大鼠骨骼肌超微结构及恢复因子的变化

摘    要:研究7天力竭运动及恢复期的大鼠骨骼肌超微结构和增殖细胞核抗原(PCNA)的变化。研究对象为8周龄健康雄性大鼠36只,随即分为6组(C、E0、E12、E24、E48和E72),每组6只。采用Western blot免疫印迹法检测大鼠增殖细胞核抗原(PCNA)的变化,电镜观察大鼠骨骼肌超微结构的变化。结果表明:大鼠力竭后以及恢复期间骨骼肌超微形态发生不同程度的改变,特别是力竭运动组(E0、E24)骨骼肌内部结构明显遭到破坏,肌节紊乱、肌间隙增宽,部分线粒体出现变形、扭曲、肿大;大鼠在7天力竭训练后,骨骼肌内增殖细胞核抗原(PCNA)发生变化,其中安静组内的PCNA含量很少,而力竭运动组的E12组-E72组的增殖细胞核抗原含量均有显著增加(P0.05),显示此时大鼠内由于损伤导致增殖细胞核抗原含量的增加,大鼠内自体修复过程在E24达到峰值(P0.01)。结论:7天力竭运动及恢复期,大鼠骨骼肌内部结构遭到了不同程度的破坏,但骨骼肌中增殖细胞核抗原(PCNA)在力竭运动后明显增加,在E24组达到顶峰,表明增殖细胞核抗原可能加快软组织损伤的修复。

Abstract:Objective: The purpose of this paper was to study the changes of skeletal muscle ultrasructure and to explore the changes of proliferating cell nuclear antigen during heavy load exercise and recovery in rat. Methods : 36 male SD rats of 8 weeks old were randomly divided into 6 groups (C, E0, E12, E24, E48 and E72) ), and each group had six rats. The part of tibialis anterior was to measure the proliferating cell nuclear antigen by Western bolt. A part of tibialis anterior were fixed for producing uhrathin sections and observed the uhrastructural changes by transmission electron microscope. Result: By ob- serving the electron micrographs of the tibialis anterior, we found the skeletal muscle structure in exhaustive exercise groups (E0,E24) were significantly damaged with the myofibril disorder and muscle gap widened, some mitochondrias were de- formed, distorted and swollen. By Western blot to measure proliferating cell nuclear antigen(PCNA) of the tibialis anterior, after exhaustive exercise of one week, we found that PCNA contents in the tibialis anterio increased significantly in the exhaus- tive exercise groups compared with the control group (P 〈 0. 05) , especially, E24 was increased significantly compared E0 (P 〈 0. 01 ) . Conclusion :The uhrastrncture of skeletal muscle structure in exhaustive exercise groups were significantly damaged during 7 days of heavy load exercise and recovery. The proliferating cell nuclear antigen (PCNA) of the tibialis ante- rior was increased after exhaustive exercise during heavy load exercise and recovery. PCNA expressed to a peak at E24, and it indicated that proliferating cell nuclear antigen can be accelerated the repair of soft tissue injury.
