导入粟DNA的小麦变异系酯酶同工酶分析

摘    要:彩和PAGE方法,对从导入珍珠粟DNA的两个春小麦受体(佛约28和青春4425)中所选的17个变异系进行了酯酶同工酶分析,结果表明,变异系与受体的酯酶同工酶图谱基本相同;同时,在变异系中出现了供体酶带和新酶带,而且在酶活性和酶带数等方面也表现出不同程度的变异。说明酯酶同工酶的变异与粟DNA导入有密切关系,变异系是外源DNA导入的结果,进上步说明花粉管通道法导入外源DNA技术是改良小麦品种、丰富小麦

Abstract:Millet DAN was introduced into two spring wheat varieties(Huyue 28 and Qingchun 4425)by the pollen tube pathway method.Seventeen variants from their descendents,the two teceptors and the donor were used as the test materials for esterase isoenzyme analysis.It was found that the variants were very similar to the receptors in esterase electrophoresis(PAGE)bands.Several kinds of variations were obviously shown.It was confirmed that the 17 variants are all the transformed strains of millet DNA.It was indicated that the pollen tube pathway technique for exogenous DNA transformation is an effective way to enrich wheat gene pool and improve wheat.
