Defining knowledge translation activities using Gibbons and Nowotny's Mode I and II forms of knowledge production, health researcher (N = 240) characteristics are compared with their knowledge translation activities. Applied researchers (n = 168) engaged in more Mode II activity (p < .001), perceived their work as having more impact (p < .001), and reported higher levels of relational capital (p < .001) when compared to basic researchers (n = 72). Those from medical schools (n = 128) had more publications (p = .004) as did applied researchers in medical school compared to applied researchers from other faculties (p = .001). These findings provide early empirical insights into emerging tensions in environments where Mode I and Mode II activities co-exist; tensions that may extract unintended costs from researchers. Such costs may be paid in the currency not only of tenure and promotion, but also of successful grant and career scientist award acquisition.  相似文献

We developed a microfluidic device to culture cellular spheroids of controlled sizes and suitable for live cell imaging by selective plane illumination microscopy (SPIM). We cocultured human umbilical vein endothelial cells (HUVECs) within the spheroids formed by hepatocellular carcinoma cells, and studied the distributions of the HUVECs over time. We observed that the migration of HUVECs depended on the size of spheroids. In the spheroids of ∼200 μm diameters, HUVECs migrated outwards to the edges within 48 h; while in the spheroids of ∼250 μm diameters, there was no outward migration of the HUVECs up to 72 h. In addition, we studied the effects of pro-angiogenic factors, namely, vascular endothelial growth factor (VEGF) and fibroblast growth factor (β-FGF), on the migration of HUVECs in the carcinoma cell spheroid. The outward migration of HUVECs in 200 μm spheroids was hindered by the treatment with VEGF and β-FGF. Moreover, some of the HUVECs formed hollow lumen within 72 h under VEGF and β-FGF treatment. The combination of SPIM and microfluidic devices gives high resolution in both spatial and temporal domains. The observation of HUVECs in spheroids provides us insight on tumor vascularization, an ideal disease model for drug screening and fundamental studies.  相似文献

Although β-Fe2O3 has a high theoretical solar-to-hydrogen efficiency because of its narrow band gap, the study of β-Fe2O3 photoanodes for water splitting is elusive as a result of their metastable nature. Raman identification of β-Fe2O3 is theoretically and experimentally investigated in this study for the first time, thus clarifying the debate about its Raman spectrum in the literature. Phase transformation of β-Fe2O3 to α-Fe2O3 was found to potentially take place under laser and electron irradiation as well as annealing. Herein, phase transformation of β-Fe2O3 to α-Fe2O3 was inhibited by introduction of Zr doping, and β-Fe2O3 was found to withstand a higher annealing temperature without any phase transformation. The solar water splitting photocurrent of the Zr-doped β-Fe2O3 photoanode was increased by 500% compared to that of the pure β-Fe2O3 photoanode. Additionally, Zr-doped β-Fe2O3 exhibited very good stability during the process of solar water splitting. These results indicate that by improving its thermal stability, metastable β-Fe2O3 film is a promising photoanode for solar water splitting.  相似文献

In this article we define an exponential informetric process to formalize these stochastic process. It is defined by combining an effort function with a density function. Without using the powerful results of Price on the cumulative advantages process this characterization clarifies the principle of least effort. Hemoglobin (Hb) Grey Lynn is a Hb variant caused by a substitution of Phe for Leu at position 91 of α1-globin chain, originally described in individual of unknown ethnic background. This article addresses the interaction of Hb Grey Lynn with a non-deletional α+-thalassemia found in Thailand, a hitherto un-described condition. The proband was adult Thai woman referred for investigation of mild anemia with Hb 90 g/L. Hb analyses using low pressure liquid chromatography raised a suspicion of abnormal Hb presence, which was failed to demonstrate by cellulose acetate electrophoresis and capillary electrophoresis. DNA sequencing identified a CTT (Leu) to TTT (Phe) mutation at codon 91 corresponding to the Hb Grey Lynn (Vientiane) [α91(FG3)Leu>Phe (α1) on α1-globin gene and a C deletion between codons 36 and 37 on α2-globin gene causing α+-thalassemia. As compared to those observed in a compound heterozygote for Hb Grey Lynn / α0-thalassemia reported previously, higher MCV (81.7 fL) and MCH (26.3 pg) values with a lower level of Hb Grey Lynn (19.7%) were observed in the proband. The normochromic normocytic anemia observed could be due to the interaction of Hb Grey Lynn with α+-thalassemia. The two mutations could be identified using PCR-RFLP and allele-specific PCR assays developed.  相似文献

A comparison of feature selection methods for an evolving RSS feed corpus   总被引:3,自引:0,他引:3  
Previous researchers have attempted to detect significant topics in news stories and blogs through the use of word frequency-based methods applied to RSS feeds. In this paper, the three statistical feature selection methods: χ2, Mutual Information (MI) and Information Gain (I) are proposed as alternative approaches for ranking term significance in an evolving RSS feed corpus. The extent to which the three methods agree with each other on determining the degree of the significance of a term on a certain date is investigated as well as the assumption that larger values tend to indicate more significant terms. An experimental evaluation was carried out with 39 different levels of data reduction to evaluate the three methods for differing degrees of significance. The three methods showed a significant degree of disagreement for a number of terms assigned an extremely large value. Hence, the assumption that the larger a value, the higher the degree of the significance of a term should be treated cautiously. Moreover, MI and I show significant disagreement. This suggests that MI is different in the way it ranks significant terms, as MI does not take the absence of a term into account, although I does. I, however, has a higher degree of term reduction than MI and χ2. This can result in loosing some significant terms. In summary, χ2 seems to be the best method to determine term significance for RSS feeds, as χ2 identifies both types of significant behavior. The χ2 method, however, is far from perfect as an extremely high value can be assigned to relatively insignificant terms.  相似文献

Many of the features to be presented are akin to, or are adaptations of, ones that have featured in the econometric literature but not in informetrics. We acknowledge in particular our debt to Lambert [Lambert, P. J. (2001). The distribution and redistribution of income. Manchester: Manchester University Press] and Kleiber and Kotz [Kleiber, C., & Kotz, S. (2003). Statistical size distributions in economics and actuarial sciences. New Jersey: Wiley] for source material in the econometrics literature. Hydrothermal fluid is essential for transporting metals in the crust and mantle. To explore the potential of Cu isotopes as a tracer of hydrothermal-fluid activity, Cu-isotope fractionation factors between Cl-bearing aqueous fluids and silicate magmas (andesite, dacite, rhyolite dacite, rhyolite and haplogranite) were experimentally calibrated. Fluids containing 1.75–14 wt.% Cl were mixed together with rock powders in Au95Cu5 alloy capsules, which were equilibrated in cold-seal pressure vessels for 5–13 days at 800–850°C and 2 kbar. The elemental and Cu-isotopic compositions of the recovered aqueous fluid and solid phases were analyzed by (LA-) ICP–MS and multi-collector inductively coupled plasma mass spectrometry, respectively. Our experimental results show that the fluid phases are consistently enriched in heavy Cu isotope (65Cu) relative to the coexisting silicates. The Cu-isotope fractionation factor (Δ65CuFLUID-MELT) ranges from 0.08 ± 0.01‰ to 0.69 ± 0.02‰. The experimental results show that the Cu-isotopic fractionation factors between aqueous fluids and silicates strongly depend on the Cu speciation in the fluids (e.g. CuCl(H2O), CuCl2 and CuCl32−) and silicate melts (CuO1/2), suggesting that the exsolved fluids may have higher δ65Cu than the residual magmas. Our results suggest the elevated δ65Cu values in Cu-enriched rocks could be produced by addition of aqueous fluids exsolved from magmas. Together with previous studies on Cu isotopes in the brine and vapor phases of porphyry deposits, our results are helpful for better understanding Cu-mineralization processes.  相似文献

This article describes a fabrication process for the generation of a leak proof paper based microfluidic device and a new design strategy for convenient incorporation of externally prepared test zones. Briefly, a negative photolithographic method was used to prepare the device with a partial photoresist layer on the rear of the device to block the leakage of sample. Microscopy and Field Emission Scanning Electron Microscopy data validated the formation of the photoresist layer. The partial layer of photoresist on the device channel limits sample volume to 7 ± 0.2 μl as compared to devices without the partial photoresist layer which requires a larger sample volume of 10 ± 0.1 μl. The design prototype with a customized external test zone exploits the channel protrusions on the UV exposed photoresist treated paper to bridge the externally applied test zone to the sample and absorbent zones. The partially laminated device with an external test zone has a comparatively low wicking speed of 1.8 ± 0.9 mm/min compared to the completely laminated device with an inbuilt test zone (3.3 ± 1.2 mm/min) which extends the reaction time between the analyte and reagents. The efficacy of the prepared device was studied with colorimetric assays for the non-specific detection of protein by tetrabromophenol blue, acid/base with phenolphthalein indicator, and specific detection of proteins using the HRP-DAB chemistry. The prepared device has the potential for leak proof detection of analyte, requires low sample volume, involves reduced cost of production (∼$0.03, excluding reagent and lamination cost), and enables the integration of customized test zones.  相似文献

Type 2 diabetes mellitus (DM) is a multifactorial disease where both genetic and environmental factors contribute to its pathogenesis. Estrogen plays an important role in type 2 DM pathogenesis. A number of polymorphisms have been reported in the estrogen receptor (ESR1), including the XbaI and PvuII restriction enzyme polymorphisms of ESR1,which may be involved in disease pathogenesis. Metallothioneins (MT) act as potent antioxidants against various oxidative damages. Very few studies have indicated the association between Estrogen Receptor-α, MT1 gene polymorphisms with type2 DM. A total of 100 type 2 diabetic women and 100 age, sex matched controls were recruited. Using the PCR based RFLP method, the PvuII and XbaI polymorphisms of ESR1 and in MT1A (rs8052394 and rs11076161) gene polymorphisms were analysed. The genotype distribution and frequency of mutated allele showed no significant differences between diabetic and non-diabetic groups in PvuII (χ2 = 2.443; P = 0.1181) or XbaI (χ2 = 1.789; P = 0.1812) and rs8052394 (χ2 = 1.154; P = 0.2840) or rs11076161 (χ2 = 0.4141; P = 0.5199), polymorphisms. This is the first Indian study to conclude that ESR1 and MT1 gene polymorphisms are not associated with increased susceptibility to type 2 diabetes in Indian women.  相似文献

For the first time, we report on the preliminary evaluation of gold coated optical fibers (GCOFs) as three-dimensional (3D) electrodes for a membraneless glucose/O2 enzymatic biofuel cell. Two off-the-shelf 125 μm diameter GCOFs were integrated into a 3D microfluidic chip fabricated via rapid prototyping. Using soluble enzymes and a 10 mM glucose solution flowing at an average velocity of 16 mm s−1 along 3 mm long GCOFs, the maximum power density reached 30.0 ± 0.1 μW cm−2 at a current density of 160.6 ± 0.3 μA cm−2. Bundles composed of multiple GCOFs could further enhance these first results while serving as substrates for enzyme immobilization.  相似文献

Cystatin C is an emerging parameter for the assessment of renal allograft function. The objective of the study was to compare the efficacy of serum cystatin C (SCys) with the established parameter serum creatinine (SCr) in the assessment of renal function in renal transplant recipients (RTR). The glomerular filtration rate (GFR) of 30 renal transplant patients and 29 control subjects was determined using 99mTc Diethylene-triamine-penta-acetate (DTPA) method. SCr was measured using an automated Jaffe’s assay and SCys was measured using latex particle enhanced turbidimetric immuno assay (PETIA). The modification of diet in renal disease (MDRD) formula was used to calculate GFR from SCr, while the Le Bricon formula was used to derive GFR based on SCys. Statistical analysis was performed using MedCalc software. SCr and SCys levels were significantly higher, while DTPA clearance was significantly lower in RTR (P < 0.0001) when compared with controls. The correlation coefficient (r value) between calculated GFR based on MDRD method and DTPA clearance was 0.343 (P = 0.06) while the calculated GFR based on Le Bricon formula was 0.694 (P < 0.001). The results have shown that SCys is a better parameter than SCr in assessing renal function in RTR. The inclusion of SCys as an additional parameter would certainly help in detection of even a marginal decline in renal function and also in adjusting the dosage of immunosuppressive drugs.  相似文献

In the past three decades, China has built more than 87 000 dams with a storage capacity of ≈6560 km3 and the total surface area of inland water has increased by 6672 km2. Leaching of N from fertilized soils to rivers is the main source of N pollution in China, but the exposure of a growing inland water area to direct atmospheric N deposition and N leaching caused by N deposition on the terrestrial ecosystem, together with increased N deposition and decreased N flow, also tends to raise N concentrations in most inland waters. The contribution of this previously ignored source of  N deposition to freshwaters is estimated in this study, as well as mitigation strategies. The results show that the annual amounts of N depositions ranged from 4.9 to 16.6 kg · ha−1 · yr−1 in the 1990s to exceeding 20 kg · ha−1 · yr−1 in the 2010s over most of regions in China, so the total mass of ΔN (the net contribution of N deposition to the increase in N concentration) for lakes, rivers and reservoirs change from 122.26 Gg N · yr−1 in the 1990s to 237.75 Gg N · yr−1 in the 2010s. It is suggested that reducing the N deposition from various sources, shortening the water-retention time in dams and decreasing the degree of regulation for rivers are three main measures for preventing a continuous increase in the N-deposition pollution to inland water in China.  相似文献

Introduction

Acute kidney injury (AKI) is common in neonatal intensive care units (NICU). In recent years, every effort is made for early detection of AKI. Our hypothesis was that serum neutrophil gelatinase-associated lipocalin (sNGAL) may be a reliable screening test for early diagnosis of AKI in premature neonates after perinatal asphyxia. Therefore, our aim was to assess the diagnostic accuracy of sNGAL for AKI in premature asphyxiated neonates.

Materials and methods

AKI was defined in the third day of life (DOL 3) as a serum creatinine (sCr) increase ≥ 26.5 μmol/L from baseline (the lowest previous sCr). According to the increase of sCr, AKI patients were divided in AKIN1 (sCr increase up to 1.9 baseline) and AKIN2 (sCr increase from 2.0 to 2.9 baseline). sNGAL levels were measured on DOL 1, 3 and 7.

Results

AKI was diagnosed in 73 (0.676) of 108 enrolled premature asphyxiated neonates. Sixty one patients (0.836) were classified in AKIN1 and 12 patients (0.164) in AKIN2. sNGAL reached the maximal concentrations on DOL 1 within 4 hours after admission to NICU, being higher in AKI compared with no-AKI group (160.8 ± 113.1 vs. 87.1 ± 81.6; P < 0.001) as well as in AKIN2 compared with AKIN1 group (222.8 ± 112.9 vs. 147.8 ± 109.9; P < 0.001). The best areas under the receiver operating characteristic curves (AUC) for prediction of AKI were 0.72 [95% (0.62-0.80) P < 0.001] on DOL1 at 2h and 0.72 [95% (0.63-0.80) P < 0.001] at 4th hour after admission respectively. The corresponding sNGAL cutoff concentrations were 84.87 ng/mL (sensitivity 69.0% and specificity 71.9%) and 89.43 ng/mL (sensitivity 65.7% and specificity 74.3%).

Conclusions

In premature asphyxiated neonates sNGAL measured within the first 4 hours of DOL 1 is predictive of the occurrence and severity of AKI. In early 2020, unprecedented lockdowns and travel bans were implemented in Chinese mainland to fight COVID-19, which led to a large reduction in anthropogenic emissions. This provided a unique opportunity to isolate the effects from emission and meteorology on tropospheric nitrogen dioxide (NO2). Comparing the atmospheric NO2 in 2020 with that in 2017, we found the changes of emission have led to a 49.3 ± 23.5% reduction, which was ∼12% more than satellite-observed reduction of 37.8 ± 16.3%. The discrepancy was mainly a result of changes of meteorology, which have contributed to an 8.1 ± 14.2% increase of NO2. We also revealed that the emission-induced reduction of NO2 has significantly negative correlations to human mobility, particularly that inside the city. The intra-city migration index derived from Baidu Location-Based-Service can explain 40.4% ± 17.7% variance of the emission-induced reduction of NO2 in 29 megacities, each of which has a population of over 8 million in Chinese mainland.  相似文献

As the reaction product of subducted water and the iron core, FeO2 with more oxygen than hematite (Fe2O3) has been recently recognized as an important component in the D” layer just above the Earth''s core-mantle boundary. Here, we report a new oxygen-excess phase (Mg, Fe)2O3+δ (0 < δ < 1, denoted as ‘OE-phase’). It forms at pressures greater than 40 gigapascal when (Mg, Fe)-bearing hydrous materials are heated over 1500 kelvin. The OE-phase is fully recoverable to ambient conditions for ex situ investigation using transmission electron microscopy, which indicates that the OE-phase contains ferric iron (Fe3+) as in Fe2O3 but holds excess oxygen through interactions between oxygen atoms. The new OE-phase provides strong evidence that H2O has extraordinary oxidation power at high pressure. Unlike the formation of pyrite-type FeO2Hx which usually requires saturated water, the OE-phase can be formed with under-saturated water at mid-mantle conditions, and is expected to be more ubiquitous at depths greater than 1000 km in the Earth''s mantle. The emergence of oxygen-excess reservoirs out of primordial or subducted (Mg, Fe)-bearing hydrous materials may revise our view on the deep-mantle redox chemistry.  相似文献

Nowadays, microfluidics is attracting more and more attentions in the biological society and has provided powerful solutions for various applications. This paper reported a microfluidic strategy for aqueous sample sterilization. A well-designed small microchannel with a high hydrodynamic resistance was used to function as an in-chip pressure regulator. The pressure in the upstream microchannel was thereby elevated which made it possible to maintain a boiling-free high temperature environment for aqueous sample sterilization. A 120 °C temperature along with a pressure of 400 kPa was successfully achieved inside the chip to sterilize aqueous samples with E. coli and Staphylococcus aureus inside. This technique will find wide applications in portable cell culturing, microsurgery in wild fields, and other related micro total analysis systems.Microfluidics, which confines fluid flow at microscale, attracts more and more attentions in the biological society.1–4 By scaling the flow domain down to microliter level, microfluidics shows attractive merits of low sample consumption, precise biological objective manipulation, and fast momentum/energy transportation. For example, various cell operations, such as culturing5–7 and sorting,8–10 have already been demonstrated with microfluidic approaches. In most biological applications, sterilization is a key sample pre-treatment step to avoid contamination. However, as far as the author knew, this important pre-treatment operation is generally achieved in an off-chip way, by using high temperature and high pressure autoclave. Actually, microfluidics has already been utilized to develop new solution for high pressure/temperature reactions. The required high pressure/temperature condition was generated either by combining off-chip back pressure regulator and hot-oil bath,11,12 or by integrating pressure regulator, heater, and temperature sensor into a single chip.13 This work presented a microfluidic sterilization strategy by implementing the previously developed continuous flowing high pressure/temperature microfluidic reactor.Figure Figure11 shows the working principle of the present microfluidic sterilization chip. The chip consists of three zones: sample loading (a microchannel with length of 270 mm and width of 40 μm), sterilization (length of 216 mm and width of 100 μm), and pressure regulating (length of 42 mm and width of 5 μm). Three functional zones were separated by two thermal isolation trenches. The sample was injected into the chip by a syringe pump and experienced two-step filtrations (feature sizes of 20 μm and 5 μm, not shown in Figure Figure1)1) at the entrance to avoid the channel clog. All channels had the same depth of 40 μm. According to the Hagen–Poiseuille relationship,15 the pressure regulating channel had a large flow resistance (around 1.09 × 1017 Pa·s/m3, see supplementary S1 for details16) because of its small width, thereby generated a high working pressure in the upstream sterilization channel under a given flow rate. The boiling point of the solution will then be raised up by the elevated pressure in the sterilization zone followed by the Antoine equation.16 By integrating heater/temperature sensors in the pressurized zone, a high temperature environment with temperature higher than 100 °C can thereby be realized for aqueous sample sterilization. The sample was collected from the outlet and cultured at 37 °C for 12 h. The results indicated that when the sterilization zone was heated up to 140 °C, the pressure regulating zone was about 40 °C. At this temperature, the viscosity of water decreases to 0.653 mPa·s from 1.00 mPa·s (at 20 °C), which will make the pressure in the sterilization zone reduced from 539 kPa (calculated at 20 °C and flow rate of 4 nl/s) to 387 kPa. The boiling point will then decrease to 142.8 °C, which will guarantee a boiling-free sterilization. In the cases without the thermal isolation trenches, the temperature of the pressure regulating zone reached as high as 75 °C because of the thermal interference from the sterilization zone, as shown in Figure Figure3.3. The pressure in the sterilization zone was then reduced to 268 kPa (calculated at flow rate of 4 nl/s) and the boiling temperature was around 130 °C, which was lower than the set sterilization temperature. E. coli with initial concentration of 106/ml was pumped into and flew through the chip with the sterilization temperatures varied from 25 °C to 120 °C at flow rates of 2 nl/s and 4 nl/s. The outflow was collected and inoculated onto the SS agar plate evenly with inoculation loops. The population of bacteria in the outflow was counted based on the bacterial colonies after incubation at 37 °C for 12 h. Typical bacterial colonies were shown in Figure Figure4.4. The low flow rate case showed a better sterilization performance because of the longer staying period in the sterilization channel. The population of E. coli was around 1.25 × 104/ml after a 432 s-long, 70 °C sterilization (at flow rate of 2 nl/s). While at the flow rate of 4 nl/s, the cultivation result indicated the population was around 3.8 × 104/ml because the sterilization time was shorten to 216 s. A control case, where the solution flew through an un-heated chip at 2 nl/s, was conducted to investigate the effect of the shear stress on the sterilization performance (see the supplementary S3 for details16). As listed in Table TableI,I, the results indicated that the shear stress did not show any noticeable effect on the bacterial sterilization. When the chip was not heated, i.e., the case with the largest shear stress because of the highest viscosity of fluid, the bacterial cultivation was nearly the same as the off-chip results (no stress). The temperature has the most significant effect on the sterilization performance. No noticeable bacteria proliferation was observed in the cases with the sterilization temperature higher than 100 °C, as shown in Figure Figure44.

In this research, we investigated whether a learning process has unique information searching characteristics. The results of this research show that information searching is a learning process with unique searching characteristics specific to particular learning levels. In a laboratory experiment, we studied the searching characteristics of 72 participants engaged in 426 searching tasks. We classified the searching tasks according to Anderson and Krathwohl’s taxonomy of the cognitive learning domain. Research results indicate that applying and analyzing, the middle two of the six categories, generally take the most searching effort in terms of queries per session, topics searched per session, and total time searching. Interestingly, the lowest two learning categories, remembering and understanding, exhibit searching characteristics similar to the highest order learning categories of evaluating and creating. Our results suggest the view of Web searchers having simple information needs may be incorrect. Instead, we discovered that users applied simple searching expressions to support their higher-level information needs. It appears that searchers rely primarily on their internal knowledge for evaluating and creating information needs, using search primarily for fact checking and verification. Overall, results indicate that a learning theory may better describe the information searching process than more commonly used paradigms of decision making or problem solving. The learning style of the searcher does have some moderating effect on exhibited searching characteristics. The implication of this research is that rather than solely addressing a searcher’s expressed information need, searching systems can also address the underlying learning need of the user.  相似文献
