通过单因子试验和多因子正交试验,优化筛选了适于绣球菌摇瓶培养条件。结果表明,适于产菌丝体的摇瓶条件为:培养温度28℃,培养基起始pH4.5,接种量8%,摇瓶转速100r/min;摇瓶培养适于产菌丝体的最适碳氮源及其浓度分别为:可溶性淀粉2.5%,蛋白胨0.15%。适于产胞外多糖的摇瓶条件为:培养温度29℃,培养基起始pH5.5,接种量8%,摇瓶转速100r/min;摇瓶培养最适碳氮源及其浓度分别为:可溶性淀粉2.5%,蛋白胨0.25%。 相似文献

利用杏鲍菇(pleurotus eyngii)降解啤酒糟液态发酵生产木聚糖酶,运用单因素与正交试验研究了啤酒糟、玉米芯与麸皮的不同组分、氮源、pH值、培养时间、转速、装瓶量等对产木聚糖酶活力的影响,并对其粗酶的酶学性质进行了研究。结果表明:碳源为3%啤酒糟、1%麸皮和1%玉米芯,氮源为0.5%酵母膏,pH值为5,培养时间为8d,转速为160 r/m in,接种量为10%,装瓶量为100 mL/250mL时,酶活力最高。其粗酶液的最适反应温度为50℃,最适反应pH为6,在pH4~6的范围内酶活性较稳定。但粗酶液的热稳定较差,当温度升至60℃时酶活力损失65%以上。 相似文献

通过摇瓶培养,研究香菇液体深层培养最适发酵培养时间、最适碳源、最适氮源及最佳碳源与氮源质量配比.结果表明最适发酵培养时间为168 h,所得菌丝体干重达0.399 g/50 mL菌液,最适碳源为蔗糖,菌丝体干重达到0.396 g/50 mL菌液;最适氮源为牛肉膏,菌丝干重达到0.421 g/50 mL菌液,并通过正交实验筛选出最佳碳源与氮源质量配比为:W(蔗糖):W(牛肉膏):W(麦芽糖)=2.5:1:1. 相似文献

以紫外分光光度法作为庆大霉素含量测定手段,采用正交试验方法,对紫色小单孢菌产庆大霉素的发酵培养基及发酵条件进行优化,通过统计学分析,确定发酵培养基最优组合为:黄豆饼粉3.5%,CaCO30.5%,淀粉5.5%,CoCl20.0006%。优化的发酵条件为:种龄60 h,接种量12 mL,装液量50 mL/500 mL摇瓶。在此优化条件下添加一定量的氨基酸培养6 d,庆大霉素效价较优化前提高了85.5%。用紫外分光光度法与微生物法测定庆大霉素发酵液效价产生的误差小于4%,该方法可以用于教学实验中庆大霉素液体发酵培养基优化的快速筛选。 相似文献

The problem on the geometrc inequalities involving an n-dimensional simplex and its inscribed simplex is studied. Aninequality is established, which reveals that the difference between the squared circumradius of the n-dimensional simplex andthe squared distance between its circumcenter and barycenter times the squared circumradius of its inscribed simplex is not lessthan the 2(n-1)th power of n times its squared inradius, and is equal to when the simplex is regular and its inscribed siplex is atangent point one. Deduction from this inequality reaches a generalization of n-dimensional Euler inequality indicating that thecircumradius of the simplex is not less than the n-fold inradius. Another inequality is derived to present the relationship betweenthe circumradius of the n-dimensional simplex and the circumradius and inradius of its pedal simplex. 相似文献

Using virtual reality for interactive design gives a designer an intuitive vision of a design and allows the designer to achieve a viable, optimal solution in a timely manner. The article discusses the process of making the Virtual Reality System of the Humble Administrator＇s Garden. Translating building data to the Virtual Reality Modeling Language （VRML） is by far unsatisfactory. This creates a challenge for computer designers to do optimization to meet requirements. Five different approaches to optimize models have been presented in this paper. The other methods are to optimize VRML and to reduce the file size.
