游泳及丙酮酸钙补充对大鼠瘦素和瘦素受体水平的影响

摘    要:研究目的:以游泳大鼠为研究对象,通过检测其补充丙酮酸过程中身体成份、瘦素和瘦素受体基因表达的变化,探讨丙酮酸补充对游泳大鼠脂肪代谢的影响及机理.研究方法:选取38只健康成年雄性Wistar大鼠,随机分为安慰剂对照组、运动对照组、补充丙酮酸对照组和补充丙酮酸运动组.采用竞争放射免疫分析法测定大鼠血清瘦素和脑脊液瘦素水平;PCR法测定下丘脑瘦素受体基因表达量.研究结果:4周丙酮酸补充可明显减缓游泳大鼠体重和体脂增长速度,明显升高大鼠和游泳大鼠脑脊液瘦素水平及下丘脑瘦素受体mRNA表达水平.研究结论:丙酮酸补充可在一定程度上减慢游泳大鼠体重和体脂的增长速度,加快其能量代谢速度;并可能通过对游泳大鼠体内瘦素水平及瘦素受体基因表达量的调控,来影响游泳大鼠的脂肪代谢过程.

Abstract:Male swimming rat s as experimental object s were selected and t he effect s and mechanism of
pyruvate supplement on lipometabolism should be discussed in an at tempt to measure some changes
regarding body composition and leptin and leptin receptor . 38 male Wistar rat s were selected and ran2
domly divided into four group s , placebo cont rol group , t raining cont rol group , supplement cont rol
group , t raining and supplement group . The serum and cerebrospinal fluid leptin were determined by
Radioimmunoassay Met hod , leptin recptor (OB2R) mRNA in hypot halamus by PCR met hod. It was
seen t hat four weeks’supplement of pyruvate could weaken weight and body fat gaining remarkably in
swimming rat s and increase cerebrospinal fluid leptin and leptin receptor level in hypot halamus in rat s
and swimming rat s dramatically. The supplement of pyruvate could weaken weight and body fat gai2
ning remarkably , which could accelerate t he speed of t he body energy metabolism. It indicated t hat
pyruvate supplement could regulate lipometabolism in swimming rat s by means of the effect on leptin
and leptin receptor level s.
