大肠杆菌抗药性的体外诱导与消除

摘    要:采用试管二倍稀释法 ,分别用林可霉素、氯霉素、青霉素对大肠杆菌进行抑菌实验 ,并测定其MIC(最低抑菌浓度 ) ;用平板抑菌圈、管碟稀释法检测大肠杆菌对单味中药三颗针、遥竹消的敏感性 ;在含 1/2MIC抗生素的药物培养基中连续传代 2 0次 ,然后 ,菌液再分别在 1/2MIC浓度的三颗针、遥竹消中连续传代 5次 ,测定抗生素的MIC 比较了传代前后大肠杆菌对抗生素耐受性的变化 结果表明在含抗生素的培养基中传代 2 0次后 ,大肠杆菌的抗生素MIC大幅度提高 ,但青霉素提高幅度不大 菌液再经过中药处理后抗生素的MIC都降低了 ,但抗药性并未完全消除

Abstract:The antimicrobial experiment of antibiotics containing Lincomycin hydr ochloride,Chloramphenicol and Penicillin was taken by using two times echelon d iluting method,and the minimal inhibitory concentration (MIC) of E.coli was det ermined;Sensitivity of E.coli to 2 single Chinese medicines called San-ke-zhen and Yao-zhu-xiao was investigated with plate method and tube-plate method;We in cubated E.coli cells under the culture media with 1/2 MIC antibiotics (Lincomyci n hydrochloride,Chloramphenicol or Penicillium) respectively for 20 times. Late ly,the culturing process continued under the media with Chinese medicine(San-ke -zhen or Yao-zhu-xiao)for 5 times .We tested the MIC of antibiotics (Lincomycin hydrochloride,Chloramphenicol and Penicillium). Comparing E.coli’ s resistance to Lincomycin hydrochloride,Chloramphenicol and Penicillin before and after in cubating in LB containing antibiotics(Lincomycin hydrochloride,Chloramphenicol or Penicillin),we tried to understand the difference in their mechanism of resi stance and how Chinese medicine eliminate bacteria’s resistance.. After 20 time s serial passage with 1/2 MIC of antibiotics (Lincomycin hydrochloride,Chloramp henicol or Penicillin),the MIC of antibiotics rose immensely except Penicillium . Continuing the function of 1/2 MIC Chinese medicine,the MIC of antibiotics de creased,but the resistance didn’t eliminate completely.
