Oxygen and glucose supply is one of the important factors for the growth and viability of the cells in cultivation of tissues, e.g., spheroid, multilayered cells, and three-dimensional tissue construct. In this study, we used finite element methods to simulate the flow profile as well as oxygen and glucose supply to the multilayered cells in a microwell array chip for static and perfusion cultures. The simulation results indicated that oxygen supply is more crucial than glucose supply in both static and perfusion cultures, and that the oxygen supply through the wall of the perfusion culture chip is important in perfusion cultures. Glucose concentrations decline with time in static cultures, whereas they can be maintained at a constant level over time in perfusion cultures. The simulation of perfusion cultures indicated that the important parameters for glucose supply are the flow rate of the perfusion medium and the length of the cell culture chamber. In a perfusion culture chip made of oxygen-permeable materials, e.g., polydimethylsiloxane, oxygen is hardly supplied via the perfusion medium, but mainly supplied through the walls of the perfusion culture chip. The simulation of perfusion cultures indicated that the important parameters for oxygen supply are the thickness of the flow channel and the oxygen permeability of the walls of the channel, i.e., the type of material and the thickness of the wall.  相似文献

In rapid interceptive actions such as hitting a baseball, cricket ball or tennis ball, ball speed varies between trials, and players have to compensate the time lag by controlling the moment of movement onset and movement duration. Previous studies have found that these two variables can flexibly co-vary and are robustly influenced by target speed (i.e. velocity-coupling effect: faster movement for faster target). However, some studies reported an interpersonal variability in the timing control strategy and the relationship between the strategy and temporal accuracy in rapid interception is unclear. We used a baseball-simulated rapid interceptive task to assess this issue. Under relatively easy time constraints, there was a large interpersonal variability, and participants were distinctively divided into two groups: those who mainly modulated their movement duration and those who mainly controlled their movement onset. When the time constraint became severe, the second strategy shifted to the first strategy in most of the second group participants. In the both cases, being able to mainly control movement onset resulted in higher temporal accuracy. These results suggest that minimising the velocity-coupling effect is an important factor to achieve high temporal accuracy in rapid interception.  相似文献

Spheroid culture is a preferable cell culture approach for some cell types, including hepatocytes, as this type of culture often allows maintenance of organ-specific functions. In this study, we describe a spheroid microarray chip (SM chip) that allows stable immobilization of hepatocyte spheroids in microwells and that can be used to evaluate drug metabolism with high efficiency. The SM chip consists of 300-μm-diameter cylindrical wells with chemically modified bottom faces that form a 100-μm-diameter cell adhesion region surrounded by a nonadhesion region. Primary hepatocytes seeded onto this chip spontaneously formed spheroids of uniform diameter on the cell adhesion region in each microwell and these could be used for cytochrome P-450 fluorescence assays. A row of microwells could also be connected to a microchannel for simultaneous detection of different cytochrome P-450 enzyme activities on a single chip. The miniaturized features of this SM chip reduce the numbers of cells and the amounts of reagents required for assays. The detection of four cytochrome P-450 enzyme activities was demonstrated following induction by 3-methylcholantlene, with a sensitivity significantly higher than that in conventional monolayer culture. This microfabricated chip could therefore serve as a novel culture platform for various cell-based assays, including those used in drug screening, basic biological studies, and tissue engineering applications.  相似文献

This study evaluated baseball pitching accuracy using a variety of parameters to quantify pitching errors and analysed the validity of the accuracy measurements by comparing the outcomes of two small groups of pitchers. Several professional (n = 5) and high school (n = 8) pitchers threw 30 pitches each, including 20 fastballs and 10 breaking balls. To assess pitching accuracy, pitch locations relative to the catcher’s mitt (as the target) were evaluated with various parameters, including major/minor radius, an area of 95% confidence ellipse, absolute error, constant error and pitch location trajectory. Compared to the high school pitchers, the professional pitchers exhibited shorter major and minor radii in their 20 fastball pitches (p < 0.05), more accurate control in the lateral direction (p < 0.05), and shorter pitch location trajectories (p < 0.05). The evaluation methods presented in this study can objectively assess pitching accuracy and may thus provide useful coaching feedback with visual information.  相似文献

To extend face-negotiation theory (FNT) to romantic relationship contexts, we examined the patterns of responses to six survey instruments among five countries. We intend this initial paper to serve as a tutorial of testing measurement invariance—specifically by elaborating on: (a) how to establish measurement equivalence and (b) how to test measurement invariance using multigroup Confirmatory Factor Analysis. The results highlight the importance of establishing equivalency and routinely testing invariance in cross- and intercultural research. This is especially important given the often-extreme equivalency challenges faced when collecting data in non-Western cultures using scales developed in Western countries.  相似文献

According to recent motor control studies, it is important to know probabilistic structure of his/her own motor errors to choose an optimal motor plan (i.e., where you aim at) to maximise the expected gain. In this study, we questioned if pitching form determines the probabilistic structure of pitching errors in baseball pitchers. Eighteen collegiate baseball pitchers with various pitching forms including right- and left-handed overarm, sidearm and underarm throwers threw 100 pitches aiming at a target located 90 cm above the ground. Two dimensional distribution of pitch location was fitted by using bivariate normal distribution and 95% confidence ellipse was calculated. In order to quantify the pitching form, the direction of the throwing arm trajectory in frontal plane was calculated. The direction of the long axis was dependent on each participant’s pitching form (e.g., right overarm pitchers pitched along a right-up–left-down ellipse and left overarm pitchers pitched along a left-up–right-down ellipse). This was confirmed by circular correlation analysis (P = 0.98). These results suggest that different mechanisms, potentially errors in pitching mechanics and errors in ball release timing, might contribute to errors along the long axis and those along the short axis.  相似文献
