溃疡性结肠炎(ulcerative colitis,UC)是一种原因不明主要发生在结肠粘膜层的炎症性病变,以溃疡糜烂为主。其病因及发病机制目前尚无定论,但倾向性意见是UC与自身免疫紊乱有关,所涉及到的自身抗体主要归纳为两类,一类是抗结肠抗体(ACA),另一类是抗人中性粒细胞胞质抗体(ANCA)。本文就溃疡性结肠炎免疫学发病机理及血清自身抗体检测的临床应用加以阐述,以便为UC病因学和诊断学的研究提供客观依据。  相似文献

将人细胞周期蛋白D1基因克隆入原核表达载体pET-20b中获得重组质粒pET-20b-eyeD,经酶切鉴定正确后转化大肠杆菌BL21PlaysS后获得表达菌株.该菌株经IPTG诱导后表达的目的蛋白有部分分泌到培养基上清中,将培养基上清中的蛋白沉淀后用Ni^2+螯合柱进行纯化,最后可得到纯度达到95%以上的目的蛋白.蛋白电泳显示纯化蛋白的分子量约为33KD,Westemblot分析表明,在电泳胶的相应分子量处出现特异性条带,说明已经成功表达和纯化了重组人细胞周期蛋白D1.  相似文献

从唐山师范学院花房中采集菘蓝根腐病病根并对其进行了病原菌的分离、纯化和鉴定及不同浓度、不同分子量的壳聚糖对菘蓝根腐病病原菌的室内抑菌作用。结果表明:分离纯化得到致病菌经鉴定为半知菌亚门(Deuteromycotina)、丝孢纲(Hyphomycetes),丛梗孢目(Stilbellales)、瘤座孢科(Tuborculadales)、镰刀属(Fusarium)真菌。在一定浓度范围内,分子量为3KD、50KD的壳聚糖随着浓度的提高,其抑菌效果也逐渐增强,6mg/ml的效果最好;分子量为150KD时,3mg/ml的效果最好;在一定分子量范围内,除0.75mg/ml的浓度外,随着壳聚糖分子量的提高,其抑菌效果也逐渐增强,分子量150KD的效果最好;本试验中,最大抑菌率为61%。  相似文献

以小麦黄化苗为材料,采用不同离子强度的缓冲液抽提,通过硫酸铵分级沉淀、离子交换层析和分子筛层等步骤分离出两种不同分布的PGIP.经SDS-PAGE分析:不溶性PGIP出现两条谱带,分子量为41KD和39KD,而可溶性PGIP出现一条谱带,分子量为41KD.  相似文献

[目的]了解胃癌患者血清中抗核抗体谱的特点.[方法]以鸡卵核为抗原,应用免疫印迹法对18份胃癌术前患者以及22份正常对照组(年龄和性别分布均无差异)的血清进行分析.[结果]胃癌术前患者血清中特异识别5条抗原条带的抗核抗体的阳性率显著高于正常对照组,靶抗原的蛋白分子质量及胃癌术前组血清中特异识别这5条抗原条带的抗核抗体的阳性率分别为:69KD(44.4%,8/18),62KD(50%,9/18),58KD(33.9%,7/18),44KD(22.2%,4/18),37KD(38.9%,7/18).[结论] 胃癌患者体内存在抗核抗体,其相对的靶抗原有多种.  相似文献

目的:探讨小鼠大脑皮层源性神经干细胞(NSCs)体外培养的两种方法并对培养的细胞进行鉴定.方法:分别取孕14~16d的昆明小鼠胚胎脑皮质并用细胞球悬浮培养和单层贴壁培养两种方法进行体外培养,用光学显微镜观察NSCs的生长情况,并用免疫细胞化学鉴定NSCs特异性抗原的表达,经过血清对NSCs分化诱导后进行胶质纤维酸性蛋白及微管相关蛋白-2的检测.结果:①培养出来的细胞可以扩增生长;②这两种方法分别培养的神经干细胞球和单层神经干细胞经抗巢蛋白免疫细胞染色均呈阳性;③两种培养方法培养出的NSCs经诱导分化后,免疫细胞化学染色显示微管相关蛋白-2、神经胶质酸性蛋白染色阳性.结论:采用无血清培养基中加入特定生长因子的神经干细胞球培养和单层贴壁两种培养技术,可培养出在体外稳定增殖并有多向分化潜能的NSCs.  相似文献

分子量是蛋白质的重要特征参数,准确测定蛋白质分子量对研究蛋白质结构与功能具有重要意义。该文以牛血清蛋白(BSA)和人血清免疫球蛋白(IgG)为研究对象,采用分析超速离心-沉降速率法(AUC-SV)测定2种蛋白的分子量,并系统研究了样品初始浓度、离心转速、离心时间、离心温度、样品上样体积和缓冲液体系等实验条件对测定结果的影响。结果表明:离心转速为20 000~40 000 r/min时,影响不明显,样品初始浓度、离心时间、离心温度、缓冲液体系的适宜范围较宽,样品上样体积对测定结果的影响显著。在优化的实验条件下,测得未知蛋白的分子量为39.9 kDa,与质谱法测定结果(40.0 kDa)一致。该方法具有适宜条件范围宽、非破坏性、无需标准品和普适性高等优点,适用于未知蛋白分子量的测定。此外,相关研究结论还可供AUC-SV初学者学习借鉴。  相似文献

分别采用截留分子量为150KD和200KD的超滤膜过滤谷氨酸发酵液,对过滤效果以及膜清洗后的通量恢复率进行了比较.试验结果表明,150KD和200KD的超滤膜对谷氨酸发酵液中菌体的去除率分别为98.96%和97.93%,对COD的去除率分别为48.72%和42.31%,对NH3-N的去除率分别为21.42%和18.57%,所得滤液的OD分别为0.005和0.010.在超滤除菌试验中,两种膜的通量衰减速率在浓缩周期的不同时段是不同的;在再生后的水流测试中,150KD膜的通量恢复率为93%~99%,200KD膜的通量恢复率为90%~98%.因此,截留分子量为150KD的超滤膜更适合应用于谷氨酸发酵液除菌工艺.  相似文献

Background

Birch pollen sensitization and associated pollen-food syndrome among Chinese allergic patients have not been investigated.

Methods

Sera from 203 allergic patients from the northern part of China and collected during February to July 2014 were investigated. Specific immunoglobulin E (IgE) against birch pollen extract Bet v and major birch pollen allergen Bet v 1 were measured using the ADVIA Centaur. The presence of major apple allergen Mal d 1 and soy bean allergen Gly m 4 specific IgE was measured by ImmunoCAP 100.

Results

Among the 203 sera, 34 sera (16.7%) had specific IgE to Bet v and of these, 28 sera (82.4%) contained Bet v 1-specific IgE. Among the 28 sera with Bet v 1-specific IgE, 27 sera (96.4%) contained Mal d 1-specific IgE and 22 sera (78.6%) contained Gly m 4-specific IgE. Of the 34 Bet v-positive sera, 6 sera (17.6%) contained no specific IgE for Bet v 1, Mal d 1, or Gly m 4. Almost all Bet v-positive sera were donated during the birch pollen season.

Conclusions

The prevalence of birch allergy among patients visiting health care during pollen season can be as high as 16.7% in Tangshan City. The majority of Chinese birch allergic patients are IgE-sensitized to the major birch pollen allergen Bet v 1 as well as to the major apple allergen Mal d 1 and soy bean allergen Gly m 4. A relatively high number of patients (17.6%) are IgE-sensitized to birch pollen allergen(s) other than Bet v 1. The high prevalence of specific IgE to Mal d 1 and Gly m 4 among Bet v 1-sensitized patients indicates that pollen-food allergy syndrome could be of clinical relevance in China.

[目的]新的大肠癌相关性抗原EID3的基因克隆及其诊断价值研究.[方法]利用大肠癌病人体内血清中所含的对肿瘤抗原产生的特异性抗体筛选睾丸组织cDNA噬菌体表达文库和大肠癌组织cDNA噬菌体表达文库(SEREX),并用RT-PCR技术研究EID3 mRNA在正常组织和大肠癌传代细胞表达.[结果]睾丸组织cDNA噬菌体表达文库筛选得到了可以诱导大肠癌病人抗体免疫应答的新抗原EID3基因(Gen-bank NM_001008394.1).它们定位于染色体19q13.2,EID3含1个外显子.通过RT-PCR分析发现,EID3基因在43例大肠癌传代细胞株中,39例阳性,阳性率为90.7%.在正常组织中,除睾丸组织外不表达或有极低水平转录.[结论]EID3 mRNA表达检测用于诊断大肠癌,可能具有高特异性和高敏感性的特点.EID3蛋白被首次发现在大肠癌病人中能够诱导机体的抗体免疫应答,为一个新的大肠癌相关性抗原分子.其功能可能与抑制细胞的恶性增殖相关,并可进一步研究其用于治疗和诊断大肠癌的可行性.  相似文献

Objective: The aims of this research were to purify and identify the 130 kDa (CagA) protein of H. pylori clinical isolate HP97002 and evaluate the relationships between the purified 130 kDa (CagA) protein and gastric diseases. Methods: The procedure for isolating the protein included 6 mol/L guanidine extract, size exclusion chromatography and elusion from gel. Sera of 68 patients with gastric diseases (44 with chronic gastritis,15 with atrophic gastritis,7 with peptic ulcer disease,2 with gastric cancer ) were obtained, and the serological response to CagA was studied by Western-blot using the purified protein. Results: The purified protein was 130 kDa and preserved good antigenicity and revealed basic isoelectric point about of 8.1. Among 68 sera, 43 sera could recognize the purified protein associated with chronic gastritis 47.7% (21/44),atrophic gastritis 86.7% (13/15),peptic ulcer disease 100% (7/7),gastric cancer 100% (2/2). Compared with each other, the difference was significant (χ2=13.327, P=0.004), and 130 kDa (CagA) protein was associated with severe gastric diseases (rs=0.442, P=0.001). Conclusion: The 130 kDa (CagA) protein was associated with severe gastric diseases.  相似文献

Assaying parathyroid hormone (PTH) in the washing liquid after fine-needle aspiration biopsy (FNAB) seems to be a valid approach to locate parathyroid tissue. PTH-FNAB was evaluated in 47 patients with a clinical picture of primary hyper-parathyroidism (PHP) and ultrasonography (US) suggestive of parathyroid lesion. The patients were subdivided into two groups on the basis of the absence or presence of US thyroid alterations. The result of PTH-FNAB was compared with those of cytology, scintigraphy and, in 24 patients, surgical outcome. PTH-FNAB samples with a value higher than that recorded in the serum and higher than our institutional cut-off were deemed to be probable samples of parathyroid tissue. Cytology proved diagnostic for benign thyroid lesions, non-diagnostic for thyroid lesions, hyperplastic parathyroid tissue, undetermined or malignant thyroid lesions and other lesions in 45%, 30%, 17%, 4%, and 4% of cases, respectively. In 47% of cases, PTH-FNAB indicated that the sample had been taken in parathyroid tissue. In patients without US alterations, the diagnostic accuracy of PTH-FNAB was greater than that of scintigraphy. After surgery, comparison between the results of PTH-FNAB and scintigraphy, in terms of positive predictive value (PPV), revealed the superiority of PTH-FNAB; PPV was 94% for FNAB and 71% for scintigraphy, while sen-sitivity was 83% and 69%, respectively. PTH-FNAB evaluation after FNAB appears to be more diagnostic than cytology and scintigraphy. Of all the procedures used, PTH-FNAB appears to be the method of choice when the target is US suggestive and reachable. PTH-FNAB appears to be a useful method of guiding surgical intervention.  相似文献

Simple procedures for extraction and chromatographic determination ofdimethachlon residues in fresh tobacco leaves and cut-tobacco are described. The determination was carried out by capillary gas chromatography (GC) with electron capture detection (ECD) and confirmed by GC-MS. The mean recoveries and relative standard deviation (RSD) were 93.2%-112.9% and 3.5%-6.7%, respectively at levels ranging from 0.01 to 0.1 mg/kg. The limit of determination was 0.001 mg/kg. Tobacco samples in routine check were successfully analyzed using the proposed method.  相似文献

The histopathological features and the associated clinical findings of ulcerative colitis (UC) are due to persistent inflammatory response in the colon mucosa. Interventions that suppress this response benefit UC patients. We tested whether sodium arsenite (SA) benefits rats with dextran sulfate sodium (DSS)-colitis. The DSS-colitis was induced by 5% DSS in drinking water. SA (10 mg/kg; intraperitoneally) was given 8 h before DSS treatment and then every 48 h for 3 cycles of 7,14 or 21 d. At the end of each cycle rats were sacrificed and colon sections processed for histological examination. DSS induced diarrhea, loose stools, hemoccult positive stools, gross bleeding, loss of body weight, loss of epithelium, crypt damage, depletion of goblet cells and infiltration of inflammatory cells. The severity of these changes increased ir the order of Cycles 1,2 and 3. Treatment of rats with SA significantly reduced this severity and improved the weight gain.  相似文献
