实验性脂肪肝大鼠的脂肪代谢、细胞因子、瘦素方面的变化

摘    要:目的:观察脂肪肝大鼠在生化、病理、脂肪代谢、细胞因子、胰岛素及瘦素各方面指标的变化.方法:雄性Wistar大鼠22只,按体重层次随机分为2组,对照组(A)11只,脂肪肝组(B)11只.对照组饲以普通鼠饲料,脂肪肝组饲以高脂饲料.8周后,全部处死,取血及肝组织,测定血中AIT、AST、TG、CHO、INS、FBS、FFA、Ieptin、II-6、TNF.肝组织匀浆测定肝组织TC,TG,SOD、MDA,观察肝脏组织学变化.结果:脂肪肝大鼠存在明显脂肪代谢紊乱,肝功能异常,胰岛素抵抗及瘦素表达增高;细胞因子升高.结论:实验性高脂饮食脂肪肝大鼠存在瘦素抵抗及胰岛素抵抗.

Abstract:Objective : To observe the change of the liver function, pathology, fat metabolism, insulin,leptin and cytokine on fat liver rats. Methods :22 male Wistar rats were randomly devided into two groups, control group 11 rats, fat liver group 11 rats. The control group were fed by normal fodder,the other group by a high-protein fodder. After eight weeks all the animals were killed. Liver index( liver/body weight ratio), serum activities of liver-associated enzymes, blood lipids, FFA, leptin, fasting blood glucose, fast plasma insulin, HOMA insulin resistance index ( HOMA -IR ), IL-6 , TNF, liver triglycerides, liver cholesterol , liver tissue MDA,SOD and the liver histology of rats of all groups were assayed. Results: The body weight, liver index, serum activities of alanine aminotransferase ( ALT ) , aspartate aminotransferase (AST), total cholesterol (TC), triglycerides (TG) and liver triglycerides, liver cholesterol , liver tissue MDA, HOMA-IR, FFA, leptin, IL-6, TNF in the fatty liver group increased significantly, while liver tissue SOD concentration decreased significantly. Fast blood glucose, fast plasma insulin showed an increasing tendency, but there was no significant difference among the two groups. The liver histology in the the fatty liver group showed moderate to severe steatosis, mainly as macro vesicle steatosis, lobular inflammatory, cell infiltration and necrosis. Conclusion: It is suggested that there is leptin resistence and insulin resistence in fatty liver rats induced by high fat feeding.
