油菜小孢子衍生植株倍性水平及结实性分析

摘    要:为了研究不同油菜基因型小孢子衍生植株倍性水平差异以及秋水仙碱处理对相应植株结实性能影响,本研究利用三个遗传背景各不相同的F1杂交组合,通过小孢子培养产生再生小苗,经FCM检测比较不同基因型染色体的倍性水平;并根据检测结果,选择性地对再生苗进行秋水仙碱加倍处理,分析不同染色体倍性间加倍率和结实性差异.结果表明:倍性水平显示二倍体的再生苗(自然加倍)约占总数的20%~28%,其中60%~80%可发育成正常二倍体植株,故不需要再行加倍处理;倍性为嵌合体的再生苗占45%~60%,未加倍处理平均仅7%植株可长成结荚正常的二倍体,经加倍处理后结实株率平均可从40%提高到75%;根据植株的结实性分析,对相同基因型的角果性状来说,自然加倍产生的正常植株优于加倍处理后产生的二倍体,但秋水仙碱加倍处理明显促进嵌合体植株的角果发育,平均角果长度增加1.2 cm,角果粒数增多4粒左右.

Abstract:This study was to investigate the genotypic difference in ploidy levels of microspore-derived plants and the effect of colchicine treatment on seed setting of the regenerated plants in oilseed rape.After measuring the ploidy levels of young plantlets by Flow-cytometry(FCM),the diploid/haploid mixed and haploid plantlets were treated with 0.5 mg.mL-1 colchicine,then chromosome doubling efficiency and seed setting characters were investigated.The results of FCM detection showed that 20-28% of regenerated plants were spontaneous doubling haploids,in which,60-80% was developed to be normal diploid plants.The majority of regenerated plantlets showed ploidy levels with the diploid/haploid mixed(45%-60%) and only 7% of them could harvest seeds from full fertile plants,but after artificially doubling treatment,the total number of productive plants was significantly increased from in average of 40% to 75%,however,most of them were partial diploids(diploid/haploid mixed).Our results demonstrated that the spontaneous doubled haploids clearly showed more pods per plant and longer pod length with more seeds number than that of diploid plants induced by colchicine treatment,while the colchicine treatment significantly promoted their pod formation and development,with an average of 1.2cm increase in pod length and 4 seeds more per pod.
