| 摘 要： | 研究目的：研究高脂饮食对大鼠血浆和下丘脑各项内分泌的影响,初步探讨肥胖大鼠形成机制。方法：高脂饮食喂养SD大鼠10周,测试比较肥胖大鼠、肥胖抵抗大鼠和正常饮食体重大鼠基础形态和血液学指标以及血浆和下丘脑摄食相关调节肽指标。结果：肥胖大鼠血浆Ghrelin和Ghrelin/Obestatin比值有下降趋势,NPY显著升高;下丘脑Ghrelin和Obestatin显著下降,Ghrelin/Obestatin比值显著下降。GHSR-1a基因表达显著下降,而NPY基因表达没有明显变化,GPR-39mRNA在下丘脑没有表达。结论：下丘脑Ghrelin/Obestatin比值在大鼠肥胖发生中可能有一定的作用;Ghrelin可能通过中枢机制影响摄食和能量代谢调节;Obestatin-GPR39通路在能量代谢调节中可能不起作用。

| Abstract: | The purpose of this study was to analyze the influence of high lipid diet on serum and hypothalamic endocrinological hormones of rats and study the mechanism of obese rats.Through 10 weeks high lipid diet,this paper analyzes some physique and serum index,serum and hypothalamic endocrinological hormones.The result shows that obese rats have distinct differences on body weight,BMI and serum preferences.The concentration of ghrelin and the ratio of ghrelin/obestatin have a decline tendency on serum,obestatin has an up tendency and NPY was higher than the normal weight rats distinctly.However,on the hypothalamic tissues,the concentrations of ghrelin,obestatin and the ratio of Ghrelin/Obestatin decreased greatly.The ghrelin receptor GHSR-1a mRNA decreased distinctly,NPY mRNA had no change and GPR-39 mRNA on hypothalamic can not been found.It concludes that 1) the ratio of ghrelin/obestatin has certain influence on the etiology of obese rats.2) ghrelin mediated food intake and energy metabolism through hypothalamic centers.3) Obestatin- GPR-39 may be no use on energy homeostasis. |
