乙酰胆碱对海马神经元网络自发同步钙振荡的调控作用

摘    要:目的以体外原代培养海马神经元网络为模型,应用钙成像技术,观察不同浓度乙酰胆碱(ACh)对海马神经元网络同步自发钙振荡的调控作用。方法取(18±1)d的SD胎鼠,分离海马组织,制成细胞悬液,进行细胞培养。培养9—13d的海马神经元用于实验。分别加入不同浓度的ACh,应用钙成像技术,观察其对钙振荡频率的影响。结果加入1umol/LACh后,钙振荡频率增加至对照期的(116.10±2.63)%(x±s,n=16);加入10umoL/L,100umol/LACh后,钙震荡频率增加到对照期的(138.39±4.03)%(n=27)和(141.37±2.33)%(n=21);而加入500umol/LACh钙震荡频率在前4min可增加到对照期的(156.81±1.65)%(n=18),但在后6min时段内海马神经元网络同步自发钙震荡频率减慢,到16—18min,钙震荡频率减慢至对照期的(52.27±0.55)%(n=18)。结论ACh在一定浓度范围内可以加快海马神经元网络同步自发钙振荡频率,调节神经元网络兴奋性。

Abstract:Objective The primarily cultured rat hippocampus neuronal networks were used for the model in this study. In order to investigate the effect of ACH,calcium imaging technique were used to observe the change of spontaneous synchronized calcium oscillation frequency in hippocampal neuronal networks. The aim is to learn how ACh acts on CNS on the cellular level,which would enrich the theory system of ACh in its clinical application. Methods Hippocampal tissues from( 18 ± 1 ) day fetal SD rats were dissected using trypsin. And then it was prepared into single cell suspension, which was then planted in dishes in appropriate concentration. The cultures were used for experiments on 9 - 13 days after planting. The change in calciium oscillation frequency was recorded before and after ACh administration by calcium imaging technique. Results After the administration of 1 umol/L ACh the frequencies of calcium oscillation were increased to 116.10 ± 2.63% ( n = 16) in the first 4 minutes whereas there were no change in the following 6 minutes. After the administration of 10umol/L and 100umol/L ACh the frequencies of calcium oscillation increased to( 138.39 ±4.03 ) % ( n = 27 ) and ( 141.37 ± 2.33 ) % ( n = 21 ) respectively compared with the control period. After the administration of 500umol/L ACh the frequencies increased to ( 156.81 ± 1.65 ) % ( n = 18 ) in the period of first four minutes whereas the frequencies decreased to (52.27 ± 0.55 ) % ( n = 18 ) in the last two minutes compared with the control period. Conclusion ACh can increase the frequencies of spontaneous synchronized calcium oscillation in hippocampal neuronal networks in appropriate concentration.
