We present a microfluidic device designed for maintenance and culture of non-adherent mammalian cells, which enables both recirculation and refreshing of medium, as well as easy harvesting of cells from the device. We demonstrate fabrication of a novel microfluidic device utilizing Braille perfusion for peristaltic fluid flow to enable switching between recirculation and refresh flow modes. Utilizing fluid flow simulations and the human promyelocytic leukemia cell line, HL-60, non-adherent cells, we demonstrate the utility of this RECIR-REFRESH device. With computer simulations, we profiled fluid flow and concentration gradients of autocrine factors and found that the geometry of the cell culture well plays a key role in cell entrapping and retaining autocrine and soluble factors. We subjected HL-60 cells, in the device, to a treatment regimen of 1.25% dimethylsulfoxide, every other day, to provoke differentiation and measured subsequent expression of CD11b on day 2 and day 4 and tumor necrosis factor-alpha (TNF-α) on day 4. Our findings display perfusion sensitive CD11b expression, but not TNF-α build-up, by day 4 of culture, with a 1:1 ratio of recirculation to refresh flow yielding the greatest increase in CD11b levels. RECIR-REFRESH facilitates programmable levels of cell differentiation in a HL-60 non-adherent cell population and can be expanded to other types of non-adherent cells such as hematopoietic stem cells. 相似文献

The present study evaluated the cardioprotective potential of vitamin-E by studying its effect on hemodynamic parameters,
lipid peroxidation, myocyte injury marker and ultrastructural changes in model of isoproterenol-induced myocardial necrosis
in rats. Wistar albino male rats (150–200 g) were randomly divided into saline, ISP control, and vit E groups. Vitamin E group
was administered vitamin E at a dose of 100mg/kg/day while saline and ISP control groups received saline orally for one month.
On 29th and 30th day, ISP (85 mg/kg, sc) was administered at an interval of 24 h to vit E and ISP control rats. On 31st day, rats of all groups were anesthetized and hemodynamic parameters were recorded. At the end of experimentation, animals
were sacrificed; hearts were excised and processed for biochemical and ultrastructural studies. ISP administration produced
marked cardiac necrosis as evidenced by significant decrease in my ocardial creatine kinase-MB as well as increase in malonaldialdehyde
levels. ISP-induced myocardial necrosis resulted in myocardial dysfunction as evidenced by significant depression in heart
rate and mean arterial pressure in the ISP control group as compared to saline control. Salient ultrastructural changes including
extensive loss of myofibrils, muscle necrosis, loss of mitochondria, and formation of several intracytoplasmic vacuoles and
lipid droplets further confirmed the ISP-induced myocardial damage. However, subsequent to ISP challenge, vit E treatment
significantly preserved the myocardium by restoring myocardial CK-MB activity, inhibiting the ISP-induced lipid peroxidation
and ultrastructural changes. Additionally, pre-and co-treatment of vit E prevented the deleterious ultrastructural changes
caused by ISP. These beneficial effects of chronic vit E treatment also translated into significant restoration of the altered
hemodynamic parameters. The present study clearly demonstrated the cardioprotective potential of vit E at dose of 100 mg/kg
in ISP-induced model of myocardial necrosis in rats. The significant restoration of altered hemodynamic parameters, myocardial
CK-MB activity, prevention of ISP-induced rise in lipid peroxidation and ultrastructural changes may confirm its cardioprotective

Mapping of curves from the physical to the hodograph plane, when the Jacobian vanishes along a curve is discussed in detail here. A certain quadratic expression occuring in the equations has been identified as the important factor in the transformation. The nature of the transformation depends on whether the roots are complex or real distinct and so on. The prevalence of non-communicable diseases like diabetes mellitus (DM) and hypertension (HTN) is growing worldwide. Both lead to nephropathy if not controlled effectively. Microalbuminuria (MAU) is recognized as an early predictor for nephropathy. Additionally, the timely detection of advanced glycation end products (AGEs) is also considered to be an important prognostic factor for diabetic nephropathies. Hence, screening for the early detection of MAU and AGEs would be an useful and relatively inexpensive laboratory test for early clinical diagnosis for the incidence of nephropathy in these diseases. This study was conducted in DM, HTN and pregnancy induced hypertensive (PIH) subjects. MAU and Nε-Carboxymethyllysine (CML) levels were estimated by in-house RIA kits in the patient groups and controls, while the total AGEs level in serum was determined by ELISA. The levels of MAU, CML and AGE-BSA were observed to be significantly higher in DM, HTN and PIH subjects compared to controls (p < 0.001). Increased serum CML and AGEs levels in DM, HTN and PIH subjects indicated ongoing glycemic damage and their susceptibility to develop renal complications. 相似文献

Based on questionaire responses from clients who have taken over indiginous technologies for commercialisation, an attempt has been made to identify and assess the relative magnitude and impact of the positive and negative factors influencing technology trnsfers. Parameters dealt with in this study relate to: (a) problems of time-lag; (b) the status, nature and extent of development of technologies; (c) sources of help in the commercial implementation of the technologies; (d) qualitative aspects of factors, such as plant design, operational difficulties, market demand and post-trnsfer assistance given by the transferring institutions; (e) capital investment needed for commercialisation of the technologies;p and (f) the general reaction of the clients to the technology transferred. The conclusions deal with minimising time-consuming procedural interlinks, selection of clients, avoidable managerial lapses in project selection, improving and augmenting the design and engineering capability of the transferring institutions, and uniform wxtension of post-transfer services to the clients. 相似文献

Exosomes, nanosized membrane-bound vesicles released by cells, play roles in cell signaling, immunology, virology, and oncology. Their study, however, has been hampered by difficulty in isolation and quantification due to their size and the complexity of biological samples. Conventional approaches to improved isolation require specialized equipment and extensive sample preparation time. Therefore, isolation and detection methods of exosomes will benefit biological and clinical studies. This paper examines some of the theoretical assumptions governing a model patent system and highlights the features of the new Indian Patent Act compared to these assumptions. An attempt has also been made to analyze and project data on foreign patent registrations in India both prior to and after the passing of the new Act. A tentative inference is that the new Act has not significantly affected the number of foreign patents seeking protection in India. A case has also been made out justifying the use of patent statistics in R & D programme formulation both at the macro- and micro-levels. 相似文献
