目的:研究大黄素对脑缺血-再灌注小鼠探索认知功能的改善作用及其机制。方法:采用改进的Himori法暂时性阻断两侧颈总动脉制备小鼠脑缺血-再灌注损伤模型,进行探索实验和跳台实验,观察腹腔注射大黄素10.0、1.0、0.1 mg·kg-1对脑缺血-再灌注小鼠探索认知功能的改善作用,并对各剂量组小鼠脑组织和血液中一氧化氮(nitric oxide,NO)含量和一氧化氮合酶(nitric oxide synthase,NOS)活力、脑组织中过氧化氢(hydrogen peroxide,H2O2)含量和过氧化氢酶(catalase,CAT)活力进行测定,并测定脑指数。结果:大黄素可改善脑缺血-再灌注损伤所致的探索认知功能障碍;减少NO和H2O2含量,降低NOS活力,提高CAT活力和增加脑指数。结论:大黄素对脑缺血-再灌注损伤小鼠探索认知功能有改善作用,其作用机制可能是通过降低NOS活力和增强CAT活力,提高脑组织对氧自由基的清除能力,从而减轻缺血-再灌注引起的脑组织损伤。  相似文献

中风回春丸对小鼠灌胃的最大耐受量力72g/kg.按最大耐受量的1/6,1/12,1/24设置药效学3个剂量组,中风回春丸灌胃给药可防止动物脑缺血再灌注死亡,使缺血再灌注损伤产生的丙二醛含量显著降低,并有保护超氧化物歧化酶作用.中风回春丸还可使血凝块溶解时间及全血浆凝块溶解时间显著缩短,抑制ADP和CaCl_2诱导的血小板聚集,增加红细胞压积,改善微循环障碍,有活血化瘀作用.  相似文献

目的:观察二氢石蒜碱(dihydrolycorine,DL)对大鼠局部脑缺血/再灌注损伤的治疗作用和可能机制。方法:线栓法制备右侧大脑中动脉局灶性脑缺血模型,缺血1 h再灌注24 h。SD大鼠随机分为假手术组、模型组和DL低、中、高剂量组,于大鼠脑缺血后即刻及复灌后11 h及23 h DL组腹腔注射DL(10,20,40 mg.kg-1)治疗,假手术组和模型组则注射生理盐水对照。观察不同剂量DL对脑缺血再灌注大鼠神经功能状态、脑梗塞面积、脑水肿程度和脑组织髓过氧化物酶(myeloperoxidase,MPO)活性的影响。结果:脑缺血后给予DL可明显降低缺血侧脑梗死及脑水肿程度,使神经病学症状得到明显改善,降低缺血再灌注损伤后脑组织内MPO的活性(P<0.05)。结论:DL对脑缺血/再灌注损伤具有治疗作用,可能与其减轻脑缺血再灌注所致脑组织的炎症反应有关。  相似文献

目的:探讨不同时相探讨山茶花提取物(extract of Camellia japonica L,ECJ)预处理对小鼠缺血再灌注损伤的影响。方法:线栓法制备小鼠缺血再灌注模型(MCAO),观察不同时相ECJ预处理后小鼠大脑中动脉缺血再灌注致脑梗死面积、血清中乳酸脱氢酶(LDH)和丙二醛(MDA)含量等指标的变化。结果:ECJ预处理延迟时相能明显减少脑梗塞面积并可明显降低血清中LDH活性、MDA的含量。结论:ECJ预处理延迟时相对小鼠脑缺血再灌注损伤有一定的保护作用。  相似文献

目的:观察脑缺血再灌注损伤后,大黄酚脂质体对小鼠海马神经元凋亡的影响,探讨大黄酚脂质体对脑缺血再灌注损伤小鼠海马的保护作用及其机制.方法:采用暂时性阻断颈总动脉的方法建立小鼠脑缺血再灌注损伤模型,腹腔注射(ip)大黄酚脂质体10.0,1.0,0.1 mg·kg-1,观察小鼠脑缺血再灌注后神经功能学和病理形态学的改变,并采用免疫组化方法检测海马神经元凋亡相关蛋白半胱氨酰天冬氨酸蛋白酶-3(caspase-3),Bcl-2和Bcl-2-Associated X(Bax)的表达.结果:大黄酚脂质体可明显抑制脑缺血再灌注引起的神经元的丢失,提高Bcl-2 的表达,降低caspase-3 和Bax 的表达,提高神经功能评分,减少病理形态改变,减轻海马神经元损伤,其中以10.0 mg·kg-1 组大黄酚脂质体的作用最为明显(P<0.05).结论:脑缺血再灌注损伤后应用大黄酚脂质体对海马神经元有明显的保护作用,其机制可能与上调Bcl-2 的表达,下调caspase-3 和Bax 的表达有关.  相似文献

目的:研究大黄素对脑缺血再灌注小鼠记忆功能的保护作用.方法:采用改进的Himori法暂时性阻断小鼠两侧颈总动脉制备脑缺血再灌注损伤模型,进行避暗实验、穿梭实验、常压耐缺氧实验、断头耐缺氧实验和亚硝酸钠中毒耐缺氧实验,观察大黄素(10.0,1.0,0.1 mg·kg-1,ip)对脑缺血再灌注小鼠记忆功能和耐缺氧能力的影响,并对各剂量组小鼠脑组织和血液中谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)活力和超氧化物歧化酶(superoxide dismutase,SOD)活力进行测定.结果:大黄素可改善脑缺血再灌注损伤所致的记忆功能障碍,明显延长小鼠耐缺氧生存时间,增加GSH-PX和SOD活力.结论:大黄素对脑缺血再灌注损伤小鼠记忆功能有保护作用,其作用机制可能是通过增强抗氧化酶GSH-PX和SOD活力,提高脑组织对氧自由基的清除能力和耐缺氧能力,从而减轻缺血再灌注引起的脑组织损伤.  相似文献

目的:研究黄芪总苷(astragalosides,AST)预处理对小鼠脑缺血再灌注损伤的保护作用及其机制。方法:采用间断静脉推注AST模拟预适应的实验方法,观察小鼠大脑中动脉栓塞再灌致脑梗死体积、血清中血清中乳酸脱氢酶(LDH)、一氧化氮(NO)及丙二醛(MDA)的含量的变化。结果:AST(25,50,100mg.kg^-1)预处理明显减少脑梗塞体积,并可明显降低血清中LDH含量,同时明显增加血清中NO的含量。结论:黄芪总苷预处理对小鼠脑缺血再灌注损伤具有明显的保护作用。  相似文献

[目的]研究硫酸镁对大鼠肾缺血再灌注损伤的保护作用。[方法]夹闭大鼠肾动脉25分钟后复流,制作肾缺血再灌注模型,设假手术阴性对照组、缺血再灌注模型组、维拉帕米治疗组及硫酸镁治疗组,比较四组大鼠肾组织匀浆SOD、MDA及血BUN、CR测定值。[结果]①缺血再灌注模型组MDA和BUN、CR比假手术阴性对照组显著升高,SOD显著降低;②维拉帕米治疗组MDA和BUN、CR比缺血再灌注模型组显著降低,SOD显著升高;③硫酸镁治疗组MDA及BUN比缺血再灌注模型组显著降低,CR无显著差异,SOD显著升高。④硫酸镁治疗组SOD、MDA及BUN、CR与维拉帕米治疗组无显著性差异。[结论]硫酸镁对大鼠实验性肾缺血再灌注损伤的有保护作用,其机制可能与减轻钙超载,促进肾脏内源性氧自由基清除剂SOD活性、灭活氧自由基;抑制脂质过氧化水平有关。  相似文献

如今延长溶栓时间窗、减轻再灌注损伤是缺血性脑血管病亟待解决的的问题,缺血后适应提供了一种可能的解决方法,故成为研究的热点。缺血后适应通常指的是在组织缺血-再灌注之后进行的一系列短暂血管闭塞/血管再灌注,诱导组织针对缺血-再灌注损伤产生内源性保护作用,减少组织器官缺血-再灌注损伤。该文将综述缺血后适应脑保护的基本机制:减少缺血-再灌注后脑血流的改变,减少氧化应激产物的产生,抗炎,相关信号传导通路改变。  相似文献

研究目的:探索小分子酪氨酸激酶抑制剂舒尼替尼(sunitinib)对结肠肿瘤微环境中的肿瘤相关成纤维细胞的作用及其机制。创新要点:舒尼替尼通过抑制肿瘤间质成纤维细胞的生长,间接发挥抗肿瘤效应,为结肠癌综合治疗的提供新途径。研究方法:通过细胞周期分析和细胞增殖测定进行舒尼替尼体外抑制肿瘤细胞的研究。采用Western-blot检测磷酸化血小板衍生生长因子β受体(PDGFR-β)、蛋白激酶B(Akt)及细胞外信号调节蛋白激酶(ERK)的蛋白水平。通过注射肠腺癌细胞株SW620和结肠成纤维细胞构建的裸鼠移植瘤模型来研究舒尼替尼的体内抑瘤效果。重要结论:舒尼替尼可有效抑制结肠癌来源的原代结肠成纤维细胞生长,该抑制作用主要通过抑制血小板衍生生长因子(PDGF)信号通路得以实现。  相似文献

研究目的:应用磁共振弥散加权成像(DWI)监测荷瘤裸鼠肝细胞性肝癌表观弥散系数(ADC)的变化,探讨ADC对索拉菲尼治疗肝细胞性肝癌的疗效预测。创新要点:探讨磁共振DWI对索拉菲尼治疗肝癌疗效的早期评估,期望能够指导临床合理用药。研究方法:荷瘤裸鼠分成两组(治疗组和对照组)共40只,使用GE 3.0T磁共振成像系统和小动物线圈,分别于服药前后1、10、14和18天(40 mg/kg,一天两次)进行T1加权成像(T1WI)、T2加权成像(T2WI)和DWI扫描。在弥散加权图像上进行ADC测量,对比不同区域和不同时间点的差异。重要结论:索拉菲尼治疗10天后,伴随肝癌肿瘤的缩小,肿瘤内部ADC值明显上升,病理切片显示治疗组瘤体中心小血管闭塞,凋亡细胞可见,此时瘤体内坏死细胞并未大量显现,说明ADC值的改变可以反映索拉菲尼的疗效。  相似文献

Background and objective

It has been shown that macrophages play an important role in the development of severe acute pancreatitis (SAP), and eventually lead to multiple organ failure (MOF). Clodronate-liposome selectively depleted macrophages. This study was to investigate the role of renal macrophage infiltration in acute renal injury in rats with SAP and to evaluate the potential of superparamagnetic iron oxide (SPIO)-enhanced magnetic resonance imaging (MRI) for diagnosis.

Methods

Superparamagnetic Fe3O4 nanoparticles were prepared by chemical coprecipitation. SPIO-liposomes and SPIO-clodronate-liposomes were prepared by the thin film method. SAP models were prepared by injection of sodium taurocholate into the subcapsular space of rat pancreas. Sprague-Dawley rats were randomly divided into a control group, SAP plus SPIO-liposome (P) group, and SAP plus SPIO-clodronate-containing liposome (T) group. Kidney injury was evaluated by T2-weighted MRI scan. The levels of serum amylase (SAM), blood urea nitrogen (BUN), and serum creatinine (SCr) were measured by an automated enzymatic method. Serum tumor necrosis factor-α (TNF-α) was measured by enzyme-linked immunosorbent assay (ELISA). Pathological changes in the pancreas and kidney were observed using hematoxylin and eosin (H&E) staining, while cell apoptosis was detected with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining. In addition, the macrophage markers (CD68) of the renal tissue were detected with immunohistochemistry.

Results

The pathological changes in the pancreas and kidneys of rats in the T group were milder than those in the P group. The MRI signal intensity of the kidneys in the P and T groups was significantly lower than that in the control group. There were significant changes in the two experimental groups (P<0.01). The levels of SAM, Bun, SCr, and TNF-α in rats in the P group were higher than those in the control group (P<0.01) and in the T group (P<0.01). The apoptosis of the kidney in the T group was higher than that in the P group at 2 and 6 h (P<0.01).

Conclusions

Clodronate-containing liposomes protected against renal injury in SAP rats, and SPIO can be used as a tracer for MRI examination to detect renal injury in SAP rats. SPIO-aided MRI provided an efficient non-invasive way to monitor the migration of macrophages after renal injury in rats with SAP.  相似文献

Objective:To investigate the effect of Anethum graveolens(AG) extracts on the mounting frequency,histology of testis and epididymis,and sperm physiology.Methods:Male rats induced by cold immobilization before treating with vehicle or AG extracts [50,150,and 450 mg/kg body weight(BW)] via gastric tube for consecutive 1,7,and 14 d were examined for mounting frequency,testicular phosphorylation level by immunoblotting,sperm concentration,sperm acrosome reaction,and histological structures of testis and epididymis,respectively.Results:AG(50 mg/kg BW) significantly increased the mounting frequency on Days 1 and 7 compared to the control group.Additionally,rat testis treated with 50 mg/kg BW AG showed high levels of phosphorylated proteins as compared with the control group.In histological analyses,AG extract did not affect the sperm concentration,acrosome reaction,and histological structures of testis and epididymis.Conclusions:AG extract enhances the aphrodisiac activity and is not harmful to sperm and male reproductive organs.  相似文献

Objective

To evaluate the potential adjuvant effect of Agrocybe aegerita lectin (AAL), which was isolated from mushroom, against a virulent H9N2 strain in vivo and in vitro.

Methods

In trial 1, 50 BALB/c male mice (8 weeks old) were divided into five groups (n=10 each group) which received a subcutaneous injection of inactivated H9N2 (control), inactivated H9N2+0.2% (w/w) alum, inactivated H9N2+0.5 mg recombinant AAL/kg body weight (BW), inactivated H9N2+1.0 mg AAL/kg BW, and inactivated H9N2+2.5 mg AAL/kg BW, respectively, four times at 7-d intervals. In trial 2, 30 BALB/c male mice (8 weeks old) were divided into three groups (n=10 each group) which received a subcutaneous injection of inactivated H9N2 (control), inactivated H9N2+2.5 mg recombinant wild-type AAL (AAL-wt)/kg BW, and inactivated H9N2+2.5 mg carbohydrate recognition domain (CRD) mutant AAL (AAL-mutR63H)/kg BW, respectively, four times at 7-d intervals. Seven days after the final immunization, serum samples were collected from each group for analysis. Hemagglutination assay, immunogold electron microscope, lectin blotting, and co-immunoprecipitation were used to study the interaction between AAL and H9N2 in vitro.

Results

IgG, IgG1, and IgG2a antibody levels were significantly increased in the sera of mice co-immunized with inactivated H9N2 and AAL when compared to mice immunized with inactivated H9N2 alone. No significant increase of the IgG antibody level was detected in the sera of the mice co-immunized with inactivated H9N2 and AAL-mutR63H. Moreover, AAL-wt, but not mutant AAL-mutR63H, adhered to the surface of H9N2 virus. The interaction between AAL and the H9N2 virus was further demonstrated to be associated with the CRD of AAL binding to the surface glycosylated proteins, hemagglutinin and neuraminidase.

Conclusions

Our findings indicated that AAL could be a safe and effective adjuvant capable of boosting humoral immunity against H9N2 viruses in mice through its interaction with the viral surface glycosylated proteins, hemagglutinin and neuraminidase.

研究目的:探讨早期经尿道前列腺隧道式电切治疗前列腺癌近距离照射治疗术后尿潴留的适应症及其对近距离照射治疗疗效的影响。创新要点:前列腺癌近距离照射治疗已经成为75岁以上患者的首选,前列腺癌近距离照射治疗术后尿潴留发生率为1.5%~22.0%。经尿道前列腺电切常用于药物治疗无效的反复尿潴留患者,且手术时机多在近距离照射治疗术后6个月以后。经尿道前列腺隧道式电切治疗前列腺癌近距离照射治疗术后尿潴留可在6个月内进行,患者尿道症状从IV(出现尿潴留,需要导尿)下降为0级(没有症状)或I级(轻度,中度尿频,2~3次/晚),较术前改善明显,且对近距离照射治疗疗效无明显影响。研究方法:浙江大学医学院附属邵逸夫医院2009年2月至2013年7月间所有接受前列腺癌近距离照射治疗的病例共190例,其中9例(4.7%)患者术后出现反复尿潴留,且早期行经尿道前列腺隧道式电切术治疗,回顾分析其临床资料。重要结论:前列腺癌近距离照射治疗术后反复尿潴留,在严格防护下早期行经尿道前列腺隧道式电切是安全有效的,且电切后对近距离照射治疗疗效无明显影响。  相似文献

研究目的:通过对一个中国Nance-Horan综合征家系的临床表型及基因突变分析,揭示本家系的致病遗传机制。研究方法:对该Nance—Horan综合征家系的一个男性患者进行全外显子组测序,结合此家系临床表型及遗传方式分析,选定x染色体上NHS基凶上的一个无义突变c.322G〉T(E108x)为可疑致病突变。通过聚合酶链式反应(PCR)和Sanger测序,对该家系内其他成员进行NHS基因突变分析,同时对50名健康对照者的NHS基因的突变检测结果进行对比。另外,将该突变的位点第108位氨基酸残基进行多物种NHS蛋白内序列比对。最后,对该家系成员眼部及全身的临床特点进行全面检查和分析。重要结论:全外显子组测序结合Sanger测序发现NHS基因第一个外显子上的c.322G〉T(E108X)突变为引起该家系临床病变的突变位点;多物种NHS蛋白内序列比对发现该突变位点第108位氨基酸残基位于高度保守区;临床表型分析发现该家系内存在表型异质性。此家系为国内首次报道的无义突变引起的Nance-Horan综合征家系。  相似文献

Objective: This study was designed to investigate the pharmacokinetics of borneol in the pathological conditions of stroke and evaluate the pharmacokinetic differences of borneol caused by stroke after oral administration of borneol and Xingnaojing (XNJ). Methods: The rats were divided into two groups, ischemia-reperfusion (IR) and sham-operated (SO) rats. Each group contained two subgroups: pure borneol and XNJ subgroups. After administration with the same dosages of borneol 162.0 mg/kg, plasma samples were collected. The cerebral ischemia-reperfusion model was created by reversible middle cerebral artery occlusion (MCAO). The blood samples were collected punc- tually after oral administration and a specific gas chromatographic system-flame ionization detector (GC-FID) method was developed and employed to determine the level of borneol in the plasma. The pharmacokinetic parameters were analyzed using non-compartmental methods with Kinetica. Results: After administration of borneol, the maximum plasma concentration (Cmax) and area under the curve (AUC) values in stroke rats significantly increased by 302% and 275%, respectively, compared with the SO rats, and the same phenomenon appeared after administration of XNJ. In the rats with the same physiological conditions, the Cmax and AUC had higher values in the borneol subgroup (P〈0.05). Conclusions: These results suggest that the pathological damages of ischemia-reperfusion have a significant impact on the pharmacokinetic traits of borneol and that there are some components in XNJ inhibiting the absorption of borneol.  相似文献

Background

Ketoconazole (KET), an antifungal drug, has adverse effects on the male reproductive system. Pre-treatments with antioxidant plant against testicular damage induced by KET are required. The flowers of Clitoria ternatea (CT) are proven to have hepatoprotective potential. However, the protective effect on KET-induced testicular damage has not been reported.

Objective

To investigate the protective effect of CT flower extracts with antioxidant activity on male reproductive parameters including sperm concentration, serum testosterone level, histopathology of the testis, and testicular tyrosine phosphorylation levels in rats induced with KET.

Methods

The antioxidant activity of CT flower extracts was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) assays. Male rats were treated with CT flower extracts (10, 50, or 100 mg/kg BW) or distilled water via a gastric tube for 28 d (preventive period: Days 1–21) and induced by KET (100 mg/kg BW) via intraperitoneal injection for 7 d (induction period: Days 22–28). After the experiment, all animals were examined for the weights of the testis, epididymis plus vas deferens and seminal vesicle, serum testosterone levels, sperm concentration, histological structures and diameter of testis, and testicular tyrosine phosphorylation levels by immunoblotting.

Results

The CT flower extracts had capabilities for DPPH scavenging and high reducing power. At 100 mg/kg BW, the extract had no toxic effects on the male reproductive system. Significantly, in CT+KET groups, CT flower extracts (50 and 100 mg/kg BW) alleviated the reduction of reproductive organ weight parameters, testosterone levels, and sperm concentration. In addition, CT flower extracts gave protection from testicular damage in KET-induced rats. Moreover, in the CT100+KET group, CT flower extracts significantly enhanced the expression of a testicular 50-kDa tyrosine phosphorylated protein compared with that of other groups.

Conclusions

C. ternatea flower extracts possessing antioxidant activity are not harmful to the male reproductive system and can protect against testicular damage in KET-induced rats.  相似文献
