目的:建立一种能延长薄荷种质资源离体保存时间的方法。方法:以薄荷试管苗为材料,接种到添加不同配比的甘露醇和蔗糖的MS(Murashige and skoog,1962)培养基中;在温度为(25±1)℃、光照强度2000 Lx,光照时间12 h/d、相对湿度60%左右的环境下培养;12个月后选成活率较高的试管苗转接到生根培养基进行恢复生长后驯化盆栽,以正常继代苗作对照,分别测量其株高、茎粗、叶片数及叶片SPAD值。结果:试管苗在添加蔗糖浓度30~90 g/L和甘露醇浓度0~15 g/L的MS培养基短期内都有部分存活,最长能保存12个月,存活率最高达60%。保存12个月的试管苗在MS培养基上均能恢复正常生长,驯化盆栽后的成活率为100%。结论:添加浓度为70 g/L蔗糖和5~10 g/L甘露醇的MS培养基适合薄荷试管苗的离体保存。 相似文献

将波士顿蕨匍匐茎接种于含铁、钙、磷等元素不同的培养基中培养,探讨铁、磷及钙对波士顿蕨试管苗生长的影响。试验结果表明,一定浓度的铁、钙、磷元素对波士顿蕨的生长有明显的促进作用,但浓度过高会抑制其生长。培养基中不同含量的铁处理中,铁盐含量为13.9~27.8 mg/L效果最佳;不同含量的钙处理中,钙盐含量为220 mg/L效果最好;不同浓度的磷处理中,磷盐含量为170 mg/L效果最好。 相似文献

目的:研究6-苄氨基腺嘌呤(6-BA)和α-萘2酸(NAA)对兰州百合鳞茎的诱导、增殖、生根及试管苗移栽的影响,筛选出不同培养阶段的最佳培养基.方法:选取生长健壮的兰州百合鳞茎,采用MS为基本培养基,添加不同浓度6-BA和NAA,进行不定芽的诱导.以鳞茎诱导的幼苗为外植体,MS添加不同浓度6-BA和NAA进行试管苗增殖培养.将试管苗转入以1/2MS、MS为基本培养基,添加不同浓度6-BA和NAA组合的生根培养基中进行试管苗的生根培养,当幼根长2～3 cm时,炼苗1～2 d,再移入蛭石和珍珠岩配比为1:1的营养钵中管理.结果:丛生芽诱导时,6-BA的最适浓度为1.0 mg/L,过高则芽的生长受到抑制;增殖试验中,NAA与6-BA配合使用,可提高芽的增殖率,而且新芽产生时间早,增殖倍数高;1/2 MS和MS培养基中NAA可以提高百合幼苗生根率,6-BA则明显抑制生根量.结论:兰州百合鳞茎片不同部位诱导丛生芽的诱导率顺序为下部＞中部＞上部;经炼苗移栽,再生株成活率可达100％. 相似文献

以甜叶菊试管苗为外植体,研究了MS培养基中微量元素对甜叶菊试管苗增殖、生根、生长及移栽等影响;同时也探讨了培养基中添加马铃薯汁、椰子汁、香蕉泥和水解乳蛋白等对试管苗增殖、生根、生长及移栽的影响。结果表明,微量元素对试管苗生长、增殖、生根产生重要作用,其中硼、锌、铜、铁的影响较为明显,尤其是锌元素是培养基中不可或缺的元素,它不仅影响试管苗的生长,而且对试管苗的移栽也产生较大影响,增加MS培养基中的锌元素的浓度,可促进试管苗的增殖、改善试管苗的质量;培养基中添加马铃薯汁、椰子汁、香蕉泥对试管苗的增殖具有一定的促进作用,其中香蕉泥能明显改进试管苗的质量,试管苗茎杆粗壮、生长旺盛,但添加水解乳蛋白却抑制试管苗的增殖和生长;在试管苗生根阶段,添加有机质作用不明显。 相似文献

以常熟白芹的带节茎段和顶芽为外植体进行离体培养,对外植体的灭菌方法、培养基中不同种类生长调节物质的浓度对增殖和生根的影响以及试管苗的保存方法等进行了研究．结果表明：以0．1％的HgCl2为灭菌剂,对水芹顶芽处理8min可以获得无菌苗30％;丛生试管苗切割成单株后接种在MS＋6-BA0．5～1．5mg·L。的增殖培养基上培养28d,平均增殖系数可达10．5以上;试管苗在1／2MS培养基上培养14d后,生根率达100％,21d时平均每株生根16．5条;试管苗用1mg·L。的NAA浸泡切口处24h,再用清水培养10d后,生根率达100％,平均每株生根22．8条;炼苗后移植成活率100％．在温度为15℃、光照时间12h·d^-1、光照强度20μmol·m^-2·s^-1的培养条件下,试管苗在含6-BA0．5mg·L^-1的增殖培养基上可保存6个月;MS培养基中Ca^2＋离子浓度加倍,可延长试管苗保存时间1-1．5个月． 相似文献

选取7种不同基质酵母浸膏配制种子培养基,设定8、10、12 g/L三个质量浓度,摇床培养cAMP合成菌,对菌落密度、种子液p H、菌体浓度(OD值)和葡萄糖比消耗率等参数进行综合分析,研究不同基质酵母浸膏种子培养基对cAMP合成菌生长的影响.结果表明:cAMP合成菌的种子培养基中酵母膏最佳质量浓度为8 g/L,以使用啤酒废酵母浸膏和蔗糖糖蜜酵母浸膏更为适宜. 相似文献

以文心兰、蝴蝶兰类原球茎薄切片为外植体,探讨了植物生长调节剂6-BA、Ad和NAA及其组合、蔗糖浓度及添加椰汁对类原球茎再生的影响.结果表明,特定浓度的3种植物生长调节剂的组合使用是PLBs高频发生的关键;培养基中添加椰汁能有效提高PLBs的诱导频率,培养基中高浓度蔗糖降低文心兰PLBs发生频率,但对蝴蝶兰没有影响;各处理诱导的PLBs在生长状况上差异不明显.适合两种兰花类原球茎薄切片高频诱导PLBs的固体培养基是1/2MS+4.0mg/L 6-BA+2.0mg/L Ad+1.0mg/L NAA+100～200ml/L椰汁+10～20g/L蔗糖+4.0g/L琼脂粉. 相似文献

以豫薯4号、豫薯5号、豫薯7号、豫薯12号、豫薯14号为材料,以MS为基本培养基对甘薯茎尖分生组织进行不同激素种类和浓度试验,结果表明,最佳诱导分化培养基为:MS+6-BAl.0mg/L+NAA0.1mg/L+GA30.05mg/L;继代培养与生根培养用·MS+NAA0.2mg/L为好.在28℃、光照14h/d条件下培养效果好.试管苗株系经指示植物、NCM-ELISA法检测,获得了5个品种的脱毒苗. 相似文献

Different nations have different social cultures, and the social culture of every nation is developing with the development of the nation, language is greatly influenced by social culture, which enables language to possess national and historical characteristics. Word is the smallest, independent, meaningful linguistic unit of language. It can well reflect the national and historical characteristics. From synchronic aspect, the social and cultural influence on word meaning is mainly embodied in conceptual and associative meanings of word. The incorrespondence of word meaning is a phenomenon that exists in both conceptual and associative meanings of word among different languages. From diachronic aspect, the development of social culture causes some changes of word meaning to some extent. He also shows how drama is valuable when discussed as a separate, autonomous subject in the school curriculum as opposed to one subsumed within English.
