膜蛋白FKBP38病理生理功能研究进展

摘    要:FKBP38亦称FKBP8,在诸多方面均体现其为FK506结合蛋白家族的一个特殊成员,仅FKBP38具有对钙调蛋白的调节活性。FKBP38活化还可通过抑制抗凋亡Bcl-2参与凋亡信号传导。研究表明,除在程序性细胞死亡中的作用外,FKBP38还发挥对诸多差异显著的细胞过程的调控作用,而这些过程不完全依赖其FKBP结构域。FKBP38的生物学功能包括调节蛋白激酶mTOR、介导线粒体自噬、调节细胞缺氧反应以及丙型肝炎病毒复制等。因此,FKBP38的药理学靶向作用研究极可能是一种有效干预对FKBP38不同依赖性过程(如癌症或神经退行性疾病的程序性细胞死亡)的策略。

Abstract:FKBP38,also known as FKBP8,which is a special member of the FK506 binding protein family in many aspects.Only FKBP38 has calmodulin-regulated activity in this protein family.The activation of FKBP38 also participates in apoptotic signal transduction by inhibiting the anti-apoptotic Bcl-2.It has been shown that besides the above-mentioned programmed cell death,FKBP38 also plays a regulatory role in many distinct cell processes,which do not depend entirely on the FKBP domain.The biological functions of FKBP38 include regulation of the kinase m TOR,regulation of mitophagy,regulation of cellular hypoxia response and hepatitis C virus replication.Therefore,the pharmacological targeting effect of FKBP38 may be an effective strategy to intervene in different dependent processes of FKBP38,such as programmed cell death in cancer or neurodegenerative diseases.
