苹果皮孔斑点病的病原菌分离与鉴定

摘    要:2010年2月下旬,从南阳市超市购买的苹果中,发现感染皮孔斑点病的病果,从病果上随机采集病斑,经表面消毒后将病斑置于PDA平板上进行病原菌分离.从19个病斑中共分离获得17株形态相同的链格孢属真菌,经单孢纯化后对代表性致病菌株AP-5-3进行病原学鉴定.根据病原菌的形态特征及其核糖体基因内转录间隔区(rDNA-ITS)序列分析,将菌株AP-5-3鉴定为细交链孢菌(Alternaria alternata).该菌的最适生长温度为28℃,最适合生长pH为6.5.本文对苹果皮孔斑点病的病因进行了讨论.

Abstract:A few diseased fruit with lenticel spot symptoms were found among the apples purchased from a supermarket in Nanyang city in late February 2010. Lesions were randomly sampled from an affected apple, and used for pathogen isolation after surface disinfection. A total of 17 Alternaria isolates showing the same morphology were isolated exclusively from 19 apple lenticel lesions. One representative isolate AP- 5 -3 was used for etio- logical characterization after being purified by single spore isolation. The isolate was identified as Alternaria alternata based on its morphological characteristics and rDNA - ITS (internal transcribed spacer) sequence analysis. The most suitable temperature and pH for the growth of the pathogen were 28 ℃ and 6.5, respectively. The causal agent of apple lenticel spot disease was discussed in the paper.
