α β γ as the weight of the sum, and show that if w is even, S(α,β,γ;p)≡0 (mod p) for p≥w 3; if w is odd, S(α,β,γ;p)≡rBp≥w (mod p) for p≥w, here r is an explicit rational number independent of p.
