葛渣异黄酮的提取及对细菌的抑制作用

摘    要:以葛根(radix puerariae)生产葛粉后的葛渣为材料,采用乙醇回流法通过正交实验对异黄酮提取条件进行优化,同时对葛根异黄酮对金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌的抑菌作用进行研究.结果表明:以乙醇作为提取溶剂,最佳提取条件为乙醇浓度95%,提取温度70℃,料液比1∶15,提取时间120 min,葛根异黄酮的得率为1.59%;葛根异黄酮对大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌的生长都具有抑制作用,最低抑菌浓度分别为125 μg/mL、62.5 μg/mL、250μg/mL,最低杀菌浓度分别为250 μg/mL、125 μg/mL、500 μg/mL.

Abstract:The ethanol refluxing method was used to extract isoflavones from the root dregs of kudzu (radix puerariae). The ex- traction conditions were studied by orthogonal design experiments. The antibacterial activity of isoflavones to escherichia coli, staphylococcus aureus, and bacillus subtilis was also studied. The experimental results show that: using ethanol as extraction sol- vent, the optimum extraction conditions is ethanol concentration 95%, extraction temperature 70 ℃, material-to-liquid ratio 1 : 15, extraction time 120 min. Under this condition, the yield of isoflavones extracting from root dregs of kudzu is 1.59%. Isofla- vones has inhibitory effect on the growth of escherichia coli, staphylococcus aureus and bacillus subtilis. The minimal inhibitory concentration (MIC) is 125 μg/mL, 62.5 μg/mL and 250 μg/mL separately; the minimum bactericidal concentration (MBC) is 250 μg/mL, 125 μg/mL and 500 Ixg/mL separately.
