耐力运动对高脂膳食雄性肥胖大鼠脂肪组织OR-RmRNA表达和性发育的影响

摘    要:目的:幼年时期营养过剩,不仅会引起肥胖,还会影响青春期的启动、发育和以后的生殖机能。我们以8周的耐力游泳为运动模型,探讨耐力运动对高脂膳食雄性大鼠脂肪组织OB-RmRNA的基因表达和性发育的影响,诣在为揭示运动减肥机制以及与性发育的关系提供实验依据。方法:Wistar雄性大鼠40只,随机分三组:普通膳食组(C)、高脂膳食不运动对照组(NE)、高脂膳食运动组(E),NE和E组进行为期8周的高脂膳食和游泳训练。采用测量法测定其脂肪垫和体重,推测Lees指数,放免法检测血清瘦素、睾酮水平,分子生物学的方法测定脂肪中OB-RmRNA基因表达。结果:1)与C组相比:8周的高脂饮食使NE组体重、Lees指数、脂肪垫、血瘦素显著升高(P<0.001;P<0.01;P<0.01;P<0.01);血睾酮水平显著下降,P<0.05;NE组RNA相对含量下降59.2%;E组下降36.2%。2)与NE组相比:8周的游泳训练使E组体重、Lees指数、脂肪垫、血瘦素水平显著下降(P<0.001;P<0.01;P<0.01;P<0.05),血睾酮水平显著升高P<0.05;以NE组为对照:E组RNA相对含量上升56.2%。3)C组与E组各指标之间没有显著性差异。结论:1)高脂膳食雄性肥胖大鼠体内出现瘦素抵抗,其机制是由于瘦素受体的基因表达下调,导致其对瘦素的敏感性下降,并影响到其性发育。2)耐力训练可有效的降低体脂,上调瘦素受体的基因表达,改善瘦素抵抗,有效的缓解了肥胖引起的性发育不良。

Abstract:Objective: Overnutrition in childhood will not only cause fatness but do harm to the initial and development of adolescence and the following reproducing mechanism.A model of 8 weeks endurance swimming is made to discuss the Influence of endurance exercise on fat tissue OR-RmRNA expression and sexual development of fat male rats in high fat diet in order to provide experimental evidence for revealing the relationship between sport weight-reducing mechanism and sexual development.Methods: 40 Wistar male rats are divided into three groups randomly: common diet group(C),high fat diet and non-exercise control group(NE),and high fat diet and exercise group(E).Group E and NE receive 8 weeks high fat diet and swimming training.Their fat layer and body weight are measured to imply their Lees index,the level of serum leptin and testosteroneare tested by immunoradiometric method,and the gene expression of OR-RmRNA in fat are tested by molecular biological method.Results: 1) Compared to Group C,8 weeks high fat diet makes the body weight,Lees index fat layer and blood leptin of Group NE rise significantly(P <0.001;P<0.01;P<0.01;P<0.01);and the level of testosteroneare decrease(P<0.05);the comparative content of RNA of Group NE decreases by 59.2%;Group E decreases by 36.2%.2) Compared to Group NE,8 weeks swimming training makes the body weight,Lees index fat layer and blood leptin of Group E rise significantly(P<0.001;P<0.01;P<0.01;P<0.01);and the level of testosteroneare increases(P<0.05);compared to Group NE,the comparative content of RNA of Group E increases by 56.2%.3) Indexes of Group E and C have no remarkable difference.Conclusions: 1) Leptin resistant appears in high fat diet male rats,whose mechanism is lower down gene expressions of leptin receivers causing the sensitivity to leptin decrease and affecting their sexual development;2) endurance exercise reduces the body fat effectively,raise the gene expression of leptin receiver,improve the leptin resistant and relief the impaired sexual development caused by fatness.
