晚期运动预适应保护效应中大鼠心肌αPKC表达和分布变化的研究

摘    要:目的观察晚期运动预适应保护效应中大鼠心肌αPKC表达和分布的变化,探讨该变化的作用及机制。方法将SD大鼠随机分为对照组(C组)、力竭组(EE组)、晚期运动预适应组(LEP组)和晚期运动预适应+力竭组(LEP+EE组)。采用一次间歇性大强度跑台运动建立运动预适应动物模型,采用一次力竭跑台运动建立力竭动物模型。运用蛋白质印迹法,定量检测大鼠心肌组织αPKC和p-αPKC表达水平;运用免疫组织化学法,定位检测大鼠心肌细胞αPKC和p-αPKC的分布。结果 1)蛋白质印迹。①αPKC:与C组比较,EE组和LEP组αPKC表达水平显著升高(P<0.05);与EE组比较,LEP+EE组显著降低(P<0.05)。②p-αPKC:与C组比较,EE组p-αPKC表达水平显著升高(P<0.05);与EE组比较,LEP+EE组显著降低(P<0.05)。2)免疫组织化学。①αPKC:免疫阳性产物呈棕黄色,以颗粒状弥散分布在胞质中或聚集在闰盘处;各组差异在于颜色有深有浅,颗粒或大或小。②p-αPKC:免疫阳性产物呈棕黄色,C组和LEP+EE组分布在胞质中;除上述现象外,EE组有细胞膜聚集和核染现象,LEP组出现细胞膜聚集现象。结论αPKC参与了LEP效应对力竭运动大鼠心肌的保护作用,在该作用中αPKC表现为表达水平和激活水平均下调与定位改变。

Abstract:The paper observes the change of expression and distribution of αPKC in the myocardium of rat during the cardio-protective effect of the late exercise preconditioning and explores the function and mechanism of the change.Methods: Rats were randomly assigned to C group,EE group,LEP group and LEP+EE group.The rat model of EP was set up by means of the single bout of intermittent high-intensity treadmill exercise,and the rat model of exhaustion in the manner of exhaustive treadmill exercise.Western Blot was introduced to quantitatively determine the content of αPKC and p-αPKC in the myocardium of rat.Immuno-histochemistry method was used to determine the distribution of αPKC and p-αPKC.Results: 1) protein mark.① αPKC: compared with C group,there is a significant increase of expression distribution for EE group and LEP group(P<0.05);compared with EE group,there is a significant decrease for LEP+EE(P<0.05).② p-αPKC: compared with C group,there is a significant increase of expression level for EE group(P<0.05);compared with EE group,there is a significant decrease for LEP+EE(P<0.05).2) Immuno-histochemistry.① αPKC: immune positive products appear orange yellow and scatter in cytoplast or gather around intercalated disc in the form of particles,with the different shades of color and particles in each group.② p-αPKC: immune positive products appear orange yellow and scatter in cytoplast in C group and LEP+EE group.Besides,they gather around cytomembrane or appear on it.Conclusion: αPKC plays a role in the myocardium of rat during the cardioprotection of LEP effect in EE,which shows the downturn of both expression level and stimulation level and the change of orientation.
