大鼠哮喘模型中嗜酸性粒细胞上前列腺素D2受体改变研究

摘    要:目的:研究大鼠哮喘模型中外周血嗜酸性粒细胞(EOS)上前列腺素D2(PGD2)受体改变,了解其对EOS的调控作用。方法:动物模型复制:20只雄性SD清洁级大鼠,随机分2组(A组:卵白蛋白(OVA)激发组,B组:应用生理盐水雾化吸入组,为正常对照组),雾化诱喘。测定外周血和肺泡灌洗液(BALF)的EOS,并用放射配体分析其外周血EOS上的PGD2受体。结果:B组外周血和BALF的EOS计数较正常组显著增加,(外周血0.48±0.01和0.12±0.03(×10^9/L),BALF:1.21±0.47和0.39±0.24(×10^7/L));外周血EOS上CRTH2受体结合容量和总结合比正常对照组和增加(fmol/10^6细胞)(CRTH 2分别为:313.58±86.17和121.32±69.88;DP总结合分别为449.19±121.49和298.85±79.64)(P〈0.01),DP1无显著性差异(A组180.72±47.44和B组198.68±87.22)(P〉0.05)。结论:在大鼠哮喘模型中外周血和BALF中EOS绝对值增高,其表面DP总结合显著增加,主要与CRTH2受体(DP2)显著增加有关,DP1受体结合容量无显著改变,CRTH2结合容量增加可能与哮喘发病机制有关。

Abstract:Object:To study the change of prostaglandin D2 receptor on eosinophil in rats asthma models.Methods:Twenty male SD rats were randomly divided into two groups: Group A: ovalbumin(OVA) challenge group,Group B: the control group.The rats in each group were sacrificed 24 h after the last challenge.In the experiment,the rat model of asthma was established by ovalbumin(OVA) challenge method,bronchoalveolar lavage fluid(BALF) was collected from the right lung.The total cell numbers,eosinophils(EOS) absolute count and differentiated cell counts in BALF were performed on different count fluids.DP were measured by using Radiological binding assay(RBA).Results: Number of EOS(×10^9/L)in peri-pheral blood in B groups(0.48±0.01)increased markedly compared with normal control group(0.12±0.03)(P〈0.01),in BALF,Number of EOS(×10^7/L)(1.21±0.47)in B groups were markedly increased than A group(0.39±0.24(×10^7/L))(P〈0.01);The combining capacity of CRTH2 and total combining capacity on Eos in A group were significantly higher than normal controls(fmol/10^6cell)(CRTH 2: 313.58±86.17 vs 121.32±69.88,DP total combining capacity: 449.19±121.49 vs 298.85±79.64)(P〈0.01).In twe groups,there are no significantly differences in DP1(A group 180.72±47.44 vs B group 198.68±87.22)(P〉0.05).Conclusion: In rats asthma models,there were a significantly higher in eosinophil counts in peri-pheral blood and BALF,combining capacity of CRTh2 and total combining capacity on Eos in peri-pheral blood were significantly higher after OVA challenge.CRTH2 plays an important role in pathogenesis of asthma.
