皖西大别山松乳菇菌丝半固体培养研究

摘    要:采用含琼脂的丰固体培养基来培养松乳菇菌丝,优化了半固体培养基碳源和氮源以及培养温度,并将含同样营养成分的液体培养、固体培养和半固体培养进行了比较,试验结果表明,松乳菇菌丝采用半固体培养时,0.5%蔗糖与1.5%淀粉的组合碳源培养菌丝生长速度最快,蛋白胨是最理想的氮源物质,25℃是松乳菇菌丝的最适培养温度.3种不同培养基培养结果是半固体培养基菌丝生长速度明显超过了其他2种培养基,且菌丝颜色可以由纯白色转变为棕黄色,同时出现部分菌丝相互靠近集中生长的现象,此现象类似子实体原基的形成.松乳菇菌丝半固体培养方法为人工栽培松乳菇提供了新的思路.

Culture for the Hyphae of Lactarius Deliciousin in Wanxi Dabie Mountains by Semisolid Medium

Abstract:The hyphae of lactarius deliciousin were cultured by semisolid medium contained 0. 9% Agar, and carbon source, nitrogen source and culture temperature were optimized in the hyphae culture tests. The contrasting tests of liquid culture, solid culture and semisolid culture were conducted by the same nutritional components. The results showed the mixture of 0.5% sucrose and 1.5% starch was the best carbon source, and peptone was the optimal nitrogen source, and 25℃ was the perfect culture temperature. The growth rate of lactarius deliciousin hyphae was maximal by semisolid culture. Also, the color of the hyphae could turn into brown from white, and the hyphae could close to each other like primordium of fruit body during the semisolid culture tests. The method of the semisolid culture would contribute to the artificial cultivation of lactarius deliciousin.
