摘    要:目的:研究乳酸对家兔离体心室乳头肌收缩力和细胞动作电位的影响。方法:应用细胞内微电极技术和微拉力描记法,研究不同浓度乳酸对家兔离体心室乳头肌收缩力(n=10)、心肌细胞动作电位(n=6)的影响。结果:一定浓度的乳酸对心肌有负性肌力作用,心肌收缩力的降低随乳酸浓度的升高而更趋明显。4mmol/L乳酸溶液灌注后,心肌细胞跨膜电位的变化为Vmax显著降低(P<0.005)。结论:乳酸溶液达4mmol/L时导致0期去极化速度的降低,即Na+通道效率降低。乳酸浓度升高引起心肌收缩力下降,其可能机制为Na+内流减少,Na+-H+交换减少,造成细胞内H+浓度升高;经Na+-Ca2+交换使Ca2+外流减少,导致细胞内Ca2+超载。

Abstract:To investigate the effect of lactate solution of different concentration on action potential of ventricular papillary muscle cell and contractile force.Through using the intracellular microelectrode technique and micro-tension depicting method,this paper studied the effect of lactate solution of different concentration on action potential of ventricular papillary muscle cell(n=6) and contractile force(n=10).The result showed that effect of lactate solution of certain concentration was obvious after perfuse with lactate solution of 4 mmol/L.The Vmax decreased significantly(P<0.05).It concluded that 4 mmol/L of lactate concentration could lead to decrease of depolarication.The efficiency of Na~+ channel decreased.The elevated lactate concentration leads to decrease of contractile force if cardiac muscle.The possible mechanism was Na~+ influx and Na~+-H~+ exchange was inhibited.It resulted the elevation of intracellular H~+ concentration.Through the Na~+-H~+ exchange,Ca~ 2+ efflux decreased and intracellular Ca~ 2+ overload.
