酸处理麦秆酶解条件的优化

摘    要:以稀硫酸预处理的麦秆为底物,通过单因素试验和正交试验优化了底物的酶解条件.结果表明:酶解条件的主次顺序为底物质量浓度、酶质量浓度、酶解时间、酶解温度和pH;最佳酶解条件为底物质量浓度0.25 g/L、酶质量浓度2 g/L、酶解时间36 h、酶解温度45℃和pH为5.0,此时总还原糖含量最大为707.07 mg/g,糖含量显著增加.酶解对于提高麦秆的糖化率是非常有必要的.

Abstract:Enzymolysis conditions were optimized by single factor experiment and orthogonal experiments using wheat straw which was pretreated by dilute sulphuric acid (H2SO4).The results showed that the primary and secondary factors of enzymolysis conditions were substrate concentration,enzyme concentration,enzymatic time, enzymatic temperature and pH.The optimal enzymolysis conditions were substrate concentration of 0.25 g/L,enzyme condition of 2 g/L,enzymatic time of 36 h,enzymatic temperature of 45 ℃ and pH of 5.0.The maximum of total reducing sugar concentration was 707.07 mg/g at the optimal enzymolysis conditions,sugar yield increased dramatically,indicating that enzymolysis is very necessary to improve the saccharification efficiency of wheat straw.
