目的:观察黄芪注射液配伍川芎嗪对家兔心肌缺血再灌注损伤的保护作用。方法:采用在体家兔冠状动脉阻断的缺血/复灌损伤模型,测定心肌梗死面积、血浆中乳酸脱氢酶(LDH)和肌酸激酶(CK)活性及各项心室力学指标。结果:与单纯缺血/复灌组相比,川芎嗪与黄芪两药明显降低心脏缺血/复灌后的梗死面积和血浆中LDH、CK含量,促进左室收缩压(LVSP)、最大左室收缩速率( dP/dtmax)和最大左室舒张速率(-dP/dtmax)的恢复,两药合用与单用比较各项指标的恢复更加显著。结论:黄芪与川芎嗪两药合用在保护心肌缺血再灌注损伤中有显著的协同作用。  相似文献

目的:研究心肌缺血预处理(IP)延迟性保护作用终末阶段(IP后48h至72h)的机制,以及神经型一氧化氮合酶(nNOS)、诱生型一氧化氮合酶(iNOS)是否中介了此过程。方法:采用在体家兔冠状动脉阻断的缺血/复灌损伤模型,测定心肌梗死面积、冠脉流出液中乳酸脱氢酶(LDH)和肌酸激酶(CK)含量及各项心室力学指标。结果:与单纯缺血/复灌组相比,IP后72h,明显降低心脏缺血/复灌后的梗死面积和血浆中乳酸脱氢酶(LDH)、肌酸激酶(CK)的含量,促进左室收缩压(LVSP)、最大左室收缩速率(+dP/dtmax)和最大左室舒张速率(-dP/dtmax)的恢复。使用iNOS的特异性抑制剂S-methylisothiourea sulfate(SMT)不能阻断IP后72h心肌保护作用。而nNOS的特异性抑制剂N-propyl-L-arginine(NPA)、NOS的非特异性抑制剂NG-硝基-L-精氨酸甲酯(L-NAME)阻断了IP后72h心肌保护作用。结论:IP后72h心肌保护作用依然存在,nNOS介导了心肌缺血预处理延迟性保护作用终末阶段,而与iNOS无关。  相似文献

目的:本实验观察复心汤灌胃对大鼠心肌缺血模型的影响.方法:Wistar大鼠60只,雌雄各半,随机分成6组,每组10只.正常对照组:生理盐水灌胃;模型组:生理盐水灌胃;地奥心血康组:等效剂量地奥心血康灌胃(0.12g/kg/d);复心汤高剂量组:6倍等效剂量灌胃(81g/kg/d);复心汤中剂量组:3倍等效剂量灌胃(40.5g/kg/d);复心汤低剂量组:等效剂量灌胃(13.5g/kg/d).每日早晚各一次,连续7天.第8天除正常对照组外,其余各组于给药1小时后,用垂体后叶素(Pituitrinum,pit)制备心肌缺血模型.观察大鼠的心电图(ECG)、心肌组织乳酸脱氢酶(LDH)和肌酸激酶(CK)含量的改变、左室压力下降最大速率(?dp/dtmax)、左室舒张末压(LVEDP)的变化.结果:与模型组比较,复心汤各剂量组和地奥心血康组表现出显著对抗作用(P<0.05,P<0.01).与正常组比较,模型组大鼠CK及LDH活力显著升高,具有极显著性差异(P<0.001).结论:复心汤有治疗和预防心肌缺血的作用,对于心脏舒张功能的防治亦有明显的作用.  相似文献

目的:研究肢体缺血预处理(LIP)对大鼠缺血再灌后心脏损伤的影响。方法:采用反复捆绑后肢造成缺血预适应的方法,观察其对心肌缺血再灌后梗死面积和形态学改变的影响,对心肌CK-MB和cTn-I的影响,以及对抗氧化物酶的影响。结果:与缺血再灌组比较,预处理能明显降低心肌梗死面积,减轻心肌细胞的损害,减少心肌酶的漏出并能提高SOD的活性。结论:LIP对心肌损伤具有保护作用。  相似文献

目的:探讨核桃分心木提取物总黄酮、总皂苷对家兔心肌缺血再灌损伤的影响。方法:30只家兔随机分为5组,假手术组单纯缺血组,缺血再灌组分心木治疗A组B组。结果:核桃分心木提取物总皂苷治疗A组、B组与再灌注组相对比,血清中MDA含量明显减少,SOD活性明显增高,A组与B组相比,血清MDA含量和SOD活性差异不显著。讨论:提示核桃分心木提取物总皂苷可通过某种机制抑制脂质过氧化过程,提高内源性抗氧化酶活性;而减轻心肌缺血一再灌注中产生的自由基对心肌细胞的损伤,具有抗氧化作用。  相似文献

目的探讨苏木素碱性品红苦味酸(HBPF)染色对诊断缺血/再灌注心肌缺血缺氧程度的价值.方法采用结扎大鼠左冠状动脉前降支,心肌缺血15min,再灌1h制作缺血-再灌注模型.用HBPF染色观察心肌形态学改变,同时与苏木素伊红(HE)染色比较.结果HBPF染色在大鼠心肌缺血再灌注1h心肌明确显示心肌细胞红染呈灶状分布,而HE染色无改变.结论HBPF染色对心肌缺血再灌注损伤程度有诊断意义。  相似文献

目的:探讨不同时相探讨山茶花提取物(extract of Camellia japonica L,ECJ)预处理对小鼠缺血再灌注损伤的影响。方法:线栓法制备小鼠缺血再灌注模型(MCAO),观察不同时相ECJ预处理后小鼠大脑中动脉缺血再灌注致脑梗死面积、血清中乳酸脱氢酶(LDH)和丙二醛(MDA)含量等指标的变化。结果:ECJ预处理延迟时相能明显减少脑梗塞面积并可明显降低血清中LDH活性、MDA的含量。结论:ECJ预处理延迟时相对小鼠脑缺血再灌注损伤有一定的保护作用。  相似文献

缺血预处理是指心肌反复多次短暂缺血/再灌注,对随后长时间心肌缺血/再灌注的一种保护.缺血预处理对心肌的保护机制十分复杂,一般公认为线粒体ATP敏感性钾通道(mitoKATP)是心肌缺血预处理保护机制的终末效应器之一.mitoKATP开放后可通过以下3条途径对抗心肌缺血损伤:减少线粒体膜Ca2 通道对Ca2 内流的驱动力,抑制Ca2 内流,减轻心肌细胞钙超载;保护线粒体呼吸链,减轻自由基释放引起的损伤;抑制线粒体转换孔开放,防止细胞色素C从线粒体释放,减少细胞凋亡.  相似文献
