| 摘 要： | 以常熟白芹的带节茎段和顶芽为外植体进行离体培养，对外植体的灭菌方法、培养基中不同种类生长调节物质的浓度对增殖和生根的影响以及试管苗的保存方法等进行了研究．结果表明：以0．1％的HgCl2为灭菌剂，对水芹顶芽处理8min可以获得无菌苗30％；丛生试管苗切割成单株后接种在MS＋6-BA0．5～1．5mg·L。的增殖培养基上培养28d，平均增殖系数可达10．5以上；试管苗在1／2MS培养基上培养14d后，生根率达100％，21d时平均每株生根16．5条；试管苗用1mg·L。的NAA浸泡切口处24h，再用清水培养10d后，生根率达100％，平均每株生根22．8条；炼苗后移植成活率100％．在温度为15℃、光照时间12h·d^-1、光照强度20μmol·m^-2·s^-1的培养条件下，试管苗在含6-BA0．5mg·L^-1的增殖培养基上可保存6个月；MS培养基中Ca^2＋离子浓度加倍，可延长试管苗保存时间1-1．5个月．

| Abstract: | The in vitro culture of Changshu Oenanthe javanica was established by using the nodal stem segment and terminal bud as explants.The suitable method for surface sterilization of explants,the effects of different plant growth regulators on the multiplication and rooting,and the appropriate measure for preserving test-tube plantlets were investigated.The results showed that 30%sterile explants could be obtained when the terminal buds were treated by surface sterilization with 0.1%HgCl_2 for 8 min.When the sing... |
