Dietary and lifestyle factors have been shown to have a profound effect on paraoxonase-1 (PON1) activity. Cigarette smoke has been shown to inhibit its mass and activity where as resveratrol has been shown to enhance it. We exposed hepatoma derived cell line (HepG2) to resveratrol and nicotine in varying doses and measured PON1 enzymatic activity and PON1 gene expression. In addition, total protein content of HepG2 cells was also measured. Resveratrol in a dose of 15 μmol/l or above significantly increased the PON1 enzyme activity (p > 0.001) where as nicotine in a dose of 1 μmol/l or higher significantly reduced it (p < 0.05). The resveratrol in this dose also enhanced the PON1 gene expression whereas nicotine decreased it as compared to controls. However, the protein conent of cells was not changed suggesting that they were not cytotoxic in the doses used. Till date the antioxidant vitamins have shown disappointing results against LDL oxidation and cardiovascular protection. However, the effect of resveratrol on PON1 gene expression and activity was significant, suggesting increase in PON1 activity and enhanced gene expression may be its alternative mechanism for offering protection against cardiovascular disease and may be an potential pharmacological agent which can be used for this. 相似文献

Over the last 35 years, centralized universal-number (in the United States, 9-1-1) systems have come to be the preferred means of emergency-response dispatching. The creation of these systems has motivated the development of information systems that render the landscape, to those with the right access, increasingly legible. This has been to the benefit of those receiving emergency services, but also to the benefit of police and commercial interests, who have used the improved infrastructure as bases for the creation of geodemographic and other profiling systems. More recently, the creation of wireless telephony has motivated the creation of a further surveillance infrastructure built on and integrated into that landscape. One consequence has been the commercialization of the systems in ways that permit the incorporation of more intimate and detailed data into preexisting systems. Public concern with locational surveillance systems has focused on privacy. Immunoreactivity properties of serum dilutions andPlasmodium falciparum malaria antigens were measured and compared by ELISA technique using different ELISA plates to evaluate the role of antigens
and serum dilutions for optimum binding. Also effort has been made to see the effect of reaction surface and material i.e.
ELISA plates for binding capacity. Serological properties were estimated by ELISA methods for detection of malaria and determination
of immunological characteristics. Three Pf antigens (PfAg) i.e. ring infected erythrocyte surface antigen: AR-1 (RESA), histidine-rich
protein 2 antigen (HRP-2) and glycophospholipid antigen (grown and developed Pf antigen from PSJ-M strain): GPL1 have been
used for serological testing of human blood samples by Enzyme Link Immunosorbant Assay (ELISA). 1∶100, 1∶1000 and 1∶10000
dilutions of Pf positive and negative serum (50 samples in each group) and 1∶1000 dilution of Pf antigens were used to measure
immunoreactive properties by ELISA method. Result of PfAg-serum immunoreactivity study showed that GPL1 has the highest degree
of immuno binding reactivity compared to other Pf antigens. HRP-2 and RESA antigens showed no significant difference to each
other. Study also found that Costar and Fastec ELISA plates have a better Ag−Ab binding capability compared to immulon and
Falcon plates at all dilutions of serum. Serum dilution of 1∶100 showed best binding and reactivity with Pf antigens followed
by 1∶1000 and 1∶10000 showed lowest reactivity. 相似文献

目的:设计获得5段hIL-18异构体,定向插入pEGFP-C1质粒形成融合基因,转染胰腺癌Bx-PC-3细胞并表达,为进一步研究改建的hIL-18蛋白功能奠定实验基础.方法:设计引物从pGEM-T-hIL-18质粒中PCR扩增得到5段不同的hIL-18片段,分别连接入pEGFP-C1载体构建不同的突变子(Mu0、Mu1、Mu2、Mu3和Mu4),转染原位胰腺癌BxPC-3细胞,以荧光显微镜观察绿色荧光蛋白的表达,RT-PCR法检测转染细胞中hIL-18表达水平.结果:(1)五种重组质粒经酶切与测序证实构建成功;(2)BxPC-3细胞转染重组质粒后可观察到绿色荧光蛋白的表达.结论:(1)成功构建了hIL-18五种异构体的绿色荧光蛋白表达载体;(2)改建的hIL-18蛋白可与绿色荧光蛋白在BxPC-3细胞融合表达. 相似文献
