酶辅助法提取大青叶中总黄酮及抗氧化性研究

摘    要:本论文采用酶辅助法提取大青叶中的总黄酮,先用纤维素酶酶解,再用乙醇回流提取大青叶中总黄酮。分别固定乙醇提取液的浓度为70%,料液比为1∶20g∕mL,回流时间为2h。初步探究了纤维素酶浓度、酶解pH、酶解温度、酶解时间四个单因素对大青叶中黄酮提取率的影响。设计正交实验确定酶辅助法提取大青叶中总黄酮的较佳条件为:纤维素酶浓度为4U/mL、酶解pH=4.5、酶解温度为40℃、酶解时间1.5h,总黄酮提取率高达2.85%。另外还研究了大青叶总黄酮提取液对羟自由基的清除活性,与BHT的抗氧化剂活性作对照实验,结果表明大青叶中的总黄酮对羟自由基有较强的清除活性。

Abstract:Enzyme - assisted extraction of total flavonoids from folium Isatidis was studied in this pa-per. Frist enzymatic hydrolysis by cellulase, and then reflux by ethanol to extract total flavonoids. Ethanolconcentration of 70% , solid -liquid ratio of I :20g/mL and reflux time of 2h were fixed. The preferred ex-traction conditions were determined by single factor experiment and orthogonal experiment. The experimentreveals that : cellulase concentration of 4U/mL, enzymatic hydrolysis pH of 4.5, enzymatic hydrolysis tem-perature of 40℃, and enzymatic hydrolysis time of 1.5 h. The extraction ratio of total flavonoids reached upto 2.85 % by using spectrophotometry. Antioxidant effects of flavonoids and BHT were measured to removehydroxyl radicals by comparative experiments. Antioxidant experiments showed that flavonoids from foliumIsatidis showed a strong scavenging effect on hydroxyl radicals.
