Global DNA hypomethylation has been associated with increased risk for cancers of the colorectum, bladder, breast, head and
neck, and testicular germ cells. The aim of this study was to examine whether global hypomethylation in blood leukocyte DNA
is associated with the risk of hepatocellular carcinoma (HCC). A total of 315 HCC cases and 356 age-, sex- and HBsAg status-matched
controls were included. Global methylation in blood leukocyte DNA was estimated by analyzing long interspersed element-1 (LINE-1)
repeats using bisulfite-polymerase chain reaction (PCR) and pyrosequencing. We observed that the median methylation level
in HCC cases (percentage of 5-methylcytosine (5mC)=77.7%) was significantly lower than that in controls (79.5% 5mC) (P=0.004, Wilcoxon rank-sum test). The odds ratios (ORs) of HCC for individuals in the third, second, and first (lowest) quartiles
of LINE-1 methylation were 1.1 (95% confidence interval (CI) 0.7–1.8), 1.4 (95% CI 0.8–2.2), and 2.6 (95% CI 1.7–4.1) (P for trend <0.001), respectively, compared to individuals in the fourth (highest) quartile. A 1.9-fold (95% CI 1.4–2.6) increased
risk of HCC was observed among individuals with LINE-1 methylation below the median compared to individuals with higher (>median)
LINE-1 methylation. Our results demonstrate for the first time that individuals with global hypomethylation measured in LINE-1
repeats in blood leukocyte DNA have an increased risk for HCC. Our data provide the evidence that global hypomethylation detected

Alkaline hydrogen evolution reaction (HER), consisting of Volmer and Heyrovsky/Tafel steps, requires extra energy for water dissociation, leading to more sluggish kinetics than acidic HER. Despite the advances in electrocatalysts, how to combine active sites to synergistically promote both steps and understand the underlying mechanism remain largely unexplored. Here, Density Functional Theory (DFT) calculations predict that NiO accelerates the Volmer step while metallic Ni facilitates the Heyrovsky/Tafel step. A facile strategy is thus developed to control Ni/NiO heterosurfaces in uniform and well-dispersed Ni-based nanocrystals, targeting both reaction steps synergistically. By systematically modulating the surface composition, we find that steering the elementary steps through tuning the Ni/NiO ratio can significantly enhance alkaline HER activity, and Ni/NiO nanocrystals with a Ni/NiO ratio of 23.7% deliver the best activity, outperforming other state-of-the-art analogues. The results suggest that integrating bicomponent active sites for elementary steps is effective for promoting alkaline HER, but they have to be balanced. 相似文献

Central precocious puberty (CPP) refers to a human syndrome of early puberty initiation with characteristic increase in hypothalamic production and release of gonadotropin-releasing hormone (GnRH). Previously, loss-of-function mutations in human MKRN3, encoding a putative E3 ubiquitin ligase, were found to contribute to about 30% of cases of familial CPP. MKRN3 was thereby suggested to serve as a ‘brake’ of mammalian puberty onset, but the underlying mechanisms remain as yet unknown. Here, we report that genetic ablation of Mkrn3 did accelerate mouse puberty onset with increased production of hypothalamic GnRH1. MKRN3 interacts with and ubiquitinates MBD3, which epigenetically silences GNRH1 through disrupting the MBD3 binding to the GNRH1 promoter and recruitment of DNA demethylase TET2. Our findings have thus delineated a molecular mechanism through which the MKRN3–MBD3 axis controls the epigenetic switch in the onset of mammalian puberty. 相似文献

Satellites have shown free-space quantum-communication ability; however, they are orbit-limited from full-time all-location coverage. Meanwhile, practical quantum networks require satellite constellations, which are complicated and expensive, whereas the airborne mobile quantum communication may be a practical alternative to offering full-time all-location multi-weather coverage in a cost-effective way. Here, we demonstrate the first mobile entanglement distribution based on drones, realizing multi-weather operation including daytime and rainy nights, with a Clauser-Horne-Shimony-Holt S-parameter measured to be 2.41 ± 0.14 and 2.49 ± 0.06, respectively. Such a system shows unparalleled mobility, flexibility and reconfigurability compared to the existing satellite and fiber-based quantum communication, and reveals its potential to establish a multinode quantum network, with a scalable design using symmetrical lens diameter and single-mode-fiber coupling. All key technologies have been developed to pack quantum nodes into lightweight mobile platforms for local-area coverage, and arouse further technical improvements to establish wide-area quantum networks with high-altitude mobile communication. 相似文献
