牡丹叶绿素的提取及其性质研究 总被引：11，自引：0，他引：11
以分光光度法首次研究了牡丹叶绿素的提取方法,用8种提取液在不同时间内优选最佳实验条件,测定了不同品种牡丹叶绿素含量及不同温度下各品种叶绿素的稳定性.结果表明:室温下,以丙酮:95%乙醇=2:1为提取液,浸取24h的方法最佳,不同品种牡丹叶绿素含量差别不大;叶绿素稳定性受温度的影响不大. 相似文献

目的 :比较三种不同的DNA提取方法提取人全血基因组DNA对DNA提取效率、纯度以及PCR扩增的效果影响。方法 :分别应用中山医科大学达安公司DNA提取液 ,珠海黑马医学仪器有限公司DNA提取液 ,胍盐酸法三种不同的方法提取人全血基因组DNA。结果 :三种方法提取的DNA纯度平均分别为 :1.4 8,1.5 1,1.5 6 ,各组间差异无显著性。 2 0 0 μL全血中所提取DNA总量平均分别为 1.6 μg ,2 .3μg ,4 .1μg。用 0 .8%琼脂糖凝胶电泳鉴定胍盐酸法制备的DNA效果较其它两种方法好 ,PCR扩增效果差异不明显。结论 :胍盐酸法提取的DNA总量明显高于其它两种方法 ,提取效率高 ,为三种全血基因组DNA提取方法中之首选。 相似文献

针对冀东地区常见的九种树木叶片及相应虫沙,对其叶绿素提取液的吸光度值及吸收光谱进行了对比研究。结果显示：鲜叶乙醇提取液在430nm和663nm有强吸收峰,虫沙乙醇提取液在410nm和663nm有强吸收峰。相同条件下,乙醇对于鲜叶片中叶绿素的提取效果优于丙酮;而丙酮对于各种虫沙中叶绿素的提取效果优于乙醇。 相似文献

以甜柿与涩柿叶片为试材,采用3种不同有机溶剂作为提取液,比较研究了甜柿和涩柿2种柿树叶片叶绿素含量的测定方法．结果表明：采用研磨法提取叶绿素时,溶剂以体积分数为95％的乙醇最好,甜柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为1．428、1．086和0．341mg／g,涩柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为0．951、0．737和0．213mg／g,均显著高于体积分数为80％的丙酮和二者的混合液;采用浸泡法提取叶绿素时,以体积分数为80％的丙酮和体积分数为95％的乙醇按体积比1：1混合的混合液提取效果最好,甜柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为1．478、1．089和0．388mg／g,涩柿叶片叶绿素总含量、叶绿素a含量和叶绿素b含量分别为1．180、0．886和0．294mg／g,均显著高于体积分数为95％的乙醇和体积分数为80％的丙酮．两种叶绿素提取方法比较发现,浸泡法优于研磨法． 相似文献
