海带多酚提取工艺优化及抗氧化性研究

摘    要:以海带多酚为研究对象,考察提取温度、料液比和超声时间对海带游离多酚和结合多酚提取率的影响、采用正交试验对多酚提取工艺进行优化,并通过测定多酚对DPPH自由基的清除能力评估其抗氧化性.结果表明,海带多酚的最佳提取工艺为:料液比为1:30(g/mL)、提取温度为40℃、超声时间为30 min.在此工艺条件下,海带总多酚的提取率为2.74%.当浓度为2.0 mg/mL时,游离多酚和结合多酚对DPPH·的清除率分别为41.72%和19.37%,表明海带多酚具有一定的抗氧化活性,且游离多酚的抗氧化活性高于结合多酚。

Abstract:This study was to explore the extraction conditions and antioxidant activity of polyphenols from laminaria japonica.The effects of extraction temperature,solid-liquid ratio and ultrasonic time on the extraction rates of free and bound polyphenols were investigated.The orthogonal experimental design was used to optimize the extraction process of polyphenols.The antioxidant capacity of polyphenols was characterized by DPPH free radical scavenging capacity.The results showed that the optimum extraction conditions of polyphenols were as follows:the extraction temperature was 40℃,the ultrasonic time was 30 min and the solid-liquid ratio was 1:30(g/mL).Under this condition,the extraction rate of total polyphenols was 2.74%.When the concentration of free and bound polyphenols were 2.0 mg/mL,the scavenging rates of DPPH free radical were 41.72%and 19.37%,respectively,kelp polyphenols had some antioxidant activity,and the anti-oxidation of free polyphenols was better than that of bound polyphenols.
