银屑病mRNA原位杂交技术的研究

摘    要:通过对38例银屑病(Psoriasis)皮肤组织中HB—EGF的表达,比较了核酸原位杂交过程中包括组织固定、脱水、浸蜡、包埋及切片过程中,外源性RNase控制与原位杂交结果之间的关系。结果表明,外源性RNase控制组的阳性信号高于非控制组,其原位杂交结果反映了基因表达的水平。提示在石蜡切片核酸原位杂交实验中,切片制备及保存过程中防止外源性RNA酶污染,是提高结果可信度的关键。

Abstract:Through 38 examples psoriasis in the skin organizations the HB - EGF expression , the dehydration compared with the nucleic acid home position hybrid process including the organization, soaks in the wax, the embedding and the slice process, extraneous source nature RNase control and between home position hybrid result relations. The result indicates that, the extraneous source nature RNase control group's masculine signal is higher than the non-control group and its home position hybrid result reflects gene expression level. The prompt in the paraffin section nucleic acid home position hybrid experiment, in the slice preparation and the preserved process prevented the extraneous source nature RNA enzyme pollution, is the key to enhance the result reliable level.
