We present a microfluidic approach to characterizing temperature-dependent biomolecular interactions. Solvated L-arginine vasopressin (AVP) and its immobilized RNA aptamer (spiegelmer) were allowed to achieve equilibrium binding in a microchip at a series of selected temperatures. Unbound AVP were collected and analyzed with matrix-assisted laser desorption∕ionization mass spectrometry (MALDI-MS), yielding melting curves that reveal highly temperature-dependent zones in which affinity binding (36–45 °C) or dissociation (25–33 °C and 50–65 °C) occurs. Additionally, temperature-dependent binding isotherms were constructed; from these, thermodynamic quantities involved in binding were extracted. The results illustrated a strong change in heat capacity of interaction for this system, suggesting a considerable thermodynamic influence controlling vasopressin-spiegelmer interaction. 相似文献

Research into children’s ideas showed that children’s interpretations of natural phenomena often differ from those of scientists.
The aim of our study was to identify children’s ideas of various age classes (7/8–14/15) about birds. A questionnaire with
31 multiple choice and open ended questions and eight photographs were administered to 495 children from 10 elementary schools
in Slovakia. Children’s ideas were examined in six dimensions (bird classification, food, senses, communication, migration
and breeding including parental care). We found several misconceptions, some of them with both anthropomorphical and teleological
reasoning of the children about birds within each dimension. In general, misconceptions were more frequently found in younger
children, but several misconceptions were similarly distributed across all age classes. 相似文献

The integral part of the optimal velocity car-following models is the optimal velocity function （OVF）, which can be derived from measured velocity-spacing data. This paper discusses several characteristics of the OVF and presents regression analysis on two classical datasets, the Lincoln and Holland tunnels, with different possible OVFs. The numerical simulation of the formation of traffic congestion is conducted with three different heuristic OVFs, demonstrating that these functions give results similar to those of the famous Bando OVF （Bando et al., 1995）. Also an alternative method is present for determining the sensitivity and model parameters based on a single car driving to a fixed barrier. 相似文献

Chemical cytometry on a single cell level is of interest to various biological fields ranging from cancer to stem cell research. The impact chemical cytometry can exert in these fields depends on the dimensionality of the retrievable analytes content. To this point, the number of different analytes identifiable and additionally their subcellular localization is of interest. To address this, we present an electroporation based approach for selective lysis of only the plasma membrane, which permits analysis of the dissolved cytoplasm, while reducing contributions from the nucleus and membrane bound fractions of the cell analytes. The use of 100 μs long pulse and a well defined DC electric field gradient of ∼4.5 kV·cm−1 generated by 3D electrodes initiates release of a cytoplasm marker in ≪1 s, while retaining nuclear fluorescence markers. 相似文献

Using the critical work of Andreas Kazamias on the history and methods of comparative education as conceptual framework, we investigate the education (over a 200-year period) of the Slovak Roma. We position our story as paradigmatic of the dual processes of enlightenment and obscurantism with which we are familiar in thinking about the history of racial violence. The article describes the encounter of a young British doctor, Richard Bright, with Roma and Slovaks in 1814, and explores the Enlightenment thinking about Roma in specific, and race in general, in Bright’s travelogue. We juxtapose these historical investigations with qualitative findings from research conducted recently in the same region of Slovakia. Our goal is to show that Kazamias’s calls for a return to the historical in educational research, and greater attention to the “paideia of the soul,” have relevance in considering the discouraging past and present of Roma in Slovak schools and society. 相似文献
