线粒体Ca2+代谢对肱三头肌超微结构损伤的影响

摘    要:目的:通过建立大鼠离心运动后肱三头肌超微结构损伤的模型,从肱三头肌线粒体超微结构变化和线粒体钙代谢方面分析和探讨线粒体在运动引起的骨骼肌超微结构损伤中的变化。方法:将70只雄性SD大鼠,随机分成7组。运动组大鼠进行持续性下坡跑运动,分别在运动后即刻、12 h、24 h、48 h、72h、5d和安静状态取材。制作肱三头肌电镜切片并测定肱三头肌线粒体Ca2+含量。结果:1)运动后24 h至运动后48 h肱三头肌肌纤维和线粒体结构损伤严重,运动后72 h有所恢复,5 d后肌纤维结构基本恢复到正常状态,但仍偶见线粒体空泡变性和嵴缺损。2)运动后即刻,线粒体Ca2+含量迅速升高(P<0.01),运动后24 h达峰值,运动后72 h基本恢复。结论:1)持续性长时间的离心运动可以使肱三头肌超微结构产生阶段性的损伤变化。2)持续长时间离心运动后,使肱三头肌线粒体钙超载,进而加重肱三头肌纤维及线粒体结构的损伤。

Abstract:Objective:To explore the effect of mitochondria changes to the exercise induced skeletal muscle injury through observing the changes of skeletal muscle micro-structure and mitochondria calcium metabolic.Methods: Untrained male SD 70 rats were randomly divided into seven groups.The exercise groups ran on a motor-driven treadmill.Triceps brachii muscle electron microscope structure,mitochondria calcium concentration are measured.Results:1) Observing the skeletal fiber and mitochondria structure from the transmission electron microscope,the micro structure damaged seriously from 24h to 48h after running.The micro structure of 72h after running recovered,and 5 days after running skeletal muscle structure and mitochondria calcium metabolic recovered to resting level,while some mitochondria cristae coloboma remained.2) After continued long time eccentric exercise skeletal muscle mitochondria calcium concentration significantly increases after eccentric exercise(P<0.01),and peak appeared at 24h after exercise,finally 72h after exercise the value recover to the normal.Conclusion: 1) Continued long time eccentric exercise lead to triceps brachii muscle micro-structure injury.2) After continued long time eccentric exercise,the significantly increase of mitochondria calcium concentration aggravates the skeletal muscle micro-injury and mitochondria structure changes.
