分光光度计有效测量范围的生化实验验证

摘    要:为了在生物化学实验讲解中更直观地说明使用分光光度计时的有效测量范围,分别测定了不同浓度的苯丙氨酸和pSK质粒的光吸收值,与相应的标准浓度对照,计算出不同光吸收值的偏差,得到光吸收值与测量偏差的曲线。实验结果表明,当光吸收值小于0.1和大于1.0时,测量偏差超过10%,而当光吸收值在0.1~1.0之间时,测量偏差小于10%(当光吸收值在0.3~0.7之间时,测量偏差小于4%),测量值有效可信。本实验以氨基酸和核酸两种物质为例,验证了分光光度计的有效测量范围为0.1~1.0。

Abstract:To show the reliability of spectrophotometric measurements in the experimental teaching,A280 for serial dilution of phenylalanine and A260 for serial dilution of plasmid pSK were taken.For each sample,the percentage standard deviation was plotted.The results show that the standard deviation is up 10% for low and high spectrophotometric values,and the standard deviation is below 10% for spectrophotometric values between 0.1 and 1.0(as little as 4% for readings between 0.3 and(0.7)).The spectrophotometric values between 0.1 and 1.0 are very reproducible.
