Liposomes, especially cationic liposomes, are the most common and well-investigated nanocarriers for biomedical applications, such as drug and gene delivery. Like other types of nanomaterials, once liposomes are incubated in a biological milieu, their surface can be immediately cloaked by biological components to form a protein corona, which confers a new ‘biological identity’ and modulates downstream interactions with cells. However, it remains unclear how the protein corona affects the transportation mechanism after liposomes interact with cells. Here, we employed home-made aggregation-induced-emission-visualized nanoliposomes TR4@Lipo as a model to investigate transportation with or without the protein corona by optical imaging techniques. The results show that the protein corona can change the cellular transportation mechanism of TR4@Lipo from energy-independent membrane fusion to energy-dependent endocytosis. The protein corona also modulates the intracellular distribution of loaded cargoes. This knowledge furthers our understanding of bio-nano interactions and is important for the efficient use of cationic liposomes.  相似文献

The discovery of ProtoRAG in amphioxus indicated that vertebrate RAG recombinases originated from an ancient transposon. However, the sequences of ProtoRAG terminal inverted repeats (TIRs) were obviously dissimilar to the consensus sequence of mouse 12/23RSS and recombination mediated by ProtoRAG or RAG made them incompatible with each other. Thus, it is difficult to determine whether or how 12/23RSS persisted in the vertebrate RAG system that evolved from the TIRs of ancient RAG transposons. Here, we found that the activity of ProtoRAG is highly dependent on its asymmetric 5′TIR and 3′TIR, which are composed of conserved TR1 and TR5 elements and a partially conserved TRsp element of 27/31 bp to separate them. Similar to the requirements for the recombination signal sequences (RSSs) of RAG recombinase, the first CAC in TR1, the three dinucleotides in TR5 and the specific length of the partially conserved TRsp are important for the efficient recombination activity of ProtoRAG. In addition, the homologous sequences flanking the signal sequences facilitate ProtoRAG- but not RAG-mediated recombination. In addition to the diverged TIRs, two differentiated functional domains in BbRAG1L were defined to coordinate with the divergence between TIRs and RSSs. One of these is the CTT* domain, which facilitates the specific TIR recognition of the BbRAGL complex, and the other is NBD*, which is responsible for DNA binding and the protein stabilization of the BbRAGL complex. Thus, our findings reveal that the functional requirement for ProtoRAG TIRs is similar to that for RSS in RAG-mediated recombination, which not only supports the common origin of ProtoRAG TIRs and RSSs from the asymmetric TIRs of ancient RAG transposons, but also reveals the development of RAG and RAG-like machineries during chordate evolution.  相似文献

Document classification, with the blooming of the Internet information delivery, has become indispensable required and is expected to be disposed by an automatic text categorization. This paper presents a text categorization system to solve the multi-class categorization problem. The system consists of two modules: the processing module and the classifying module. In the first module, ICF and Uni are used as the indictors to extract the relevant terms. While the fuzzy set theory is incorporated into the OAA-SVM in the classifying module, we specifically propose an OAA-FSVM classifier to implement a multi-class classification system. The performances of OAA-SVM and OAA-FSVM are evaluated by macro-average performance index.  相似文献

The problem considered here is the optimal scheduling of a Boolean expression on a single-processor system. We consider a Boolean expression in which each Boolean variable represents the binary outcome (or return code) of a program module. The result of the expression may be known without executing all modules (e.g. an OR-operation is terminated as soon as one of the operands returns the value 1), so that the order in which the modules are executed influences the cost involved in finding the result. The optimisation discussed here consists in finding the operand arrangement that minimizes the average execution cost. The cost of each module represents the consumption of resources such as CPU time, elapsed time, number of input-outputs, etc. The assumptions regarding the modules are: (i) No module execution is the pre-requisite for the execution of another module. (ii) At each execution, the return code and the cost of a module do not depend on the sequence of execution. (iii) Over a number of executions, the return code and the cost of a module may be statistically dependent or a deterministic function of return codes and costs of other modules.A prototype scheduler is presented, that implements a sub-optimal strategy. It stores the Boolean expression in tabular form, passes control to a module at a time, as needed for the Boolean computation, measures the cost of execution and decides empirically what operand arrangement is more advantageous.  相似文献
