白藜芦醇/富勒烯纳米组装体的制备——在胆管支架抗炎涂层中的应用评估

摘    要:采用白藜芦醇(Res)和富勒烯(C60)结合制备Res/C60纳米组装体并验证其安全性和抗炎性。通过扫描电子显微镜(SEM)、拉曼光谱和X射线衍射研究了纳米组装体形貌、粒径大小及波谱峰值特征。其次,通过紫外分光光度法研究了Res的药物缓释行为。最后,通过MTT法在人脐静脉内皮细胞(HUVECs)层面验证了Res/C60纳米组装体的安全性以及体外的抗炎性。研究发现,Res/C60纳米组装体呈短棒状,横截面平均直径为(379.74±33.47)nm。拉曼光谱和X射线衍射特征峰显示Res/C60纳米组装体被成功制备。Res/C60纳米组装体的聚氨酯(PU)涂层中Res药物在30 d内累积释放量可达60%左右。胆管支架涂层对HUVECs细胞毒性较低,且通过释放Res可显著抑制炎症因子的释放。

Abstract:The molecular self-assembly technology is used to assemble resveratrol(Res)and fullerene(C60)with into nanostructures,and polyurethane(PU)is used as the carrier for anti-inflammatory bile duct stents coating field.The Res/C60 nano-assembly has a short rod shape with an average cross-sectional diameter of 379.74±33.47 nm.The results of Raman spectroscopy and X-ray diffraction patterns prove that the Res/C60 nano-assembly is successfully prepared.The Res drug release amount of the PU coating loaded Res/C60 nano-assemblies showed the cumulative release amount was up to about 60%within 30 days.The cytotoxicity test and anti-inflammatory experiment show that the coating is less cytotoxic to HUVECs,and it can significantly inhibit the release of inflammatory factors by releasing Res.
